[Method] The template cDNA which was reversely transcribed from total RNA of sheep liver tissue, was subjected to polymerase chain reaction (PCR) using specific primers of ActRIIB. The ful-length cDNA of ovine ActRIIB was obtained by pMD18-T cloning and sequencing for bioinformatics analysis. Ovine ActRIIB encoding sequence was subcloned into prokaryotic expression vector pET41a with restriction sites BamHl/Notl, and then transformed into BL21 (DE3). The induced products by lPTG were analyzed with SDS-PAGE and Western Blot. [Result] The amplified ful-length cDNA of ovine Ac-tRllB gene was 1 564 bp in length (Genbank accession number: JX422071.1) with an open reading frame of 1 539 bp, encoding 512 amino acides. Ovine ActRllB shared the highest homology (99.6%) with bovine ActRllB. ActRllB had highly ho-mologous C-terminal domains and belonged to the TGFβ family. After prokaryotic expression, an approximately 92 kD His-tagged ActRllB recombinant protein was obtained, which was consistent with the excepted result. [Conclusion] Cloning and successful expression of ovine ActRIIB laid solid foundation for further investigation of its biological function.展开更多

Co-administration of Chios mastic gum (CMG), which inhibits H. pylor/NAP, with an H. pylori eradication regimen might add clinical benefits against H. pylori-related gastric pathologies, but pos- sibly not CMG as main therapy. Although H. pylori NAP and other H. pylori-related cytotoxins [i.e., vaculating cytotoxin (VacA)] appear to play a major role in gener- ating and maintaining the H. pylori-associated gastric inflammatory response and H. pylor/NAP is a promising vaccine candidate against H. pylori infection (H. pylori-1), concerns regarding its potential drawbacks, particularly neurogenic ones, due to possible cross- mimicry, should be considered. Possible cross-mimicry between H. p, vlor/ NAP and/or bacterial aquaporin (AQP) and neural tissues may be associated with the anti-AQP-4 antibody-related neural damage in multiple sclerosis (MS)/neuromyelitis optica patients. Moreover, the sequence homology found between H. pylori VacA and human Na+/K+-ATPase A subunit suggests that antibodies to VacA involve ion channels in abaxonal Schwann cell plasmalemma resulting in demyelination in some patients. A series of factors have been im- plicated in inducing blood-brain barrier (BBB) disrup- tion, including inflammatory mediators (e.g., cytokines and chemokines induced by H. pylor/-I) and oxidative stress. BBB disruption permits access of AQP4-specific antibodies and T lymphocytes to the central nervous system, thereby playing a major role in multiple sclero- sis pathogenesis. Relative studies show a strong asso- ciation between H. pylori-I and MS. H. pylor/-I induces humoral and cellular immune responses that, owing to the sharing of homologous epitopes (molecular mim- icry), cross-react with components of nerves, thereby contributing and perpetuating neural tissue damage. Finally, H. pylori NAP also plays a possible pathoge- netic role in both gastric and colon oncogenesis.展开更多

Fourteen positive clones were selected for nucleotide sequencing and seven cDNAs encoding various snake venom C-type lectin-like protein precursors, designated as DRS-L1, DRS-L2, DRS-L3, DRS-L4 DRS-L5, DRS-L6 and DRS-L7, were obtained. Amino acid sequences of these proteins were deduced and each contains a carbohydrate recognition domain. Of all the deduced protein sequences, only DRS-L1 seemed to represent a closer sequence similarity to α subunits of other known snake venom C-type lectin-like proteins using the BLAST program. Homology comparison combined with analysis of cysteine position indicate that DRS-L1 and DRS-L2 are probably the light chain LC2 and LC1 of factor X activator from Daboia russellii siamensis venom, respectively. DRS-L3 and DRS-L4 might be the β subunits of higher molecular weight C-type lectin-like proteins while DRS-L5 and DRS-L6 might be β subunits of lower molecular weight C-type lectin-like proteins. DRS-L7 might be the β subunit of a platelet membrane glycoprotein Ib-binding protein similar to echicetin.展开更多

