[Method] With 16 hybrids and their parents and 202 inbred elites as materials for screening primers, the artificial groups of two standard diad and two standard triad were respectively established as the verification materials. Genomic DNA of seedlings was extracted by using CTAB method. 137 pairs of SSR primers were selected for SSR amplification and product detection, which was used for the parentage identification of maize varieties. [ Result] Twenty pairs of corn primers with high polymorphism information content ( PIC value), clearly amplified bands and good reproducibility were screened from 137 pairs of corn SSR primers tested. The identification results of using SSR molecular were consistent with the actual situation. [ Conclusion] It is feasible to identify the parentage of maize variety using SSR markers.展开更多

[Method] The phenotypic characteristics and germination rate of the seeds of four excellent lupin pedigrees were measured, and then their correlations with seeding growth were analyzed. [Result] There were abundant variations among the four ornamental lupin pedigrees. Pink seeds had the largest volume and red seeds had the largest 1 000seed weight. The variation coefficients for the seed morphological traits among the four pedigrees ranged from 2.97% to 14.34%. Seed specific weight and 1 000-seed weight could be used as important indicators for selection in breeding because of their higher variability. Seed weight variation of ornamental lupin was mainly depen- dent on seed width variation. There was small variation in seed length. The seeds of the four ornamental lupin pedigrees started to germinate one day after sowing, and the germination period was 5 d. Germination rate coefficient and germination index of pink and red seeds were higher than those of blue seeds, but blue seeds had the largest germination rate. 1 000-seed weight shared significantly positive correlations with seed germination rate, germination potential and seedling retention rate. Round and large seeds had some advantages in germination. Full seeds had higher germination rate and speed, and seedling retention rate. There was a significant relationship between seed length-width ratio and the number of leaflets of seedlings. [Conclusion] The results provided references for the evaluation of seed phenotypic diversity and breeding research of ornamental lupins.展开更多

It was composed of adjustable screw, stirrer, metering device housing and central metering sheave and so on. The sowing rate can be set by turning the screw to change the working length of the central metering sheave relative to the metering device housing. The stirrer inside of the sheave housing is used to prevent seeds overhead. And metering of different sizes of seed is adjusted by changing the position of internal components of the slot wheel mechanism. Innovative design on the structure of the central metering sheave was finished. According to the structure parameters and physical characteristic parameters, different seed sowing rate of per hectares was calculated. And then the working length scale of the central metering groove wheel was made. And there is a one-to-one correspondence between scale values and sowing quantity per hectare of different kinds of seed.展开更多
Introgression with Saceharum spontaneum intended to broaden the genetic base of sugarcane resulted in an increase in desirable genes for broader adapt ability as well as undesirable genes for high starch content. Mark...Introgression with Saceharum spontaneum intended to broaden the genetic base of sugarcane resulted in an increase in desirable genes for broader adapt ability as well as undesirable genes for high starch content. Markers could provide a quick and efficient method of screening parental genotypes for low starch. The objectives of this study were to identify potential markers associated with starch in S. spontaneum population and evaluate their potential for screening for starch content. Data for starch content and (simple sequence repeats) SSR markers were collected from 51 S. spontaneum clones grown in replicated pots. The mixed procedure of statistical analysis system (SAS) was used to determine markers significantly associated with starch. Thirty-nine out of 357 polymorphic markers were significantly (P 〈 0.05) associated with starch content. Eighteen were positively associated and 21 were negatively associated. The presence of a positive marker produced 39% more starch than absence while the absence of a negative marker produced 57% more starch than presence. Selecting parents using negative markers may be more efficient than selecting using positive markers.展开更多
