In the present study full-length cDNA of N...Previously the partial sequence of an ethylene receptor gene NTHK2 was isolated from tobacco (Nicotiana tabacum L. var. Xanthi) plants and it was wound and drought inducible. In the present study full-length cDNA of NTHK2 was cloned by 5'-RACE method. NTHK2 gene has 3 216 bp, with 509 bp of 5'-non-coding region and 427 lip of 3'-non-coding region, and encodes an ethylene-receptor homolog of 760, amino acids. NTHK2 protein has a putative signal peptide, three transmembrane domains, a histidine kinase domain and a receiver domain. In the putative histidine kinase domain, the histidine at the phosphorylation site was replaced by an asparagine. To study the biochemical property of NTHK2, its kinase domain was expressed as a fusion protein with glutathione S-transferase (GST) using yeast Schizzosaccharomyces pombe as an expression system. In vitro kinase assay showed that NTHK2 kinase domain can autophosphorylate in the presence of Mg2+, indicating that NTHK2 may function as a kinase. Further studies will elucidate the function of NTHK2 in plant.展开更多

To improve document clustering, a document similarity measure based on cosine vector and keywords frequency in documents is proposed, but also with an input ontology. The ontology is domain specific and includes a list of keywords organized by degree of importance to the categories of the ontology, and by means of semantic knowledge, the ontology can improve the effects of document similarity measure and feedback of information retrieval systems. Two approaches to evaluating the performance of this similarity measure and the comparison with standard cosine vector similarity measure are also described.展开更多

A 165 kD polypeptide, which is equivalent to XCAP-C in molecular weight, was demonstrated in the nuclei by SDS-PAGE, and was then proved to be an XCAP-C-like protein by Western blot using an anti-XCAP-C antiserum, but neither the polypeptide nor the XCAP-C-like protein was detected in die nuclear matrix. The nuclei, Chromosomes and chromosome scaffolds were observed to emanate strong, specific fluorescence after labeled with the anti-XCAP-C antiserum and an FITC-conjugated secondary antibody, indicating their containment of the XCAP-C-like protein. It was confirmed by viewing with immunoelectron microscopy that the gold particles representing the localization of the XCAP-C-like protein were found to be mainly distributed in the condensed chromatin regions of the nuclei and chromosomes.展开更多

The part-level representation is implemented, which enables a more compact shape description of 3-D objects. The proposed classification method consists of two key processing stages: the improved constrained search on an interpretation tree and the following shape similarity measure computation. By the classification method, both whole match and partial match with shape similarity ranks are achieved; especially, focus match can be accomplished, where different key parts may be labeled and all the matched models containing corresponding key parts may be obtained. A series of experiments show the effectiveness of the presented 3-D object classification method.展开更多

The immunohistochemical technique was performed to detect the expression of like-muscarinic acetylcholine receptor M2 in the brain of three castes of Polyrhachis vicina. A positive expression of like-muscarinic acetylcholine receptor M2 was observed in the mushroom body, central body and antennal lobes of the ant brain; but there is great diversity in their location and intensity among worker, queen and male ants. It is speculated that like-muscafinic acetylcholine receptor M2 plays a critical role in the central nervous system, in terms of projecting visual information and olfactory information into the protocerebrum and integrating many inputs.展开更多
AIM To develop a MRI-based method for accurate determination of liver volume(LV) and to explore the effect of long-term everolimus(EVR) treatment on LV in PCK rats with hepatomegaly. METHODS Thirty-one female PCK rats...AIM To develop a MRI-based method for accurate determination of liver volume(LV) and to explore the effect of long-term everolimus(EVR) treatment on LV in PCK rats with hepatomegaly. METHODS Thirty-one female PCK rats(model for polycystic-liverdisease: PCLD) were randomized into 3 groups and treatment was started at 16 wk, at the moment of extensive hepatomegaly(comparable to what is done in the human disease). Animals received: controls(n = 14), lanreotide(LAN: 3 mg/kg per 2 wk)(n = 10) or everolimus(EVR: 1 mg/kg per day)(n = 7). LV was measured at week 16, 24, 28. At week 28, all rats were sacrificed and liver tissue was harvested. Fibrosis was evaluated using quantitative image analysis. In addition, gene(quantitative RT-PCR) and protein expression(by Western blot) of the PI3K/Ak T/m TOR signaling pathway was investigated. RESULTS LV determination by MRI correlated excellent with the ex vivo measurements(r = 0.99, P < 0.001). The relative changes in LV at the end of treatment were:(controls) +31.8%;(LAN) +5.1% and(EVR) +8.8%, indicating a significantly halt of LV progression compared with controls(respectively, P = 0.01 and P = 0.04). Furthermore, EVR significantly reduced the amount of liver fibrosis(P = 0.004) thus might also prevent the development of portal hypertension. There was no difference in phosphorylation of Akt(Threonine 308) between LAN-treated PCK rats control PCK rats, whereas S6 was significantly more phosphorylated in the LAN group. Phosphorylation of Akt was not different between controls and EVR treated rats, however, for S6 there was significantly less phosphorylation in the EVR treated rats. Thus, both drugs interact with the PI3K/Ak T/m TOR signaling cascade but acting at different molecular levels.CONCLUSION Everolimus halts cyst growth comparable to lanreotide and reduces the development of fibrosis. m TORinhibition should be further explored in PCLD patients especially those that need immunosuppression.展开更多