[Method] The cDNA sequence of GnRHR gene was amplified from Apis cerana cerana by using RT-PCR techniques. It was conducted with bioinformatics analysis and the in situ hybridization histochemistry of its expression products was studied. [Result] The sequence analy- sis showed that the full cDNA sequence was 1 050 bp with the open reading frame of 1 050 bp, and it encoded 349 amino acid residues. The deduced amino sequence included 7 transmembrane regions, and the predicted molecular mass and isoelectric point were 40.6 kD and 9.54, respectively. The cluster analysis showed that the GnRHR from ',4. cerana cerana had close relationship to the GnRHR II from other insects. In situ hybridization showed that Bee-GnRHR staining was specifically localized to the brain, intestine, fat body and testis. [Conclusion] The results indicated that the GnRHR provided molecular bond for the reproduction and metabolism for insects, and suggested a functional role for bee-GnRHR signaling in the coupling of reproduction activities and environment conditions.展开更多

Through comparative analysis of PTA gene (pta) and its deduced amino acid sequence with those of other Araceae species, pta was found to encode a precursor lectin with signal peptide and to have extensive homology with those of other Araceae species. PTA was a heterotetrameric mannose-binding lectin with three mannose-binding boxes like lectins from other Araceae and Amaryllidaceae species. Southern blot analysis of the genomic DNA revealed that pta belonged to a low-copy gene family. Northern blot analysis demonstrated that pta constitutively expressed in various plant tissues including root, leaf, stem and inflorescence. The pta cDNA sequence encoding for mature PTA protein was cloned into pET-32a plasmid and the resulting plasmid, pET-32a-PTA containing Trx-PTA fusion protein, was investigated for the expression in E. coli BL21. SDS-PAGE gel analysis showed that the Trx-PTA fusion protein was successfully expressed in E. coli BL21 when induced by IPTG. Artificial diet assay revealed that PTA fusion protein had significant levels of resistance against peach potato aphids when incorporated into artificial diet at 0.1% (w/v). The cloning of the pta gene will enable us to further test its effect in depth on aphids by transferring the gene into crop plants.展开更多

Time series of hydrological data were from 1957 to 2009 for Bao′an station, from 1955 to 2009 for Baoqing station, from 1956 to 2009 for Caizuizi station and from 1978 to 2009 for Hongqiling station. The influences of climate change and human activities on runoff change were investigated, and the causes of hydrological regime change were revealed. The seasonal runoff distribution of the Naoli River was extremely uneven, and the annual change was great. Overall, the annual runoff showed a significant decreasing trend. The annual runoff of Bao′an, Baoqing, and Caizuizi stations in 2009 decreased by 64.1%, 76.3%, and 84.3%, respectively, compared with their beginning data recorded. The wet and dry years of the Naoli River have changed in the study period. The frequency of wet year occurrence decreased and lasted longer, whereas that of dry year occurrence increased. The frequency of dry year occurrence increased from 25.0%-27.8% to 83.9%-87.5%. The years before the 1970s were mostly wet, whereas those after the 1970s were mostly dry. Precipitation reduction and land use changes contributed to the decrease in annual runoff. Rising temperature and water project construction have also contributed important effects on the runoff change of the Naoli River.展开更多

First, the formulas implemented in the programs were presented. These formulas considerably increase the description accuracy of final deformations by taking into uncaved strip along extraction rib (extraction margin). They also improve the deformation description of areas located far from the extraction place. Then, the research results aiming to improving the description of deformation with time were introduced. Finally, the Windows based version of the program for the creation of mining geological opinions were presented in the form accepted by Mining Offices of Poland.展开更多

H. pylor/-I induces humoral and cellular immune responses that, owing to the sharing of homologous epitopes (molecular mim- icry), cross-react with components of nerves, thereby contributing and perpetuating neural tissue damage. Finally, H. pylori NAP also plays a possible pathoge- netic role in both gastric and colon oncogenesis.
文摘Total mRNA was extracted from a venom gland of snake Daboia russellii siamensis following the manufacturer's protocol of the PolyATtract System 1000 kit purchased from Promega Biotech. cDNAs encoding C-type lectins were amplified by RT-PCR and subcloned into a pMD18-T vector. DRS-L7 might be the β subunit of a platelet membrane glycoprotein Ib-binding protein similar to echicetin.

