低聚木糖对肉鸭空肠菌群的影响 被引量:2

将1600只1日龄健康雏鸭按完全随机设计分为试验分组1和试验分组2,每个试验分组各有1个对照组和3个试验组,每试验组再分4个重复栏饲养,进行添加低聚木糖(Xylo-oligosaccharides,XOS)的饲养试验,试验时间42d。试验结束后,每组每个重复中... 将1600只1日龄健康雏鸭按完全随机设计分为试验分组1和试验分组2,每个试验分组各有1个对照组和3个试验组,每试验组再分4个重复栏饲养,进行添加低聚木糖(Xylo-oligosaccharides,XOS)的饲养试验,试验时间42d。试验结束后,每组每个重复中随机抽取2只(每组8只)体质量基本一致的健康鸭进行屠宰,立即剖腹取出肠道,结扎取样部位,于实验室内无菌采取空肠中部食糜以备分析。结果表明,在添加大蒜素的基础上,添加XOS仍能明显降低肠道大肠杆菌的数量,增加乳酸菌的数量,表现出同体外试验一致的效果。添加100g/tXOS与其他有益添加物配伍后,仍能明显降低肠道大肠杆菌的数量,但在增加乳酸菌的数量方面表现还不一致,机制有待今后进一步研究。 展开更多

旨在研究中药复方(苦参、百草霜、伏龙肝)对黄羽肉鸡生长性能、器官指数及生长过程中空肠微生物菌群变化的影响。选取1日龄快大型黄羽肉鸡180只,按体质量(BW)随机分为2个组,每组6个重复,每个重复15羽,对照组(CON组)饲喂基础日粮,试验组(... 旨在研究中药复方(苦参、百草霜、伏龙肝)对黄羽肉鸡生长性能、器官指数及生长过程中空肠微生物菌群变化的影响。选取1日龄快大型黄羽肉鸡180只,按体质量(BW)随机分为2个组,每组6个重复,每个重复15羽,对照组(CON组)饲喂基础日粮,试验组(CHM组)在基础日粮中添加中药复方(1 g/kg),试验期42 d。采集1,14,28和42日龄肉鸡组织器官和空肠内容物,计算器官指数,分析空肠微生物菌群多样性的变化及其代谢途径差异性。结果表明:CHM组显著提高29-42日龄和1-42日龄肉鸡的BW、平均日增质量(ADG),显著降低F/G(P<0.05)。CHM组显著提高14 d肉鸡肝脏和肾脏指数,28 d胸腺指数以及28和42 d法氏囊指数(P<0.05)。CHM组在14 d的Chao 1、Observed species和Shannon指数显著高于CON组(P<0.05)。CHM组在14 d对软壁菌门(Tenericutes)、在42 d对拟杆菌门(Bacteroidetes)和蓝细菌门(Cyanobacteria)的丰度显著升高,在14 d对糖杆菌门(TM7)丰度显著降低(P<0.05)。CHM组在14 d肠杆菌(Enterococcus)丰度显著降低,在28 d梭菌科梭菌属(Clostridiaceae_Clostridium)的丰度显著升高(P<0.05)。CHM组显著促进次生代谢物的生物合成(类黄酮生物合成)、氨基酸代谢(丙氨酸、天冬氨酸和谷氨酸代谢的生物合成)和消化系统(蛋白质消化和吸收)途径的富集(P<0.05)。综上,中药复方可以提高肉鸡的生长性能,降低料肉比以及提高器官指数,同时能改变早期空肠微生物菌群的多样性和丰富度,并在营养物质的代谢中发挥作用。 展开更多

