[Method] In order to seek standardizing PCR reaction set-up, an orthogonal design was used to optimize ISSR-PCR amplification system of Bryaceae in five factors (Mg2+, dNTPs, primer, DNA template, Taq DNA polymerase) at four levels respectively. [Result] A suitable ISSR reaction system was obtained, namely: 20 μl reaction system containing 5 ng of DNA template, 0.2 μmol/L primer, 2.25 mmol/L MgCl2, 0.6 U of Taq DNA polymerase, 0.4 mmol/L dNTPs. Proper annealing temperature was found at 48-50 ℃.The above system and six ISSR-PCR primers were used for the PCR amplification of 14 samples from Bryaceae and the related species in Mniaceae. A total of 86 bands were amplified, all showed polymorphism. NJ cluster analysis showed a star-shaped cladogram. [Conclusion] The results manifested that ISSR fingerprinting could provide the appropriate degree of polymorphism at low taxonomic level, so it would be a useful tool to provide additional evidence for resolving taxonomic relationships at the species level of Bryaceae.展开更多

The results showed that seed rate had great effects on root twining power and qualityof rice seedlings; with the increase of seed rate, the root twining power of rice seedlings was increased, but the quality of rice seedlings was significantly reduced; under the condition of high seed rate, rice seedlings were weak with less accumulate dry matter and nonuniform growth, and the proportion of weak seedlings was higher. When the seed rate of both rice cultivars was reduced by 10 g/tray, the average seedling height was increased by 0.5 cm, leaf age was increased by 0.1, stem base diameter was increased by 0.1 cm, and root number per plant was increased by 0.3. When the seed rate was increased by 10 g/tray, the accumulated dry matter per plant was reduced by 5.1 mg. With the increase of seed rate, the seedling quality of Zhongzheyou No.1 was decreased rapidly, and the proportion of weak seedlings was increased rapidly. However, Yongyou 12 has a larger elasticity in seed rate. When the seed rate reached to 50 g/tray, blanket seedlings of Yongy- ou 12 were nursed with uniform growth and higher quality, but the shoots were sparse with lower leaf area index. However, when the seed rate was up to 50 g/tray, blanket seedlings of Zhongzheyou No.1 were nursed with uniform growth and higher quality, as well as higher-density shoots. The results showed that the project of "planting three to produce four" performed significantly in terms of increasing yields and incomes. For example, per unit area yield of one-season late hybrid rice and middleseason hybrid rice proved the highest, and net income from middle-season rice in Longhui was the highest. It is obvious that to increase per unit yield is key for improving net incomes and to reduce machine fees and worker fees is crucial for decreasing fees.展开更多

Rice allelopathy against weeds was reported since 1989, which offers an integrated weed management with substantially reduced herbicide usage. Application of allelopathic rice cultivars is thought a resources conservation and environmental friendly way of weed bio-control, and could promote the sustainable development of agriculture. Screening or evaluating the allelopathic potential rice variety is the first step. In this paper, a new bioassay method was set up by the allelopathic potential of 9 rice lines on the target weed barnyardgrass (Echinochloa crusgalli), and comparing with bioassay methods such as relay seeding in filter paper (RSF) and relay seeding in agar (RSA). The results indicated that three methods had a same tendency in evaluating the allelopathic potential of rice; there existed a significant difference among different bioassay methods, and an interaction between bioassay methods and rice lines. The method of root exudates (RE) with the highest value and a correlation efficiency of 0.98 was considered as ideal bioassay method for evaluation of allelopathic potential.展开更多
