神经干细胞移植量对PST大鼠神经病理性疼痛的影响 被引量:2

目的观察不同数量神经干细胞(NSCs)鞘内注射对坐骨神经半切断(PST)大鼠神经病理性疼痛及脊髓背角、背根神经节(DRG)胶质源性神经营养因子(GDNF)表达的影响。方法 84只成年雄性SD大鼠随机均分为七组:假手术组(Ⅰ组)、PST组(Ⅱ组)、PST+NS... 目的观察不同数量神经干细胞(NSCs)鞘内注射对坐骨神经半切断(PST)大鼠神经病理性疼痛及脊髓背角、背根神经节(DRG)胶质源性神经营养因子(GDNF)表达的影响。方法 84只成年雄性SD大鼠随机均分为七组:假手术组(Ⅰ组)、PST组(Ⅱ组)、PST+NSCs1×103组(Ⅲ组)、PST+NSCs1×104组(Ⅳ组)、PST+NSCs1×105组(Ⅴ组)、PST+NSCs1×106组(Ⅵ组)、PST+NSCs1×107组(Ⅶ组)。NSCs于术后3d鞘内注射。记录术前1d、术后1、3、7、14、21d机械痛及热痛阈;术后7、21d免疫组织化学与RT-PCR检测同侧脊髓背角及DRGGDNF表达。结果与Ⅰ组比较,Ⅱ~Ⅶ组术后1d痛阈下降,术后7、14d最低(P<0.05);与Ⅱ组比较,术后7dⅣ、Ⅴ、Ⅵ、Ⅶ组随着NSCs量的增加,痛阈逐渐升高,脊髓背角及DRGGDNF表达逐渐上调(P<0.05);术后21d,Ⅴ、Ⅵ、Ⅶ组痛阈均与术前差异无统计学意义,但其GDNF表达Ⅴ组最高(P<0.05)。结论鞘内注射1×105NSCs能最有效缓解PST大鼠神经病理性疼痛。 展开更多

背景:有研究表明神经干细胞移植治疗神经病理性疼痛有一定效果,但神经干细胞移植数量对神经病理性疼痛的疗效并不确切。目的:观察不同数量神经干细胞鞘内注射对坐骨神经半切断大鼠神经病理性疼痛及脊髓背角、背根神经节胶质源性神经... 背景:有研究表明神经干细胞移植治疗神经病理性疼痛有一定效果,但神经干细胞移植数量对神经病理性疼痛的疗效并不确切。目的:观察不同数量神经干细胞鞘内注射对坐骨神经半切断大鼠神经病理性疼痛及脊髓背角、背根神经节胶质源性神经营养因子表达的影响。方法:选取1只孕14-16 d SD大鼠制备神经干细胞悬液,分别以细胞数1×10^-3,1×10^-4,1×10^-5,1×10^-6,1×10^-7经坐骨神经半切断伤模型大鼠鞘内注入30μL,并设置模型组和假手术组作对照。记录术前1 d,术后1,3,7,14,21 d机械痛及热痛阈;术后7,21 d免疫组织化学与RT-PCR检测同侧脊髓背角及背根神经节胶质源性神经营养因子的表达。结果与结论:与假手术组比较,其他各组术后1 d痛阈下降,术后7,14 d最低(P〈0.05);与模型组比较,术后7 d,随着移植神经干细胞的浓度不断增加,神经干细胞1×10^-4,1×10^-5,1×10^-6,1×10^-7组痛阈逐渐升高,脊髓背角及背根神经节胶质源性神经营养因子表达逐渐上调(P〈0.05);术后21 d,神经干细胞1×10^-5,1×10^-6,1×10^-7组痛阈均与术前差异无显著性意义,但其胶质源性神经营养因子表达在神经干细胞1×10-5组最高(P〈0.05)。说明鞘内注射神经干细胞能缓解大鼠的神经病理性疼痛,注射剂量以1×10^-5神经干细胞效果最佳。 展开更多

Study on Fractionated Total Body Irradiation before Hematopoietic Stem Cell Transplantation

OBJECTIVE To observe the dose and the complicationsfrom total body irradiation before hematopoietic stem celltransplantation.METHODS This study involved 312 patients with total bodyirradiation before hematopoietic stem cell transplantation. Theywere entered into the treated research from May 1999 to October2005. All patients had received the irradiation from ^(60)Co of anabsorbed dose rate of (5.2 ± 1.13) cGy/min. The total dose of TBIwas 7~12 Gy, 1 f/d × 2 d. A high-dose rate group (≥10 Gy) included139 cases and a low-dose rate group (< 10 Gy) included 173 cases.RESULTS The probability of acute gastrointestinal reactionsin the high-dose rate group was more compared with that in thelow-dose rate group. The differences for other reactions, such ashematopoietic reconstitution and graft survival rate, between thetwo groups were insignificant.CONCLUSION Using fractional total body irradiation at a doserate of 5 cGy/min, with a total dose of 7~12 Gy, 1 f/d × 2 d , withthe lung receiving under 7.5 Gy is a safe and effective pretreatmentfor hematopoietic stem cell transplantation. 展开更多

OBJECTIVE:To investigate the effect of Soothing liver therapy on infertile women undergoing in vitro fertilization and embryo transfer(IVF-ET)and to explore its mechanism.METHODS:Fifty-eight women with tubal infertility were randomized into two groups:30 in an experimental group treated with Xiaoyao powder(Shugan)plus gonadotropin-releasing hormone analog(GnRHa)/follicle-stimulating hormone(FSH)/human chorionic gonadotropin(hCG)and 28 in the control group who were treated with GnRHa/FSH/hCG only.The total gonadotropin(Gn)doses required,endometrial thickness,oocyte numbers,high quality embryo production rate and pregnancy rate of the two groups were compared.The concentration of growth differentiation factor-9(GDF-9)in follicular fluid was detected by western blotting and the expression of GDF-9 mRNA in granulosa cells was measured using reverse tran-scription-polymerase chain reaction amplification.RESULTS:In the experimental group,the Gn dose was significantly lower than that in the control group;the endometrial thickness,high quality embryo production and pregnancy rates were significantly higher and the expression of GDF-9 mRNA was also significantly higher than in the control group(all P<0.05).CONCLUSION:Shugan treatment can improve the pregnancy rate of women with tubal infertility;its mechanism is possibly related to the increased expression of GDF-9 in granulosa cells. 展开更多
