Lonicera hypoglauca is a traditional Chinese medicinal plant.In this study,the tender young leaves of L.hypoglauca were used for the first time as the explants to establish a rapid in vitro propagation and regeneratio...Lonicera hypoglauca is a traditional Chinese medicinal plant.In this study,the tender young leaves of L.hypoglauca were used for the first time as the explants to establish a rapid in vitro propagation and regeneration system.The results revealed that the optimal time for disinfection of the explants was 8 min and the optimal medium for callus induction was MS+2,4-D 4.0 mg·L^(-1)+sucrose 30 g·L^(-1),with an average callus induction rate of 86.67%.The optimal medium to induce differentiation of callus to bud was MS+6-BA 1.0 mg·L^(-1)+NAA 0.10 mg·L^(-1)+sucrose 30 g·L^(-1),with an average germination rate of 83.33%.The optimal medium to induce multiplication was MS+6-BA 1.5 mg·L^(-1)+NAA 0.05 mg·L^(-1)+sucrose 30 g·L^(-1),with a multiplication coefficient of 5.42.The optimal medium for root induction was 1/2 MS+NAA 0.15 mg·L^(-1)+activated carbon 0.3 g·L^(-1)+sucrose 15 g·L^(-1),with an average rooting rate of 91.11%.The survival rate of tissue-cultured seedlings in nutrient soil cultivation medium was as high as 100%.The total flavonoid content and chlorogenic acid content in the explant,callus tissue and regenerated plant were 1.83%,2.27%,1.33%and 2.77%,1.83%,1.74%respectively.This study provides novel insights into the rapid propagation and mass production of L.hypoglauca seedlings at an industrial scale and that it exhibits important application value and future prospects.展开更多

The rooting efficiency index (REI) showed a positive correlation with the rooting rate. The female flowers from previously bagged buds were treated respectively with paper bags, net bags and hand pollination by comparing with natural pollination. The results showed that female flowers treated with paper bags, net bags, hand pollination, and natural pollination all set fruits and seeds, but there was great difference in their fruit set, seed set, pre-emergent reproductive success (PERS) and seed germination rate for two populations. In both populations, PERS of hand pollination and natural pollination were higher than those of paper bagged and net bagged treatments. Among them, PERS of hand pollination was the highest, and PERS of net bagged treatment was the lowest. Both sexual reproduction and apomixis occurred simultaneously in W septentrionalis and seeds from both sexual and apomixis could germinate into seedlings. Therefore, the breeding system of W septentrionalis belonged to facultative apomixis. The apomixis in Magnoliaceae was reported for the first time.展开更多

[Method] Three commercial PTGMS lines, Annong 810S, P88S and C815S, were used in this study. In the first year, two sterile line populations with same genetic background were multiplied by dividing seedling; meanwhile, se- lection and multiplication of the core individuals were conducted; within the year, the core seeds harvested were multiplied under the isolated condition in Hainan, and at the same time, superior individuals were selected and used as the seeds for two populations with same genetic background in the next year. In the second year, seeds harvested in Hainan were planted in Changsha, while the core individuals were selected and multiplied; the core seeds harvested were continued to multiply in Hainan. The above procedures were repeated in the following years. [Result] The test results showed that in two years the proportion of core individuals in the popu- lation of Annong 810S, P88S and C815S increased from 40.3%, 0.7% and 0.3% in the first year to 71.67%, 66.00% and 68.67% in the second year respectively. [Conclusion] This suggested that this method of purification and multiplication of the PTGMS lines for consecutive years can effectively control the drift of the critical sterility inducing temperature, and at the same time, core seeds with high purity and controlled inducing temperature can be harvested.展开更多

[Methed] Through hybridization, a new breeding material Mian 7MB-1 in three-line genic temporary maintainer line propagated by tissue culture was used to improve the sterile plant rate of rapeseed in dual-purpose recessive GMS line, such as Mian 7AB type, S45AB type, and etc. And then the variety comparative test was performed. [Result] In order to avoid some fertility restoration phenomena occurring during the process of self-reproduction, Mian 7AB was propagated in bulk with somatic tissue culture of temporary maintainer line plant stem. The propagated temporary maintainer line seedlings were applied to the breeding and seed production of net room male sterile line parent, promoting the sterile plant rate of the male sterile line parent to 91.7% -93.5%. The male sterile line parents per hectare were enough for the seed production of hybrid F1 in 7 500 -15 000 hm^2. [ Conclusion ] Compared with the original dual-purpose GMS line, the seed production ultilizing male sterile line with high sterile plant rate greatly reduced the labor, significantly improved the seed yield, ensuring the seed quality and forming a perfect breeding and seed production system.展开更多

