Under a given condition of crystallization, dark brown short rhombohedron crystals could be obtained from Δ nifZ MoFe protein purified from a nifZ deleted mutant strain of Azotobacter vinelandii Lipmann....Under a given condition of crystallization, dark brown short rhombohedron crystals could be obtained from Δ nifZ MoFe protein purified from a nifZ deleted mutant strain of Azotobacter vinelandii Lipmann. Systematic studies on the effect of concentrations of PEG 8000,MgCl 2, NaCl,Tris and buffer pH on the crystallization and crystal growth of the protein showed that the protein could not be crystallized in lower concentrations of the chemicals and lower buffer pH. A large amount of smaller crystals of the protein appeared in a week with gradual increasing in the chemical concentrations and pH≥8.0. When the chemical concentrations were further increased, the time for crystallization was increased and a few high grade crystals of larger size were formed. If the concentrations of the chemicals were continuously increased, many crystals with smaller size, and, sometimes of poor quality appeared again and eventually ceased to produce any crystals. The optimal concentration for each of the above mentioned chemicals varies with other variable factors. Only one bigger crystal (both of the longest two sides: 0.16 mm) could be obtained in a hanging drop of protein sample when the concentrations of PEG 8000, MgCl 2, NaCl,Tris and protein were kept at 1.86%, 300 mmol/L, 400 mmol/L, 53 mmol/L and 4.64 g/L , respectively, with Tris buffer pH 8.2.展开更多
AIM: To describe the clinical and histological characteristics of a group of adults with small-bowel nodular lymphoid hyperplasia (NLH). METHODS: Patients were searched for five years in pathology records of our i...AIM: To describe the clinical and histological characteristics of a group of adults with small-bowel nodular lymphoid hyperplasia (NLH). METHODS: Patients were searched for five years in pathology records of our institution. The biopsy material was reassessed using strict histopathological criteria. Clinical data were obtained from medical records. RESULTS: Small-bowel NLH was diagnosed in 18 cases. The female: male ratio was 2 : 1. The most frequent symptoms were diarrhea (72%), involuntary weight loss (72%) and abdominal pain (61%). Nine patients (50%) had immunodeficiency. Small-bowel bacterial overgrowth was found in three (17%) cases. At small-bowel NLH diagnosis, three (17%) had associated lymphoma: two intestinal and one extra-intestinal lymphomas. In two patients with villous atrophy and anti-endomysial antibodies the diagnosis of celiac disease was established. Giardia larnblia infection was found in only one patient with hypogammaglobulinemia (Herman's syndrome). CONCLUSIONS: NLH is uncommon in adult patients. Associated diseases are immunodeficiency and lymphoid tissue malignancies.展开更多
The study was aimed at evaluating growth response of Heteroclarias fingerlings fed diet in which fish meal was substituted with graded levels of earthworm meal. The replacement levels were 0.00%, 25%, 50%, 75% and 100...The study was aimed at evaluating growth response of Heteroclarias fingerlings fed diet in which fish meal was substituted with graded levels of earthworm meal. The replacement levels were 0.00%, 25%, 50%, 75% and 100% designated as Eo, E25, Es0, E75 and El00, respectively. Fingerlings used for the work were obtained through artificial insemination of brood stock (purchased from local farmers) with ovaprim in the laboratory. The resulting fry were reared and fed with cultured plankton for 2 weeks followed by 100% fish meal for another two weeks. Thereafter sixteen fingerlings weighing 4.73 g and measuring 6.512 mm standard length (on the average) were starved over night and reared in five indoors tanks (0.8 ~ 0.5 ~ 0.5 m) in duplicate for eight weeks. Fingerlings were fed on graded earthworm meals of 40% protein twice daily at 08:00 and 16:00. The highest mean weight (6.77 g), specific growth rate (0.86) and protein efficiency ratio (0.6) were obtained at 50% replacement level. Also, Food conversion ratio was the highest at 50% substitution level and lowest at 0.00% substitution, indicating that fingerlings use earthworm containing meal more efficiently. Furthermore, cost benefit analysis revealed that the profit index for 50% inclusion of earthworm meal was the highest (1.71) and the highest net profit of N374.32 (circa $2.3) was achieved at this level. Water quality parameters observed were within tolerable values for the fingerlings well being. Consequently, farmers are advised to use 50% earthworm meal in place of fish meal for formulation of Heteroclarias fingerlings diet. This will drastically reduce production cost, increase farmers profit and enhance the biological value of our menu.展开更多
The heat shock cognate protein 70 (Hsc70) is a member of a 70-kDa heat shock protein (HSP70) family that functions as molecular chaperones.In this study,a novel Hsc70 gene from Chinese soft-shelled turtle (Pelodiscus ...The heat shock cognate protein 70 (Hsc70) is a member of a 70-kDa heat shock protein (HSP70) family that functions as molecular chaperones.In this study,a novel Hsc70 gene from Chinese soft-shelled turtle (Pelodiscus sinensis) (tHsc70) was identified.The tHsc70 full-length complementary DNA (cDNA) is 2 272 bp long with a 1 941-bp open reading frame (ORF) encoding 646 amino acids.Three characteristic signature regions of the HSP70 family,two major domains of an adenosine triphosphate (ATP)/guanosine triphosphate (GTP) binding domain (ABD),and a substrate-binding domain (SBD) were present in the predicted tHsc70 amino acid sequence.The tHsc70 gene was expressed in Escherichia coli BL21 and the expression product reacted with the anti-Hsc70 mouse monoclonal antibody by Western blotting.Homology analysis revealed that tHsc70 shared identity from 53.9% to 87.7% at the nucleotide level,and 49.1% to 99.5% at the amino acid level with the known Hsc70s.Phylogenetic analysis showed that tHsc70 was clustered together with the Hsc70 gene of another reptile species (Alligator mississippiensis).The tHsc70 was expressed in the liver,lung,heart,and skeletal muscle.The expression patterns of tHsc70 messenger RNA (mRNA) differed among different tissues under different durations of heat stress at 40 °C.Adaptation at 25 °C for 1 h after heat stress was also different among tissues and length of heat stress.Irrespective of different profiles of expression under heat stress,tHsc70 may play roles in protecting turtles from thermal stress.展开更多

Comparing the morphology and molecular biology of cave species and their close surface relatives is a powerful tool for studying regressive eye evolution and other adaptive phenotypes. We compared two co-occurring and closely-related species of the fish genus Sinocyclocheilus, which is endemic to China and includes both surface- and cave-dwelling species. Sinocyclocheilus tileihornes, a cave species, had smaller eyes than Sinocyclocheilus angustiporus, a surface species. Histological and immunohistochemical analyses revealed that the cave- fish had shorter cones and more disorderly rods than did the surface-dwelling species. Using quantitative PCR and in situ hybri- dization, we found that rhodopsin and a long-wavelength sensitive opsin had significantly lower expression levels in the cavefish. Furthermore, one of two short-wavelength-sensitive opsins was expressed at significantly higher levels in the cavefish. Changes in the expression ofopsin genes may have played a role in the degeneration of cavefish eyes [Current Zoology 59 (2): 170-174, 2013].展开更多
