【目的】了解新疆不同动物源喹诺酮类耐药大肠杆菌携带质粒介导的喹诺酮耐药因子(plasmid mediated quinolone resistance,PMQR)的流行现状,及其与主要的β-内酰胺酶基因和16S rRNA甲基化酶基因的共存情况。【方法】通过PCR方法对猪源(7... 【目的】了解新疆不同动物源喹诺酮类耐药大肠杆菌携带质粒介导的喹诺酮耐药因子(plasmid mediated quinolone resistance,PMQR)的流行现状,及其与主要的β-内酰胺酶基因和16S rRNA甲基化酶基因的共存情况。【方法】通过PCR方法对猪源(79株)、牛源(8株)和羊源(96株)喹诺酮类(环丙沙星,诺氟沙星和恩诺沙星)耐药大肠杆菌进行PMQR(qnrA,qnrB,qnrC,qnrD,qnrS,qepA,oqxA,oqxB,aac(6’)-Ib-cr)、β-内酰胺酶基因(blaTEM,blaCTX-M,blaSHV,blaKPC,blaCMY-2,blaLAP-1)及16S rRNA(armA,rmtB)等基因检测,对目的条带进行DNA测序,确定阳性菌株,对检出携带β-内酰胺酶基因和16S rRNA甲基化酶基因的大肠杆菌进行β-内酰胺类及氨基糖苷类药敏试验,分析其携带的基因型与耐药表型之间的关系。【结果】猪源大肠杆菌中检出的PMQR因子为qnrS(5/79,6.33%),oqxA(35/79,44.30%),oqxB(40/79,50.60%)和aac(6’)-Ib-cr(4/79,5.06%),检出的β-内酰胺酶基因为blaTEM(79/79,100%),检出的16S rRNA基因为rmtB(3/79,3.80%);牛源大肠杆菌中检出的PMQR因子为qnrS(1/8,12.50%),oqxA(1/8,12.50%),oqxB(1/8,12.50%)和aac(6’)-Ib-cr(1/8,12.50%)和qepA(1/8,12.50%),检出的β-内酰胺酶基因为blaTEM(8/8,100%)和blaSHV(1/8,12.50%);羊源大肠杆菌中检出的PMQR因子为qnrS(6/96,6.25%),oqxA(32/96,33.3%),oqxB(37/96,38.5%)和aac(6’)-Ib-cr(22/96,22.91%),检出的β-内酰胺酶基因为blaTEM(96/96,100%)和blaCTX-M(2/96,2.08%),检出的16S rRNA基因为rmtB(2/96,2.08%);未检出qnrA,qnrB,qnrC,qnrD,blaKPC,blaCMY-2和blaLAP-1被检基因。不同动物源大肠杆菌中存在基因共存现象,携带β-内酰胺酶基因和16S rRNA甲基化酶基因的喹诺酮类耐药大肠杆菌对β-内酰胺类和氨基糖苷类药物耐药呈一定的相关性。【结论】不同动物源喹诺酮类耐药大肠杆菌中存在PMQR因子,可与主要的β-内酰胺酶和/或16S rRNA甲基化酶基因共存。此外,首次在羊源大肠杆菌中检出PMQR因子、β-内酰胺酶及16S rRNA甲基化酶基因。 展开更多

【目的】研究不同禽源碳青霉烯类耐药大肠杆菌(carbapenem-resistant Escherichia coli,CREC)的群间分布、耐药特征及菌株间亲缘关系,为阻断CREC的潜在危害提供技术支持。【方法】在胶东地区采集肉鸡、蛋鸡、水禽三类家禽泄殖腔拭子1131... 【目的】研究不同禽源碳青霉烯类耐药大肠杆菌(carbapenem-resistant Escherichia coli,CREC)的群间分布、耐药特征及菌株间亲缘关系,为阻断CREC的潜在危害提供技术支持。【方法】在胶东地区采集肉鸡、蛋鸡、水禽三类家禽泄殖腔拭子1131份,利用选择性分离培养、质谱鉴定、PCR、微量肉汤稀释法、多位点序列分型(MLST)及全基因组测序(WGS)等方法进行CREC菌株鉴定与分析。【结果】共分离出364株bla NDM基因阳性的CREC菌株,分离率为32.18%,阳性场总体占比为76.32%,肉鸡场和个体阳性率均最高,分别为93.33%和55.56%。三类禽源菌株均多重耐药,83.65%菌株对8类及以上药物同时耐药,对多数测试药物耐药率在80%以上。肉鸡多重耐药问题最严重,水禽次之,蛋鸡相对较轻。45株全基因组测序的CREC菌株携带12类52种耐药基因,4种bla NDM变异体,以bla NDM-5(77.78%)为主,三类家禽对同类耐药基因的检出率存在差异。共检出46种ST型,多样性比为44.23%。三类家禽CREC菌株间单核苷酸多态性(SNPs)差异位点为82~71307个,且携带的优势型不同。【结论】携带bla NDM的CREC在不同家禽养殖场尤其是肉鸡场广泛存在,以bla NDM-5亚型最为常见,对多种抗菌药普遍耐药,且携带大量耐药基因,以质粒传播为主。CREC菌株在三类家禽中呈多样化分布,且多数菌株间亲缘关系相差较远。提示应针对不同动物群体,加强监测和影响因素研究,以降低CREC的潜在风险。 展开更多