本试验旨在研究凉茶渣替代象草对育肥牛空肠组织形态、屏障功能以及菌群结构的影响。选取120头健康且体重相近的西门塔尔杂交肉牛,随机分为2个组,分别为对照组(饲喂基础饲粮)、试验组(50%凉茶渣替代部分象草),每组3个重复,每个重复20头... 本试验旨在研究凉茶渣替代象草对育肥牛空肠组织形态、屏障功能以及菌群结构的影响。选取120头健康且体重相近的西门塔尔杂交肉牛,随机分为2个组,分别为对照组(饲喂基础饲粮)、试验组(50%凉茶渣替代部分象草),每组3个重复,每个重复20头。试验期共67 d,包括7 d的预试期,60 d的正试期。在试验期结束后,采集试验牛血液、空肠组织、空肠内容物,通过苏木精-伊红(HE)染色测量空肠绒毛高度、隐窝深度;通过酶联免疫吸附测定(ELISA)试剂盒检测血清中炎症因子、D-乳酸(D-Lac)以及二胺氧化酶(DAO)浓度;通过荧光定量PCR测定空肠紧密连接蛋白基因闭合小环蛋白-1(ZO-1)、闭锁蛋白(Occludin)、闭合蛋白-1(Claudin-1)的mRNA表达水平;利用16S rDNA高通量测序分析空肠菌群结构。结果表明:1)与对照组相比,试验组育肥牛空肠绒毛高度显著增加(P<0.05),血清中D-Lac浓度显著降低(P<0.05),空肠紧密连接蛋白ZO-1的mRNA表达水平显著上调(P<0.05)。2)在门水平上,对照组和试验组厚壁菌门(Firmicutes)相对丰度分别为85.79%、76.96%,拟杆菌门(Bacteroidetes)相对丰度分别为0.54%、3.11%。在属水平上,对照组和试验组之间罗姆布茨菌属(Romboutsia)、罗斯氏菌属(Rothia)、布劳特氏菌属(Blautia)等相对丰度表现出显著差异(P<0.05)。3) Pearson相关性分析表明,空肠绒毛高度与Blautia、Rothia、短波单胞菌属(Brevundimonas)、四球虫属(Tetrasphaera)相对丰度呈显著正相关(P<0.05),相关系数为0.46~0.57;血清中D-Lac浓度与Romboutsia、土孢杆菌属(Terrisporobacter)相对丰度呈显著正相关(P <0.05),相关系数分别为0.56、0.62;空肠紧密连接蛋白ZO-1的mRNA表达水平与梭菌属(Solobacterium)相对丰度呈显著负相关(P<0.05),相关系数为-0.50。由此可见,饲粮中添加50%凉茶渣作为功能性粗饲料对育肥牛空肠绒毛高度、屏障功能有积极的影响,凉茶渣可能通过改变空肠菌群组成和相对丰度,进而影响空肠组织形态与屏障功能,最终促进肠道健康。 展开更多

亮氨酸对哺乳期湖羊羔羊小肠发育的影响

本试验旨在研究亮氨酸对哺乳期湖羊羔羊小肠组织形态、屏障功能及菌群组成的影响。选取30只体重[(3.60±0.37)kg]相近的5日龄湖羊羔羊,随机分成对照组(Con组,未添加亮氨酸)和亮氨酸组(Leu组,添加0.66 g/d亮氨酸),每组5个重复,每个重... 本试验旨在研究亮氨酸对哺乳期湖羊羔羊小肠组织形态、屏障功能及菌群组成的影响。选取30只体重[(3.60±0.37)kg]相近的5日龄湖羊羔羊,随机分成对照组(Con组,未添加亮氨酸)和亮氨酸组(Leu组,添加0.66 g/d亮氨酸),每组5个重复,每个重复3只羔羊。试验期25 d。结果显示:1)与Con组相比,Leu组的空肠绒毛高度有增加的趋势(P=0.073),空肠隐窝深度显著降低(P<0.05),空肠绒毛高度/隐窝深度显著增加(P<0.05);回肠绒毛高度显著增加(P<0.05)。2)与Con组相比,Leu组的空肠上皮超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物(GSH-Px)的基因相对表达量均显著升高(P<0.05)。3)与Con组相比,Leu组的空肠上皮咬合蛋白(Occludin)和闭锁小带蛋白-1(ZO-1)的基因和蛋白相对表达量均显著或极显著升高(P<0.05或P<0.01)。4)与Con组相比,Leu组的空肠布雷德菌属(Bulleidia)的相对丰度有增加的趋势(P=0.076),而空肠颤螺旋菌属(Oscillospira)和链球菌属(Streptococcus)的相对丰度显著降低(P<0.05)。5)相关性分析结果显示,空肠Bulleidia的相对丰度与空肠ZO-1的基因相对表达量呈极显著正相关(P<0.01),空肠Streptococcus的相对丰度与空肠隐窝深度呈极显著正相关(P<0.01)。由此可见,添加亮氨酸可增强湖羊羔羊空肠抗氧化能力,调控肠道上皮紧密连接蛋白的表达,改变空肠菌群结构,从而促进哺乳期羔羊小肠发育。 展开更多