We summarized the incidence of macroalgae blooms throughout the world and their impact on seagrass beds by direct and indirect ways. The competition for living space and using resources is the most direct effect on seagrass beds when macroalgae are blooming in an aquatic ecosystem. The consequence of macroalgae blooms(e.g., light reduction, hypoxia, and decomposition) can produce significant indirect effects on seagrass beds. Light reduction by the macroalgae can decrease the growth and recruitment of seagrasses, and decomposition of macroalgae mats can increase the anoxic and eutrophic conditions, which can further constrict the seagrass growth. Meanwhile, the presence of seagrass shoots can provide substrate for the macroalgae blooms. Controlling nutrient sources from the land to coastal waters is a general efficient way for coastal management. Researching into the synergistical effect of climate change and anthropognic nutrient loads on the interaction between searsasses and macroalgae can provide valuable information to decrease the negative effects of macroalgae blooms on seagrasses in eutrophic areas.展开更多
Nursery plant propagation by grafting has been widely used in modern viticulture to minimize the damage caused by biotic and abiotic stresses.In grapevine(Vitis spp.),an effective way to control disease damage is to p...Nursery plant propagation by grafting has been widely used in modern viticulture to minimize the damage caused by biotic and abiotic stresses.In grapevine(Vitis spp.),an effective way to control disease damage is to provide producers and growers with pathogen-free stock plants.In this study,five grapevine rootstock varieties,‘SO4’,‘101-14’,‘5BB’,‘110R’and‘1103P’,were selected as explants to establish an in vitro culture protocol,and three species of grapevine viruses were tested by real-time RT-PCR.The results showed that MS medium supplemented with 0.2 mg/L IBA,1.0 mg/L 6-BA,0.5 mg/L KT,4.0 mg/L adenine for culture initiation,and WPM supplemented with 0.2 mg/L IBA for subculture were suitable for all five rootstock varieties,with multiplication coefficients ranging from 1.6 to 4.4.Virus testing showed that single RT-PCR was more effective for detecting the three viruses compared to double or triple RT-PCR.Only plantlets free from the aforementioned viruses were retained for subculture.Plantlets were hardened at room temperature under natural lighting in Hoagland solution for 2 weeks and transplanted to pots filled with mixed media in a greenhouse.This protocol is applicable for rapid propagation of the five grapevine rootstock varieties and can be used for commercial production of virus-free grapevine stocks.展开更多

Isozyme banding patterns indicated these hybrids were diploid. Loci that were observed as being monomorphic in inbred populations of C. farreri were also found to be monomorphic in filial progeny; loci that observed to be polymorphic in parental type gen- erations were also polymorphic in hybrid generations. Differences existed among allelic frequency of the four types of cross. Within the reciprocal hybrids the expression of malic enzyme (ME) isozyme was sufficient to distinguishing individual hybrids because of the band, Rf=0.38. However, there were no noticeable variations among all the samples to differentiate one from another. Inbreeding was likely to be the main problem in aqua- culture. The introduction of new broodstock can improve the genetic diversity. Hybrid vigor has manifested to a certain extent in the present study.展开更多

Documenting the sex and sexual reproduction of the microalgae is very difficult, as most of the results are based on the microscopic observation that can be heavily influenced by genetic, physiological and environment...Documenting the sex and sexual reproduction of the microalgae is very difficult, as most of the results are based on the microscopic observation that can be heavily influenced by genetic, physiological and environmental conditions. Understanding the reproduction strategy of some microalgae is required to breed them in large scale culture industry. Instead of direct observation of sex and sexual reproduction under microscope, the whole set or the majority of core meiosis genes may evidence the sex and sexual reproduction in the unicellular algae, as the meiosis is necessary for maintaining the genomic stability and the advantages of genetic recombination. So far, the available genome sequences and bioinformatic tools (in this study, homolog searching and phylogenetic analysis) allow us to propose that at least 20 core meiosis genes (among them 〉6 must be meiosis specific) are enough for an alga to maintain its sexual reproduction. According to this assumption and the genome sequences, it is possible that sexual reproduction was carried out by Micromonas pusilla and Cyanidiosehyzon merolae, while asexual reproduction was adopted by Bigelowiella natans, Guillardia theta, Nannochloropsis gaditana, N. oeeanica, Chlorella variablis, Phaeodactylum tricornutum and Thalassiosira pseu- donana. This understanding will facilitate the breeding trials of some economic microalgae (e.g., N. gaditana, N. oceanica, C. vari- ablis and P. tricornutum). However, the reproduction strategies of these microalgae need to be proved by further biological experiments.展开更多