文摘[Objective] This study was to clone the GnRH (gonadotropin-releasing hormone), and to investigate its expression in Apis cerana cerana. [Conclusion] The results indicated that the GnRHR provided molecular bond for the reproduction and metabolism for insects, and suggested a functional role for bee-GnRHR signaling in the coupling of reproduction activities and environment conditions.
文摘The full-length cDNA of Pinellia ternata agglutinin (PTA) was cloned from inflorescences using RACE-PCR. The cloning of the pta gene will enable us to further test its effect in depth on aphids by transferring the gene into crop plants.

文摘Runoff change and trend of the Naoli River Basin were studied through the time series analysis using the data from the hydrological and meteorological stations. Rising temperature and water project construction have also contributed important effects on the runoff change of the Naoli River.

文摘We determined the linear complexity of a family of p2-periodic binary threshold sequences and a family of p2-periodic binary sequences constructed using the Legendre symbol, both of which are derived from Fermat quotients modulo an odd prime p.If 2 is a primitive element modulo p2,the linear complexity equals to p2-p or p2-1,which is very close to the period and it is large enough for cryptographic purpose.
文摘This paper discussed the current of works on computerisation of all problems related to mining subsidence, including the time factor, carried out in the Division of Mining Geodesy of Technical University of Silesia, Poland. Finally, the Windows based version of the program for the creation of mining geological opinions were presented in the form accepted by Mining Offices of Poland.
文摘In this paper, we discuss the counting prob lem of an order n-group of set (A 1,A 2,…,A n) which satisfies ∪ni=1A i={a 1,a 2,…,a m} and one of the following: (1) ∩ni=1A i=Φ; (2) ∩ni=1A i={b 1,b 2,…,b k};(3) ∩ni=1A 1{b 1,b 2,…,b k}; (4) A i≠Φ (i=1,2,…,k). We solve these problems by element analytical meth od.

文摘[Objective] cDNA of growth hormone releasing hormone (GHRH) receptor gene from Wuzhishan miniature pig was cloned and its sequence was also analyzed. [Method] Using genomic DNA extracted from porcine ear tissues of Wuzhishan miniature pig as the template, three pairs of primers were designed by the reported cDNA sequence of porcine GHRH, and cDNA was also amplified by RT-PCR. After being recovered and purified, PCR products were ligated to pMD18-T and then transformed into Escherichia coli DH5a. The transformation products were analyzed by PCR and double enzyme digestion to screen positive clones, and the positive clones were sequenced after identification in LB liquid medium. [ Result] cDNA of Wuzhishan miniature pig GHRH receptor gene was obtained successfully, and its length was 1 577 bp coding 423 amino acids. BLAST analysis showed that there were only 23 nuoleotides in difference between this fragment and pomine GHRH receptor gene, and its homology was 98%. However, both GHRH receptor genes were constituted by 423 amino acids with the sequence homology of 96%. [ Conclusion] This study provides theoretical basis for further studies on the dwarf mechanism of Wuzhishan miniature pig.

文摘Total atomic scattering factors for the 1s^23s ^2S stages for the lithium isoelectronic sequence from Z = 3 - 10 are calculated by using the full core plus correlation wave function. The influence of electron correlation on total atomic scattering factors is considered sufficiently in our calculation. For the 1s^2 3s ^2S states of the lithium isoelectronic sequence, the general functional behaviour of total atomic scattering factors is analyzed together for each state of the isoelectronic sequence.
