目的通过体内及体外途径研究系统性红斑狼疮(SLE)患者在应用地塞米松前后外周血CD4+ CD25- T淋巴细胞凋亡率的变化,探讨影响SLE患者经激素治疗后CD4+CD25+调节性T淋巴细胞(Treg)比率上调的机制。方法应用AnnexinV检测试剂盒及流式细胞... 目的通过体内及体外途径研究系统性红斑狼疮(SLE)患者在应用地塞米松前后外周血CD4+ CD25- T淋巴细胞凋亡率的变化,探讨影响SLE患者经激素治疗后CD4+CD25+调节性T淋巴细胞(Treg)比率上调的机制。方法应用AnnexinV检测试剂盒及流式细胞仪检测未经激素治疗的10例SLE患者经地塞米松治疗0、1、3、7d后外周血单个核淋巴细胞(PBMC)中CD4+ CD25- T淋巴细胞的早期凋亡率;结合细胞培养技术检测未经激素治疗时SLE患者PBMC在体外经地塞米松刺激培养1、3、7d后CD4+ CD25- T淋巴细胞的早期凋亡率。结果地塞米松治疗后与未治疗之前相比,SLE患者PBMC中CD4+ CD25- T淋巴细胞早期凋亡率升高(P<0.05),治疗0、1、3、7d的凋亡率分别是0.21±0.03、1.39±0.22、1.57±0.11、1.83±0.31;而CD4+ CD25+ Treg细胞早期凋亡率无明显变化(P>0.05),治疗0、1、3、7d的凋亡率分别是0.44±0.06、0.47±0.04、0.43±0.07和0.35±0.07;未治疗的SLE患者PBMC经过体外地塞米松刺激培养后CD4+ CD25- T淋巴细胞早期凋亡率升高(P<0.05)。体外培养1、3、7d的凋亡率分别是2.02±0.29、2.50±0.59和3.72±0.36;未治疗的SLE患者PBMC经过体外地塞米松刺激培养后,CD4+ CD25- T淋巴细胞早期凋亡率随激素剂量的增加而上升(P<0.05)。体外培养7d后,DMX剂量为0、5、10、25μg时凋亡率分别为0.60±0.26、1.51±0.69、2.20±0.80和3.65±0.72;SLE患者CD4+ CD25- T淋巴细胞早期凋亡率与抗ds-DNA抗体、抗Sm抗体、IgG、IgA、IgM、抗SSA抗体、抗SSB抗体、C3、C4等均无相关性(P>0.05),与SLEDAI评分呈负相关(P<0.05)。结论 SLE患者经过地塞米松治疗后外周血CD4+ CD25- T淋巴细胞的凋亡增加并与剂量相关,引起外周血CD4+ CD25- T淋巴细胞数量的减少,可能是导致CD4+ CD25+ Treg细胞比例相对增加的原因。 展开更多

本文采用实时荧光定量PCR技术对荧光假单胞菌PEF-5#18诱导番茄系统性抗性(ISR)的作用机理进行了研究,包括了诱导进程中番茄根系病程蛋白基因PRs族、GR等重要防卫基因以及乙烯、水杨酸、茉莉酸等诱导信号调控路径的关键基因的表达情况。... 本文采用实时荧光定量PCR技术对荧光假单胞菌PEF-5#18诱导番茄系统性抗性(ISR)的作用机理进行了研究,包括了诱导进程中番茄根系病程蛋白基因PRs族、GR等重要防卫基因以及乙烯、水杨酸、茉莉酸等诱导信号调控路径的关键基因的表达情况。结果表明,尖孢镰刀菌Forl和荧光假单胞菌PEF-5#18对番茄植株分别具有SAR和ISR诱导抗性能力,而在"Forl-番茄-PEF-5#18"互作模式下,番茄植株受诱导所产生的抗性则受到SAR和ISR共同作用的影响;与对照相比,Forl或荧光假单胞菌PEF-5#18均能诱导番茄病情蛋白基因PRs (PR1、PR4、PR5、PR6)与GR、POX、PAL等相关防卫基因的表达进行上调,而相关受诱导基因的表达程度与动态变化则与所诱导的SAR和ISR抗性反应类型以及诱导时间有关;此外,对于调控路径的分析结果显示出尖孢镰刀菌Forl对番茄SAR诱导抗性主要依赖于水杨酸路径进行调控,而荧光假单胞菌PEF-5#18对番茄ISR诱导抗性则主要依赖于茉莉酸和乙烯路径来调控。 展开更多

Effect of Chaetomium globosum ND35 on Plant Growth and Preliminary Study of Its Biocontrol Efficacy

[Objective] The aim was to study effect of Chaetomium globosum ND35 on plant growth and preliminary study of its biocontrol efficacy, and provide basis for popularization and application of this strain.[ Method] With endophytic fungus C. globosum ND35 as a tested strain, effect of C. globosumND35 on plant growth and its biocontrol on five plant diseases were investigated in the greenhouse and field,[Result]The results showed that ND35 promoted growth of lateral root and diameter of breast height of poplar. ND35 can induce poplar to resist Poplar Valsa Canker caused by Valsa sordida and Poplar Rust caused by Melampsora puplicola. ND35 was also able to induce tomato and bean to resist Botrytis cinera. Biocontrol of Bean Stem Rot Rhizoctonia by ND35 was effective as well. [Conclusion] Induced systemic resistance by endophytic C. globosum ND35 plays an important role in biocontrol of plant diseases. 展开更多