It is notable that a good choice of planting site is a guar- antee of the technology and to establish the method cultivating sunflowers with three generations per year is of great significance in terms of reducing breeding process of inbred lines and breeding costs. The results showed,using this method,that proliferation rate of ROC16 improved by 40 times,per flask generated about 800 plantlets; of ROC22 improved by 30 times,per flask generated about 400-600 plantlets. Propagation can be done by cuttings, l...The genus Echeveria has about 143 species that are unique to the Americas, 117 of which are presented in the Mexican Republic, mainly in the states of Hidalgo, Puebla and Oaxaca. Propagation can be done by cuttings, leaf cuttings or seeds, but these methods are insufficient to avoid the predation of many species. NOM-059-ECOL-2001 shows Echeveria elegans as species reported endangered. The objectives in this study were achieved in vitro propagation from axillary buds, as an alternative to multiplication and preservation species. We evaluated different concentrations and combinations of two growth regulators, 6-BAP (2.22, 4.44, 6.66 and 8.88 μM) and u-NAA (1.35 and 2.70 μM) on shoot formation from axillary buds and two culture media with different concentrations of MS salts, to achieve root plants grown in vitro. It was found by combining 6.66 μM of 6-BAP and 1.35μM of α-NAA favored the formation and development of new shoots. In the rooting stage, the best results are achieved with the treatment containing 50% of MS salts. Then the plants were transplanted into greenhouses and achieved a successful acclimatization. During this last phase of development, there were no phenotypic changes or presence of somaclonal variants.展开更多

The male sterile line parents per hectare were enough for the seed production of hybrid F1 in 7 500 -15 000 hm^2. [ Conclusion ] Compared with the original dual-purpose GMS line, the seed production ultilizing male sterile line with high sterile plant rate greatly reduced the labor, significantly improved the seed yield, ensuring the seed quality and forming a perfect breeding and seed production system.
文摘Rong 18A is an Indica three-line sterile line bred with M5B×D Xing B, and has the characteristics of stable sterility, high outcrossing rate, good combining ability and good quality. With Rong 18A as an experimental material and Ⅱ-32A, Jin 23A and Gang46 A as the CK materials, a seed setting rate estimation method by daily pollination was used for measuring its stigma vigor, to comparatively analyze the decreasing rates and lasting time of stigma vigor of various materials under natural condition. According to experimental results and combining with field breeding practice, the application of stigma vigor of sterile lines on hybrid rice seed production was discussed.
文摘[Objective] The aim was to improve part of auto-control system of aquatic seedling breeding and to lay foundation for highly-efficient and stable use of the sys- tem. [Method] The executive and drive modules of the auto-control system were im- proved, after which real-time monitor, data display and alarm modules were tested on time. [Result] In the test, for the improved system, stability and sensitivity were enhanced; different components matched better; transmitting electric resistanc6 was reduced. [Conclusion] The research provides references for auto-control of water temperature and dissolved oxygen, and faults clearing in aquatic seedling breeding.

文摘The genetic parameters of main reproductive traits for the new Yorkshire line with high prolificacy were evaluated by multiple animal model DFREML The re- suits showed that the heritability evaluates of the total number of born (TNB), alive number of born (ANB), litter weigh at birth (LWB) and litter weight at age of 21 days were 0.10, 0.15, 0.09 and 0.14, respectively. The genetic correlations between these traits, for example, the correlations between TNB/ANB, NB/LWB, TNB/LW21, ANB/LWB, ANB/LW21 and LWB/LW21 were 0.87, 0.74, 0.43, 0.69, 0.59 and 0.51, respectively.

