Advanced glycation end-products (AGEs) are products of non-enzymatic glycation of proteins, lipids or nucleic acids and other macromolecules. To be spe- cific, Nε-(carboxymethyl)-Iysine (CML) is one of the most important components of AGEs, which is wildly distributed in the body and can be formed in vivo or in food processing and heating processes. Previous studies have shown that CML is a ma- jor immunological epitope in AGEs and plays an important role in diabetes and its complications, as well as in the development and progression of aging. This review summarized recent advances in major source, toxicological hazard and control mea- sures of CML. 展开更多

目的:探讨培养的主动脉平滑肌细胞(ASMC)上是否表达糖基化终产物(AGEP)高亲性受体和地尔硫对AGEP升高胞浆游离钙的抑制.方法:用放射配基结合方法研究ASMC与AGEP的相互作用;用钙离子荧光指示剂Fura2... 目的:探讨培养的主动脉平滑肌细胞(ASMC)上是否表达糖基化终产物(AGEP)高亲性受体和地尔硫对AGEP升高胞浆游离钙的抑制.方法:用放射配基结合方法研究ASMC与AGEP的相互作用;用钙离子荧光指示剂Fura2AM测定ASMC胞浆游离钙.结果:ASMC上有AGEP高亲和性受体表达,其解离常数为653±15nmol·L-1,最大结合容量157±004nmol/g细胞蛋白;通过该受体介导,平滑肌细胞可内化、代谢AGEP.AGEP使胞浆游离钙呈浓度依赖性升高;地尔硫呈时间、浓度依赖性抑制此效应.结论:大鼠ASMC上有AGEP高亲和性受体表达;地尔硫可抑制AGEP介导的胞浆游离钙升高. 展开更多

Potential for Pakistani traditional medicinal plants to combat diabetes

OBJECTIVE: To investigate the potential of medicinal plants used in traditional healing in Pakistan to inhibit the glycation process. This may be useful in combatting diabetes mellitus and its complications.METHODS: The in vitro antiglycation activity of 10 medicinal plants was examined by testing the ability of the extracts to inhibit the methyl-glyoxal-mediated development of fluorescence of bovine serumalbumin.RESULTS: Of the tested plants, Persicaria barbata,Geranium collinum and Berberis lyciumshowed significant inhibition of the formation of advanced glycation end products. The inhibitory capacity of these plants was 68.89%, 62.06% and 54.23%, respectively, compared with the positive control(rutin; 86%). All other plants inhibited AGE formation non-significantly.CONCLUSION:These findings will be helpful for further research into the use of traditional herbal medicines with antiglycation properties in the treatment of diabetes. 展开更多