目的调查正常粪便中大肠埃希菌(E.coli)的耐药性及产超广谱β内酰胺酶(ESBLs)菌株的检出率,了解普通人群体内肠道菌对常用抗菌剂的耐药性。方法对来自骨科初入院患者、常规筛查结果正常的粪便标本进行培养,分离获得E.coli菌株后进行药... 目的调查正常粪便中大肠埃希菌(E.coli)的耐药性及产超广谱β内酰胺酶(ESBLs)菌株的检出率,了解普通人群体内肠道菌对常用抗菌剂的耐药性。方法对来自骨科初入院患者、常规筛查结果正常的粪便标本进行培养,分离获得E.coli菌株后进行药敏试验及ESBLs表型确证试验。结果分离获得的222株E.coli中产ESBLs菌株87株(占39.2%)。所有菌株仅对阿米卡星、头孢哌酮/舒巴坦、亚胺培南和美罗培南的敏感率大于90%。产ESBLs菌株仅对阿米卡星、亚胺培南、美罗培南的敏感率大于90%。结论医院应对携带产ESBLs菌株的初入院患者制定完善的管理流程,防止菌株播散,以控制或避免医院感染;非胃肠疾病、且入院后2日内未大量使用抗菌剂的患者肠道基础菌的耐药性较高,治疗后耐药性有可能更高,需加强对抗菌剂使用的管理。 展开更多

Mucosal permeability to lipopolysaccharides in the colon in chronic alcoholic rats

AIMS To evaluate the effects of chronic alcohol abuse on the mucosal permeability to lipopolysaccharide in the colon in rats. METHODS Escherichia coil lipopolysaccharide (LPS,20 μg/ml) was injected into the colon of chronic alcoholic rats (n=10) and the rats were supplied with Lieber diets every other day for 6 weeks. Before LPS injection and 5,10,20,30 minutes after injection, blood samples from the portal vein were obtained and contents of LPS in the blood were measured. The dis- tribution of LPS in the colon tissues was observed with a confocal laser scanning microscope by immunofluo- rescent technique using a monoclonal antibody specific to the lipid A region of LPS. Normal rats were used as controls (n=6). RESULTS Before LPS injection in the colon,LPS levels in the blood of portal vein of chronic alcoholic rats were significantly higher than those of normal con- trols (3.56±0.67 pg/ml,vs 2.45±0.15 pg/ml,P <0.01). At 5,10,20,30 minutes after injection of LPS,LPS contents were significantly higher than those before LPS injection (173.56±23.45 pg/ml,154.78 ±20.57 pg/ml,43.89±8.67 pg/ml,45.38± 7.89 pg/mls vs 3.56±0.67 pg/ml,P<0.01 respectively). Most mucosal cells showed strong posi- tive reactions to LPS in the rats of chronic alcohol abuse,but no significant changes of LPS contents in blood from the portal vein and fluorescent reactions to LPS in mucosal cells of normal rats were found after LPS injection. CONCLUSIONS Chronic alcohol abuse resulted in a significant increase of permeability to LPS in colon mu- cosal cells in rats. 展开更多

The morphology and structure of the olfactory organ of Cynoglossus semilaevis Gunther are described. The oval olfactory sacs on both sides differ in size and in the number of lamellae, With those on the abocular side having smaller sacs and fewer lamellae than those on the ocular side. On the ocular side, the average ratio of sac length to eye diameter is 2.1 (i.e.〉1) with an average of 91 lamellae, while on the abocular side, the values were 1.7 (i.e.〉1) and 69, respectively. In addition, the surface morphology varies in different parts of the lamella. The frontal part, near the anterior nostril, is a non-sensory margin with cilia-free epidermal cells. Within this is an internal ciliated sensory area, which is intercalated with ciliated receptor cells and a few ciliated non-sensory cells. Additionally, some dense ciliated non-sensory cells make up a non-sensory area, which also contains cilia-free epidermal cells distributed in patches. In the rear of the olfactory sac near the posterior nostril, the lamellae differ in morphology from those of the frontal olfactory sac but are similar in having few ciliated receptor cells. In other words, the surface of the lamellae in the rear part of the olfactory sac is mainly non-sensory. At present, four types of lamellae (~ E IlIand IV) have been recognized in relation to the pattern of the sensory epithelium. In this study, the frontal and rear lamellae resembled types I and IV, respectively, but are referred to as types r and IV because they are slightly less developed. Data on the ratio of length of lamellae to eye diameter, number of lamellae and the type of surface pattern of the lamellae show that the development of the olfactory system of C. semilaevis facilitates prey capture. 展开更多