In this paper,the fault detection filter(FDF) design problem for networked control systems(NCSs) with both network-induced delay and data dropout is studied.Based on a new NCSs model proposed recently,an observer-based filter is introduced to be the residual generator and formulated as an H∞-optimization problem for systems with two successive delay components.By applying Lyapunov-Krasovskii approach,a new sufficient condition on stability and H∞ performance is derived for systems with two successive delay components in the state.A solution of the optimization problem is then presented in terms of linear matrix inequality(LMI) formulation,dependently of time delay.In order to detect the fault,the residual evaluation problem is also considered.An illustrative design example is employed to demonstrate the validity of the proposed approach. 展开更多

AIM: To investigate the changes of platelet endothelial cell adhesion molecule-1 (PECAM-1) expression on polymorphonuclear leukocytes (PMNs) in peripheral circulation and pancreatic microcirculation in cerulein-induced acute edematous pancreatitis (AEP).METHODS: Fifty Wistar rats were randomly divided into control group (n=10) and AEP group (n=40). A model of AEP was established by subcutaneous injection of cerulein 5.5 and 7.5 μg/kg at 0 and 1 h after the beginning of experiment respectively. PECAM-1 expression on PMNs from splenic vein and inferior vena cava was determined by RT-PCR at mRNA level and determined by flow cytometry at protein level.RESULTS: In experimental rats, an increased PECAM-1mRNA expression was seen from 4 to 8 h of AEP in peripheral circulation (0.77±0.25%, 0.76±0.28%, 0.89±0.30%,1.00±0.21% ), while in pancreatic microcirculation,expression decreased from 2 h and reached the lowest level at 6 h of AEP (0.78±0.29%, 0.75±0.26%, 0.62±0.28%,0.66±0.20%). There were significant differences at 8-h time point of AEP between peripheral circulation and pancreatic microcirculation (1.00±0.21% vs0.66±0.20%, P<0.05).Meanwhile,the difference at protein level was also found.CONCLUSION: A reverse expression of PECAM-1 on PMNs was found between peripheral circulation and pancreatic microcirculation, suggesting that inhibition of PECAM-1expression may improve the pathological change of AEP. 展开更多

Effects of arbuscular mycorrhizal fungi on carbon assimilation and ecological stoichiometry of maize under combined abiotic stresses

Arbuscular mycorrhizal fungi(AMF)enhance plant tolerance to abiotic stresses like salinity and improve crop yield.However,their effects are variable,and the underlying cause of such variation remains largely unknown.This study aimed to assess how drought modifed the effect of AMF on plant resistance to high calcium-saline stress.A pot experiment was performed to examine how AMF inoculation affects the growth,photosynthetic activity,nutrient uptake and carbon(C),nitrogen(N)and phosphorus(P)stoichiometric ratio(C:N:P)of maize under high calcium stress and contrasting water conditions.The results showed that high calcium stress signifcantly reduced mycorrhizal colonization,biomass accumulation,C assimilation rate and C:N stoichiometric ratio in plant tissues.Besides,the adverse effects of calcium stress on photosynthesis were exacerbated under drought.AMF inoculation profoundly alleviated such reductions under drought and saline stress.However,it barely affected maize performance when subjected to calcium stress under well-watered conditions.Moreover,watering changed AMF impact on nutrient allocation in plant tissues.Under well-watered conditions,AMF stimulated P accumulation in roots and plant growth,but did not induce leaf P accumulation proportional to C and N,resulting in increased leaf C:P and N:P ratios under high calcium stress.In contrast,AMF decreased N content and the N:P ratio in leaves under drought.Overall,AMF inoculation improved maize resistance to calcium-salt stress through enhanced photosynthesis and modulation of nutrient stoichiometry,particularly under water defcit conditions.These results highlighted the regulatory role of AMF in carbon assimilation and nutrient homeostasis under compound stresses,and provide signifcant guidance on the improvement of crop yield in saline and arid regions. 展开更多

The global bifurcation of strongly nonlinear oscillator induced by parametric and external excitation is researched. It is known that the parametric and external excitation may induce additional saddle states, and result in chaos in the phase space, which cannot be detected by applying the Melnikov method directly. A feasible solution for this problem is the combination of the averaged equations and Melnikov method. Therefore, we consider the averaged equations of the system subject to Duffing-Van der Pol strong nonlinearity by introducing the undetermined fundamental frequency. Then the bifurcation values of homoclinic structure formation are detected through the combined application of the new averaged equations with Melnikov integration. Finally, the explicit application shows the analytical conditions coincide with the results of numerical simulation even disturbing parameter is of arbitrary magnitude. 展开更多
