Store-operated Ca2+-release-activated Ca2+ (CRAC) channels constitute the sole pathway for Ca2+ entry following antigen-receptor engagement. In a set of breakthrough studies over the past two years, stromal interaction molecule 1 (STIM1, the ER Ca2+ sensor) and Orai1 (a pore-forming subunit of the CRAC channel) have been identified. Here we review these recent studies and the insights they provide into the mechanism of store-operated Ca2+ channels (SOCCs).展开更多
Lonicerae flos,a widely used traditional Chinese medicine(TCM),has been used for several thousand years in China.As a famous traditional Chinese herbal medicine,it was used as heat-clearing drug and alexipharmic agent...Lonicerae flos,a widely used traditional Chinese medicine(TCM),has been used for several thousand years in China.As a famous traditional Chinese herbal medicine,it was used as heat-clearing drug and alexipharmic agent and was widely cultivated in Hunan province.L.flos mainly contains biologically active compounds such as caffeic acid derivatives,essential oil,flavonoids,iridoid glycosides and terpenoids.A range of biological activities has been reported from plant extracts including anti-inflammatory,antitumor,antioxidant,antiallergy,immunomodulating and antibacterial activity.In this study,the author investigated ancient books of TCM and nowadays reports,summarized the chemical constituents,biological activity of L.flos to provide a comprehensive systematic review.展开更多
Since the 1980s, people' s material and spiritual life are constantly enriched and improved. People' s values are constantly changing, more and more people realize that participate in physical exercise for their own...Since the 1980s, people' s material and spiritual life are constantly enriched and improved. People' s values are constantly changing, more and more people realize that participate in physical exercise for their own importance in sports. The status of people' s lives has been greatly improved, for most residents; they participate in physical activity and have gradually become a richer cultural life, and they enhance the basic needs of physical, mental lifelong sports have gradually deep heart. While people continue to enhance the sports consciousness exercise, we also see some potential problems, such as the rapid development of science and technology is to bring more and more problems to people. As people, enjoy the level of automation in the material aspects of life. They gradually have cars, telephones, washing machines, refrigerators, microwave ovens, rice cookers. And the household products go into our lives, we enjoy the material on those by making more and more people gradually reduce the daily activities. And the most prominent is the elevators and cars, reducing active time and the amount of activity, a lot of heat stored in the human body is causing various diseases, from the results of medical research over the past decade. Various diseases associated with obesity have been become the most important factor in human deaths worldwide, the rapid emergence of this disease and the rapid development of social science and technology form a clear lack of coordination, for which many experts at home and abroad for such issues of deep study and research.展开更多
Sphingosine-1-phosphate(S1P) is a bioactive lipid messenger in the cells that regulate gene expression and NF-κB signal pathway through unknown mechanisms.Recently,Cheng Luo,associate professor of DDDC in Shanghai ...Sphingosine-1-phosphate(S1P) is a bioactive lipid messenger in the cells that regulate gene expression and NF-κB signal pathway through unknown mechanisms.Recently,Cheng Luo,associate professor of DDDC in Shanghai Institute of Materia Medica,whose project was funded by the National Natural Science Foundation of China,joined in a research team led by Professor Sarah Spiegel of Virginia Commonwealth University.The team continuously made significant breakthroughs in understanding the regulation mechanism of Sphingosine-1- Phosphate.In September 2009,in a paper published on SCIENCE magazine(Science 2009, 325:1254-7),they firstly demonstrated that S1P is a physiologically important regulator of histone deacetylases(HDACs),HDACs are direct intracellular targets of S1P.Furthermore,they identified the mechanism that S1P regulates gene expression through regulating the activity of HDACs.In June 24th,2010,in another paper to be published on NATURE magazine(Nature 2010,June 24th,advance online publication,(doi:10.1038/ nature09128)) which reports the regulation of NF-κB signaling pathway by S1P.They demonstrate that S1P is the missing cofactor for TRAF2(tumour-necrosis factor receptor-associated factor 2) and indicate a new paradigm for the regulation of lysine-63- linked poly-ubiquitination.The study also highlight the key role of SphK1 and its product S1P in TNF-αsignalling and the canonical NF-κB activation pathway, and then play crucial role in inflammatory,antiapoptotic and immune processes.The identification of new mechanisms fay which S1P regulates gene expression and TNF and NF-κB signaling pathway will light up the road to develop novel inhibitors that might be useful for treatment of cancer and in- flammatory diseases.展开更多