文摘Seagrass decline caused by the macroalgae blooms is becoming a common phenomenon throughout temperate and tropical regions. We summarized the incidence of macroalgae blooms throughout the world and their impact on seagrass beds by direct and indirect ways. The competition for living space and using resources is the most direct effect on seagrass beds when macroalgae are blooming in an aquatic ecosystem. The consequence of macroalgae blooms(e.g., light reduction, hypoxia, and decomposition) can produce significant indirect effects on seagrass beds. Light reduction by the macroalgae can decrease the growth and recruitment of seagrasses, and decomposition of macroalgae mats can increase the anoxic and eutrophic conditions, which can further constrict the seagrass growth. Meanwhile, the presence of seagrass shoots can provide substrate for the macroalgae blooms. Controlling nutrient sources from the land to coastal waters is a general efficient way for coastal management. Researching into the synergistical effect of climate change and anthropognic nutrient loads on the interaction between searsasses and macroalgae can provide valuable information to decrease the negative effects of macroalgae blooms on seagrasses in eutrophic areas.
文摘Nursery plant propagation by grafting has been widely used in modern viticulture to minimize the damage caused by biotic and abiotic stresses.In grapevine(Vitis spp.),an effective way to control disease damage is to provide producers and growers with pathogen-free stock plants.In this study,five grapevine rootstock varieties,‘SO4’,‘101-14’,‘5BB’,‘110R’and‘1103P’,were selected as explants to establish an in vitro culture protocol,and three species of grapevine viruses were tested by real-time RT-PCR.The results showed that MS medium supplemented with 0.2 mg/L IBA,1.0 mg/L 6-BA,0.5 mg/L KT,4.0 mg/L adenine for culture initiation,and WPM supplemented with 0.2 mg/L IBA for subculture were suitable for all five rootstock varieties,with multiplication coefficients ranging from 1.6 to 4.4.Virus testing showed that single RT-PCR was more effective for detecting the three viruses compared to double or triple RT-PCR.Only plantlets free from the aforementioned viruses were retained for subculture.Plantlets were hardened at room temperature under natural lighting in Hoagland solution for 2 weeks and transplanted to pots filled with mixed media in a greenhouse.This protocol is applicable for rapid propagation of the five grapevine rootstock varieties and can be used for commercial production of virus-free grapevine stocks.

文摘Chinese and Japanese population of Chlamys farreri and their reciprocal hybrids were surveyed in isozyme variability at 13 loci by polyacrytamide gel electrophoresis (PAGE). Isozyme banding patterns indicated these hybrids were diploid. Loci that were observed as being monomorphic in inbred populations of C. farreri were also found to be monomorphic in filial progeny; loci that observed to be polymorphic in parental type gen- erations were also polymorphic in hybrid generations. Differences existed among allelic frequency of the four types of cross. Within the reciprocal hybrids the expression of malic enzyme (ME) isozyme was sufficient to distinguishing individual hybrids because of the band, Rf=0.38. However, there were no noticeable variations among all the samples to differentiate one from another. Inbreeding was likely to be the main problem in aqua- culture. The introduction of new broodstock can improve the genetic diversity. Hybrid vigor has manifested to a certain extent in the present study.

The reproduction of living beings is self-generation while the reproduction of the solar system and other galaxies is generation by augmentation. Man-made objects, society and galaxies are formed with automatic organization of thoughts, thus called generation by transformation. The solar system and the earth have reproduction function. Living beings began with the combined action of the earth, the solar system and the universe. Life forms of the universe include dominant life and recessive life. Different forms of life transform continuously.

文摘Documenting the sex and sexual reproduction of the microalgae is very difficult, as most of the results are based on the microscopic observation that can be heavily influenced by genetic, physiological and environmental conditions. Understanding the reproduction strategy of some microalgae is required to breed them in large scale culture industry. Instead of direct observation of sex and sexual reproduction under microscope, the whole set or the majority of core meiosis genes may evidence the sex and sexual reproduction in the unicellular algae, as the meiosis is necessary for maintaining the genomic stability and the advantages of genetic recombination. So far, the available genome sequences and bioinformatic tools (in this study, homolog searching and phylogenetic analysis) allow us to propose that at least 20 core meiosis genes (among them 〉6 must be meiosis specific) are enough for an alga to maintain its sexual reproduction. According to this assumption and the genome sequences, it is possible that sexual reproduction was carried out by Micromonas pusilla and Cyanidiosehyzon merolae, while asexual reproduction was adopted by Bigelowiella natans, Guillardia theta, Nannochloropsis gaditana, N. oeeanica, Chlorella variablis, Phaeodactylum tricornutum and Thalassiosira pseu- donana. This understanding will facilitate the breeding trials of some economic microalgae (e.g., N. gaditana, N. oceanica, C. vari- ablis and P. tricornutum). However, the reproduction strategies of these microalgae need to be proved by further biological experiments.

