[ Method ] The free radical scavenging ability and reducing capacity of DPPH as well as inhibitory rate of nonenzymatic glycation reaction were measured targets for comparing changes of anti-oxidation and inhibitory effect on nonenzymatic glycation reaction of fermentation lyophilizer and fermentation extraction before and after EGB fermention adsorbed by AB-8 macroporous resin. The EGB fermention was biotransformed by Hericium erinaceus. [ Result ] After adsorbed by AB-8 macroporous resin, the bioactive matters were concentrated and separated. The free radical scavenging rate, reducing capacity and inhibitory rate of nonenzymatic glycation reaction were increased significantly after adsorbed by AB-8 macroporous resin. [ Conclusion] AB-8 macroporous resin could be used for preliminary concentration of EGB fermentation which was biotransformed by Hericium erinaceus.展开更多

However, the differences did not reach significance level. AIM:To investigate the changing patterns of glycogen and enzyme histochemical activities in rat liver graft under a different warm ischemia time (WIT) and to predict the tolerant time limitation of the liver graft to ...AIM:To investigate the changing patterns of glycogen and enzyme histochemical activities in rat liver graft under a different warm ischemia time (WIT) and to predict the tolerant time limitation of the liver graft to warm ischemia injury. METHODS: The rats were randomized into five groups, WIT was 0,15,30,45,60 min, respectively, and histochemical staining of liver graft specimens was observed. The recovery changes of glycogen and enzyme histochemistry activities were measured respectively 6 and 24 h following liver graft implantation. RESULTS: The activities of succinic dehydrogenase, cytochrome oxidase, apyrase (Mg++-ATPase) and content of glycogen were decreased gradually after different WIT in a time-dependent manner. The changes were significant when WIT was over 30 min. CONCLUSION: Hepatic injury is reversible within 30 min of warm ischemia injury. Glycogen and enzyme histochemistry activities of liver grafts and their recovery potency after reperfusion may serve as criteria to evaluate the quality of liver grafts.展开更多

METHODS:The antioxidant effect of COSs on pancreatic islet cells was detected under optical microscopy and with colorimetric assay and gel electrophoresis.The activities of glutathione peroxidase and superoxide dismutase,total antioxidant capacity,and content of malondialdehyde in serum and tissue slices of pancreas were examined after 60 d to determine the effect of COSs in streptozotocin-induced diabetes in rats. RESULTS:COSs can prohibit the apoptosis of pancreatic islet cells.All concentrations of COSs can improve the capability of total antioxidant capacity and activity of superoxide dismutase and decrease the content of malondialdehyde drastically.Morphological investigation in the pancreas showed that COSs have resulted in the reduction of islets,loss of pancreatic cells,and nuclear pyknosis of pancreatic cells. CONCLUSION:COSs possess various biological activities and can be used in the treatment of diabetes mellitus.展开更多
Sea cucumbers, Apostichopus japonicus Selenka, were fed diets containing non-immunostimulant (basal diet), 0.2% β-glucan and 0.02% glycyrrhizin in a recirculatory water system for 45 days, and subsequently challenged...Sea cucumbers, Apostichopus japonicus Selenka, were fed diets containing non-immunostimulant (basal diet), 0.2% β-glucan and 0.02% glycyrrhizin in a recirculatory water system for 45 days, and subsequently challenged with Vibrio splendidus by injection at 1.0×108 cfu/sea cucumber for 15 days.Phagocytic capacity (PC), intracellular superoxide anion production (ISAP), ly-sozyme (LSZ) activity and superoxide dismutase (SOD) activity in the coelomic fluid were analyzed on the 0th, 5th, 10th and 15th days after injection.Results showed that after the 45-day feeding period, PC, ISAP, LSZ activity and SOD activity in sea cucumbers fed with dietary β-glucan or glycyrrhizin were significantly higher than in those fed with the basal diet.On the 5th day after infection, all the immune parameters examined in the sea cucumbers injected with V.splendidus decreased in value significantly.On the 15th day, PC, ISAP and LSZ activity returned to levels similar to those on the 0th day.For the sea cucumbers injected with saline, there were no significant differences in all the immune parameters examined and in the cumulative morbidity during the 15-day challenging trial.After injecting with V.splendidus, the cumulative morbidity of sea cucumbers fed with the basal diet was significantly higher than those fed with dietary β-glucan or glycyrrhizin when challenged with V.splendidus challenged sea cucumber fed with the basal diet was significantly higher than those fed with dietary β-glucan or glycyrrhizin.There was no significant difference in cumulative morbidity between the dietary β-glucan and glycyrrhizin treatments over time.展开更多