In order to improve the production of human-like collagen III(HLC III)by fed-batch culture of recombinant Escherichia coli BL21,the Plackett-Burman and Box-Behnken design were applied to optimize the fermentation process parameters.Three variables(induction time,inoculum age and pH),which have significant effects on HLC III production,were selected from eight variables by Plackett-Burman design.With the regression coefficient analysis in the Box-Behnken design,a relationship between HLC III production and three significant factors was obtained,and the optimum levels of the three variables were as follows:induction time 3.2h,inoculum age 12.6 h and pH 6.7.The 3D response surface plots and 2D contour plots created by the Box-Behnken design showed that the interaction between induction time and pH and that between innoculum age and pH were significant.An average 9.68 g·L1HLC III production was attained in the validation experiment under optimized condition,which was 80%higher than the yield of 5.36 g·L1before optimization. 展开更多

The kinetics of batch and fed-batch cultures of recombinant Escherichia coli producing human-like collagen was investigated. In the batch culture, a kinetic model of a simple growth-association system was concluded without consideration of cell endogeneous metabolism. The cell lag time, the maximum specific growth rate and Yx/s were determined as 1.75h, 0.65h^-1 and 0.51g·g^-1, respectively. In the fed-batch culture, different specific growth rates were set at (0.15, 0.2, 0.25h^-1) by the method of pseudo-exponential feeding, and the expressions for the specific rate of substrate consumption, the growth kinetics and the product formation kinetics of each phase were obtained. The result shows that the concentrations of cell and product can reach 77.5g·L^-1 and 10.2g·L^-1 respectively. The modal predictions are in good agreement with the experimental data. 展开更多

Objective: To amplify human era (Hera) gene, then express it in E.coli. Methods: Human era gene, after amplified by PCR and identified by sequencing, was inserted into the expression vector pGEX-4T3 in which exogenous gene was controlled by Ptac promoter. The recombinant plasmid pGEX-Hera was transformed into DH5 (and induced with IPTG chemically. Results: The human era gene was amplified and the sequence was correct. When the bacteria with pGEX-Hera was induced, an anticipated 65 000 protein band appeared on SDS-PAGE gel and amounted to 23% of total bacterial protein. Conclusion: The human era gene has been successfully amplified and efficiently expressed in E.coli. 展开更多

Designed based on the exponential reaching law, paratmter e of conventional sliding mode controller is the key factor of system chattering, and is ~onal to it as well. In view of this, fuzzy control theory was introduced into the design to ensure the real- time adjusting of parameter e. Sinmlation results show that the sys tem chattering is eliminated perfectly, and the dynamic performance of the control system is improved effectively. 展开更多

目的调查武汉大学人民医院分离的肠杆菌科细菌中质粒介导喹诺酮类耐药基因qnrA、qnrB、qnrS基因的现状、基因型以及qnr基因阳性菌株所携带的广谱B内酰胺酶基因型。方法PCR法对179株肠杆菌科细菌(129株大肠埃希菌、37株肺炎克雷伯菌和1... 目的调查武汉大学人民医院分离的肠杆菌科细菌中质粒介导喹诺酮类耐药基因qnrA、qnrB、qnrS基因的现状、基因型以及qnr基因阳性菌株所携带的广谱B内酰胺酶基因型。方法PCR法对179株肠杆菌科细菌(129株大肠埃希菌、37株肺炎克雷伯菌和13株阴沟肠杆菌)进行qnrA、qnrB、qnrS基因检测。KB纸片法检测对15种抗菌药的体外抗菌活性。琼脂平皿二倍稀释法检测阳性菌株对环丙沙星的MIC值。质粒接合试验分析qnrA、qnrB、qnrS基因的水平转移能力。对qnr阳性株,检测Ⅰ类整合酶基因及SHV-1、TEM-1、CTX—M、OXA—Ⅰ、OXA-Ⅱ、OXA-Ⅲ、DHA、EBC型B内酰胺酶基因。结果179株肠杆菌科细菌中,共检测出含qnr基因阳性菌株25株(13.97%),包括6株(3.35%)含有qnrA基因,9株(5.02%)含有qnrB基因,10株(5.59%)含有qnrS基因。阳性株菌均对亚胺培南敏感且对多种抗生素耐药,其中2株qnrA阳性菌株和4株qnrB阳性菌株对喹诺酮类药物敏感。2株qnrA阳性菌株和4株qnrB及qnrS阳性菌株质粒接合成功。23株qnr阳性株Ⅰ类整合酶基因阳性,1株qnrB及qnrS阳性菌株Ⅰ类整合酶基因阴性,14株菌携带TEM-1型广谱β内酰胺酶基因、6株菌携带OXA—Ⅲ型广谱β内酰胺酶基因、7株菌携带EBC型AmpC酶。结论湖北地区肠杆菌科细菌中存在qnrA、qnrB、qnrS基因的流行,qnr阳性菌株呈多重耐药,且喹诺酮类敏感菌株中有qnr基因的存在。qnr阳性菌株可携带多种广谱β内酰胺酶基因。 展开更多
