The unicellular green alga Dunaliella is outstanding for its ability of massive accumulation of carotenoids. To elucidate the carotenoids synthesis pathway in this alga,phytoene desaturase(pds) gene cDNA together with...The unicellular green alga Dunaliella is outstanding for its ability of massive accumulation of carotenoids. To elucidate the carotenoids synthesis pathway in this alga,phytoene desaturase(pds) gene cDNA together with its DNA sequences were isolated and their structures and functions analyzed. The full-length pds cDNA of 2290 bp(GenBank Accession No. DQ243892) was de-duced from RACE results,including untranslated 21 bp 5'-and 520 bp 3'-flanking regions and an open reading frame of 582 amino acids,coding a protein of 64.196 kDa. The DNA sequence of 2908 bp(GenBank Accession No. DQ845248) including five introns was obtained. The fifth intron was uncompleted and complex,including two bases' perfect repeats(GT) 10 and large different-sized repeats within the last 400 bp. The Southern blot hybridization result demonstrated that this gene occurred as a single copy in this species,and the quantitative RT-PCR result showed that the transcription of this gene was constitutive. The viability of most tomato varieties cultivated in Libya have been tested to infect with potato spindle tuber viroid/potatoes (PSTVd) and its impact on growth and production of some of these varieties, which were ...The viability of most tomato varieties cultivated in Libya have been tested to infect with potato spindle tuber viroid/potatoes (PSTVd) and its impact on growth and production of some of these varieties, which were mechanically inoculated with Libyan isolate of viroid PSTVd as follows: Vlkato, Sankarh, Lebda, Jasmine, Kenza and Hana. The percent of incidence were 95.95%, 90%, 90.80%, 80% and 20%, respectively. The following varieties have been contagious mechanically with viroid of PSTVd: Vlkato, zahra, Toria, Lebda, Hoda, Farwa, Alkaraz, Naziha, Rim Star and Kartika. The percent of incidence were 95.95%, 85%, 85.80%, 80%, 70.40%, 0.0%, 0.0%, respectively. The varied symptoms of wrinkle, twist, warp, swell the veins of the leaves, dark brown spots formation and a large yellow spots turned into white patches. Also the effect of the Egyptian isolate viroid PSTVd in the growth and production of varieties Jasmine, Lebda, Soberhalim, and treasure No. 185 had been studied, as the average rates of decline in the production of the fruits tomatoes/tomato 43.4% and 17% length of plants, and in the fresh weight and dry root of the sum of 35% and 37% respictively.展开更多
Cowpea (Vigna unguiculata L. Walp.) and tomato (Lycopersicum esculentum L. Karst) are attacked by a wide range of insect pests worldwide. In many cases, pesticides are used to control the pests. In 2010 and 2011, ...Cowpea (Vigna unguiculata L. Walp.) and tomato (Lycopersicum esculentum L. Karst) are attacked by a wide range of insect pests worldwide. In many cases, pesticides are used to control the pests. In 2010 and 2011, we conducted field experiments to evaluate and compare the effectiveness of a biorational (Agroneem~) and conventional pesticide (imidacloprid or thiamethoxam) on insect pests of these crops. In the first year, two varieties of cowpea (Mississippi Silver and Pinkeye Purple Hull) and two of tomato (Mariana and German Johnson) were used; in the second year, the better performing of each group (Mississippi Silver and Mariana) were used for the study. A split-plot design with four replications in Year 1, and a randomized complete block design with six treatment combinations in Year 2. In the first year, both pesticide groups were applied following manufacturer's recommendation (10-14 d cycles); and in the second year, the application of the pesticides was driven by economic thresholds level (ETL) of insect pests. The most prevalent species of insects recorded in both crops were in the families: Chrysomelidae, Pentatomidae, Cicadellidae, Vespidae, Sarcophagidae, Thripidae and Sphingidae. In both years, the diversity of insects on cowpea was greater than on tomato and more insects were observed in the second year compared to the first, despite the absence of significant difference (P 〉 0.05) between varieties in the treated plots. The yield of cowpea and tomato was comparable in all sprayed plots.展开更多