Methods: Corresponding treatment was given to rats in model group, Ramipril group, normal control group, low dosage SCF group and high dosage SCF group respectively for 32 days. The expressions of PPARγ and adiponectin Messenger RNA (mRNA) were detected by real-time reverse transcriptase poly-merase chain reaction. Results: The expressions of PPARγ and adiponectin mRNA increased significantly in both low and high dosage SCF groups as compared with the model group, and a positive linear correlation was found between the expressions of PPARγ and adiponectin mRNA. Conclusions: SCF can prevent macrovascular lesion in early stage of diabetes, which is possibly related with up-regulating expressions of PPARγ and activating PPARγ.展开更多

A plate microfiltration module was found suitable for such purpose. By feeding whole medium in MMF, the productivity of GOD was much higher than that by feeding glucose alone. With increasing dilution rate the enzyme productivity increased and average enzyme activity decreased. The enzyme productivity of MMF under D = 0.12h-1 and 0.20h-1 were 3871 and 3945U·h-1 respectively, which was about 3 times as that of batch fermentation (BF) and the average enzyme activity was still as high as STU·mL-1 under D = 0.12h-1. The relative efficiency of MMF applied to low yield strain was higher than that applied to high yield strain.展开更多

This study assessed the changes in enzyme activity with slope position and eros...Determining how soil erosion affects enzyme activity may enhance our understanding of soil degradation on eroded agricultural landscapes. This study assessed the changes in enzyme activity with slope position and erosion type by selecting water and tillage erosion-dominated slopes and performing analyses using the 1376s technique. The 137Cs data revealed that soil loss occurred in the upper section of the two eroded slope types, while soil accumulation occurred in the lower section. The invertase activity increased downslope and exhibited a pattern similar to the 137Cs data. The spatial patterns of urease and alkaline phosphatase activities were similar to the 137Cs inventories on the water and tillage erosion-dominated slopes, respectively. On both the eroded slope types, the invertase activity and soil organic carbon content were correlated, but no correlation was observed between the alkaline phosphatase activity and total phosphorus content. Nevertheless, the urease activity was correlated with the total nitrogen content only on the water erosion-dominated slopes. The enzyme activity-to-microbial biomass carbon ratios indicated high activities of invertase and urease but low activity of phosphatase on the water erosion-dominated slopes compared with the tillage erosion-dominated slopes. Both the invertase activity and the invertase activity-to-microbial biomass carbon ratio varied with the slope position. Changes in the urease activity-to-microbial biomass carbon ratio were significantly affected by the erosion type. These suggested that the dynamics of the invertase activity were linked to soil redistribution on the two eroded slope types, whereas the dynamics of the urease and alkaline phosphatase activities were associated with soil redistribution only on the water or tillage erosion-dominated slopes, respectively. The erosion type had an obvious effect on the activities of invertase, urease and alkaline phosphatase. Soil redistribution might influence the involvement of urease in the N cycle and alkaline phosphatase in the P cycle. Thus, enzyme activity-to-microbial biomass ratios may be used to better evaluate microbiological activity in eroded soils.展开更多
A pot experiment was carried out to study the effects of three arbuscular mycorrhizal fungi (AMF), including Glomus intraradices, Glomus constrictum and Glomus mosseae, on the growth, root colonization and Cd accumu...A pot experiment was carried out to study the effects of three arbuscular mycorrhizal fungi (AMF), including Glomus intraradices, Glomus constrictum and Glomus mosseae, on the growth, root colonization and Cd accumulation of marigold (Tagetes erecta L.) at Cd addition levels of 0, 5 and 50 mg kg-1 in soil. The physiological characteristics, such as chlorophyll content, soluble sugar content, soluble protein content and antioxidant enzyme activity, of Tagetes erecta L. were also investigated. The symbiotic relationship between the marigold plant and arbuscular mycorrhizal fungi was well established under Cd stress. The symbiotic relationship was reflected by the better physiobiochemical parameters of the marigold plants inoculated with the three AMF isolates where the colonization rates in the roots were between 34.3% and 88.8%. Compared with the non-inoculated marigold plants, the shoot and root biomass of the inoculated marigold plants increased by 15.2%- 47.5% and 47.8%-130.1%, respectively, and the Cd concentration and accumulation decreased. The chlorophyll and soluble sugar contents in the mycorrhizal marigold plants increased with Cd addition, indicating that AMF inoculation helped the marigold plants to grow by resisting Cd stress. The antioxidant enzymes reacted differently with the three AMF under Cd stress. For plants inoculated with G. constrictum and G. mosseae, the activities of superoxide dismutase (SOD) and catalase (CAT) increased with increasing Cd addition, but peroxidase (POD) activity decreased with increasing Cd addition. For plants inoculated with G. intraradices, three of the antioxidant enzyme activities were significantly decreased at high levels of Cd addition. Overall, the activities of the three antioxidant enzymes in the plants inoculated with AMF were higher than those of the plants without AMF inoculation under Cd stress. Our results support the view that antioxidant enzymes have a great influence on the biomass of plants, and AMF can improve the capability of reactive oxygen species (ROS) scavenging and reduce Cd concentration in plants to alleviate Tagetes erecta L. from Cd stress.展开更多