文摘Large high-dimensional data have posed great challenges to existing algorithms for frequent itemsets mining.To solve the problem,a hybrid method,consisting of a novel row enumeration algorithm and a column enumeration algorithm,is proposed.The intention of the hybrid method is to decompose the mining task into two subtasks and then choose appropriate algorithms to solve them respectively.The novel algorithm,i.e.,Inter-transaction is based on the characteristic that there are few common items between or among long transactions.In addition,an optimization technique is adopted to improve the performance of the intersection of bit-vectors.Experiments on synthetic data show that our method achieves high performance in large high-dimensional data.

文摘B ased on the data of phytoplankton and environmental factors in the Bohai Bay, the dependence between the concentration of phytoplankton and environmental factors is analysed by linear correlation coefficient, rank correlation coefficient and Hoeffding test of independence .The result shows that wind-speed, air-pressure, surface temperature, field pH, salinity, DO, silicate and NO3- have a great impact on the concentration of phytoplankton.

文摘An experiment was performed in a sunflower field based on sunflower characters and relevant researches available to explore multiple-generation cultiva- tions in Hainan in order to establish a cultivation method of sunflower inbred line with three generations per year and reduce breeding process of inbred lines. The results showed that the cultivation site is chosen as per relevant standards that ef- fective accumulative temperature (≥5 ℃) is equal to or higher than 1 900 ~C within 110 d from sowing date. It is notable that a good choice of planting site is a guar- antee of the technology and to establish the method cultivating sunflowers with three generations per year is of great significance in terms of reducing breeding process of inbred lines and breeding costs. In addition to that, it provides refer- ences for rapid cultivation of multi-generation sunflowers and improving breeding effi- ciency.
文摘This paper gave the standards to evaluate Hequ Tibetan Mastiff on body type, appearance, body measures and temperament. A nucleus group of breeding consisted of high-grade individual according this standards, and it was for enhancing the proceeding of selecting and breeding of Hequ Tibetan Mastiff. By this way, after working on three generations it had been gained that effective population size of the nucleus group of breeding was 96, increasing numbers of inbreeding coefficient was 0.005, male and female body height were 72.1 cm and 70.2 cm, their cannon circumference were 16.3 cm and 15.2 cm, respectively.

文摘By employing temporary immersion bioreactor system(TIBs),we studied virus-free culture of seedlings from sugarcane varieties ROC16 and ROC22,from medium recipe,inoculation amount,sucrose concentration,and variety difference. The results showed,using this method,that proliferation rate of ROC16 improved by 40 times,per flask generated about 800 plantlets; of ROC22 improved by 30 times,per flask generated about 400-600 plantlets. The results provided basis for using TIBs in rapid propagation of plantlets via tissue culture.
文摘The genus Echeveria has about 143 species that are unique to the Americas, 117 of which are presented in the Mexican Republic, mainly in the states of Hidalgo, Puebla and Oaxaca. Propagation can be done by cuttings, leaf cuttings or seeds, but these methods are insufficient to avoid the predation of many species. NOM-059-ECOL-2001 shows Echeveria elegans as species reported endangered. The objectives in this study were achieved in vitro propagation from axillary buds, as an alternative to multiplication and preservation species. We evaluated different concentrations and combinations of two growth regulators, 6-BAP (2.22, 4.44, 6.66 and 8.88 μM) and u-NAA (1.35 and 2.70 μM) on shoot formation from axillary buds and two culture media with different concentrations of MS salts, to achieve root plants grown in vitro. It was found by combining 6.66 μM of 6-BAP and 1.35μM of α-NAA favored the formation and development of new shoots. In the rooting stage, the best results are achieved with the treatment containing 50% of MS salts. Then the plants were transplanted into greenhouses and achieved a successful acclimatization. During this last phase of development, there were no phenotypic changes or presence of somaclonal variants.