The results showed that number of FCB in marine water samples ranged from no detection ( 〈2 cfu/100 mL) to 540 cfu/100 mL in Luoyuan Bay. Values of FCB during August to October were significantly higher than those during January to April during three year period. Monthly changes of FCB values at each year period were mainly due to monthly precipitation. In addition, compared with FCB values in difference sample sites, values of FCB in Bay-heed were significantly higher than those in middle of bay and Bay-mouth, Luoyuan Bay. However, values of FCB in Bay-mouth were significantly less than those in middle of bay. The ratios to the par of FCB in 2003, 2004, and 2005 years were 100%, 98.0%, and 97.9%, respectively. Therefore, we considered that the pollution of FCB of surface marine water in Luoyuan Bay was not serious. 展开更多

AIM: To studythe role of intestinal flora in inflammatory bowel disease (IBD).METHODS: The spatial organization of intestinal flora was investigated in normal mice and in two models of murine colitis using fluorescence in situ hybridization.RESULTS: The murine small intestine was nearly bacteriafree. The normal colonic flora was organized in three distinct compartments (crypt, interlaced, and fecal), each with different bacterial compositions. Crypt bacteria were present in the cecum and proximal colon. The fecal compartment was composed of homogeneously mixed bacterial groups that directly contacted the colonic wall in the cecum but were separated from the proximal colonic wall by a dense interlaced layer. Beginning in the middle colon, a mucus gap of growing thickness physically separated all intestinal bacteria from contact with the epithelium. Colonic inflammation was accompanied with a depletion of bacteria within the fecal compartment, a reduced surface area in which feces had direct contact with the colonic wall, increased thickness and spread of the mucus gap, and massive increases of bacterial concentrations in the crypt and interlaced compartments. Adhesive and infiltrative bacteria were observed in inflamed colon only, with dominant Bacteroides species.CONCLUSION: The proximal and distal colons are functionally different organs with respect to the intestinal flora, representing a bioreactor and a Segregation device.The highly organized structure of the colonic flora, its specific arrangement in different colonic segments, and its specialized response to inflammatory stimuli indicate that the intestinal flora is an innate part of host immunity that is under complex control. 展开更多

Levels of pollutants at Zomar stream, the only recreational area for the inhabitants of the northern part of the West Bank, Palestine, were recently determined, and have shown to be relatively high. Three bathing sites along the Zomar stream area were monitored for one year (fortnightly). Wastewater samples were subjected to microbiological analysis (fecal coliforms, Escherichia coli, Pseudomonas, Enterococcus and Klebsiella), physico-chemical analyses (water temperature, pH, electroconductivity and DO (dissolved oxygen)), biochemical oxygen demand, total Kjeldahl nitrogen and ammonia. The study results demonstrated seasonal and locational variations in all parameters studied. The highest levels of pollution were detected during summer, especially before a rainfall or after a discharge from onsite cesspits. Locations associated with sewage discharge were found to have the highest fecal indicator levels. 展开更多
