[Method] The effects of four factors such as buffer system,solid-liquid ratio,temperature,and extracting time on the lectin extraction result from P. yezoensis were investigated by the use of specific activity as the indicator. Then the lectin was purified by DEAE-Sepharose FF chromatography. The hemagglutination activity was used as the indicator,and the properties of the lectin such as carbohydrate specificity,divalent cations dependence,temperature,acidic and alkaline stability and others were detected. [Result] The optimal extraction conditions were:extraction ageat Tris-HCl buffer (25 mmol/L,pH=7.5 ),solid-liquid ratio of 1:5,extracting temperature of 40 ℃,and extracting time of 16 h. The activity of the lectin of P. yezoensis didn't change after 30 min of being heated at 50 ℃,which showed a certain extent of thermal stability,and the suitable pH value was 7-9. The lectin of P. yezoensis could combine with maltose,D-galactose,D-xylose and L-Arabinose,in which the binding force with the maltose was the strongest. The lectin activity was depended on the addition of Ca2+ and Mg2+. [Conclusion] The research had provided theoretical basis for the clarification of immune mechanism of P. yezoensis,as well as its high effective utilization.展开更多

Paclitaxel (PTX) is one of the most efficient anticancer drugs for the treatment of cancers through β-tubulin-binding. Our previous work indicated that a PTX-derivative hydrogelator Fmoc-Phe-Phe-Lys(paclitaxel)-...Paclitaxel (PTX) is one of the most efficient anticancer drugs for the treatment of cancers through β-tubulin-binding. Our previous work indicated that a PTX-derivative hydrogelator Fmoc-Phe-Phe-Lys(paclitaxel)-Tyr(H2PO3)-OH (1)could promote neuron branching but the underlying mechanism remains unclear. Using tubulin assembly-disassembly assay, in this work, we found that compound 1 obviously delayed more microtubule aggregation than PTX did. Under the catalysis of alkaline phosphatase, Fmoc-Phe-Phe-Lys(paclitaxel)- Tyr(H2PO3)-OH could self-assemble into nanofiber Fmoc-Phe-Phe-Lys(paclitaxel)-Tyr-OH with width comparable to the size of αβ-tubulin dimer. Therefore, we proposed in this work that nanofiber Fmoc-Phe-Phe-Lys(paclitaxel)-Tyr-OH not only inhibits the αβ-tubulin dimer binding to each other but also interferes with the plus end aggregation of microtubule. The constraint on the total energy in a given spatial region is given from holography by the mass of a black hole that just fits in that region, which leads to an UV/IR relation: the maximal energy density in that re...The constraint on the total energy in a given spatial region is given from holography by the mass of a black hole that just fits in that region, which leads to an UV/IR relation: the maximal energy density in that region is proportional to Mp^2/L^2, where Mp is the Planck mass and L is the spatial scale of that region under consideration. Assuming the maximal black hole in the universe is formed through gravitational collapse of perturbations in the universe, then the "Jeans" scale of the perturbations gives a causal connection scale RCC. For gravitational perturbations, RCC^-2= Max (H+ 2H^2, -H) for a fiat universe. We study the cosmological dynamics of the corresponding vacuum energy density by choosing the causal connection scale as the IR cutoff in the UV/IR relation, in the cases of the vacuum energy density as an independently conserved energy component and an effective dynamical cosmological constant, respectively. It turns out that only the case with the choice RCC^-2 = H+ 2H^2, could be consistent with the current cosmological observations when the vacuum density appears as an independently conserved energy component. In this case, the model is called holographic Ricci scalar dark energy model in the literature.展开更多

The adsorption properties of CFBC ash...Circulating fluidized bed combustion (CFBC) ash can be potentially used as supplementary cementitious materials for concrete production due to its desirable pozzolanic activity. The adsorption properties of CFBC ash-cement pastes were studied, and ordinary pulverized coal combustion (PCC) fly ash-cement pastes were used as control. The water-adsorption and superplasticizer (SP)-adsorption properties of the pastes were evaluated by water demand and UV-visible absorption spectroscopy respectively. The results show that CFBC ash-cement system has greater compressive strength as compared with PCC fly ash-cement system at a given curing age, although the water demand of the former is significantly higher than that of the latter. CFBC ash-cement pastes possess higher adsorption ability of aliphatic SP than PCC fly ash-cement pastes and the adsorption amount increases with an increase in ash replacement ratio. CFBC ash- cement pastes exhibit lower workability with higher slump loss. It is concluded that CFBC ash can be potentially used as supplementary cementitious material in concrete production, but the mix design of CFBC ash concrete needs to be appropriately adjusted. It is suggested that CFBC ash is used for the production of the concrete needing low flowability.展开更多

In order to i...Given the morphological characters of purple crabs identical with that of the tea-green ones except the body color, it is expected that the purple crabs and the tea-green ones were from the same species. In order to identify the phylogenetic relationships between them at the molecular level, we amplified and compared two conserved mitochondrial gene fragments of purple and tea-green crabs through PCR-SSCP and DNA sequencing analyses. The results showed that there was no distinct variation of DNA bands of both the CO1 and 16srRNA genes detected in polyacrylamide gels through SSCP analysis between purple and tea-green crab samples, and the following sequence analysis of color-different crab individuals presented 99.81% and 99.91% nucleotide sequence identity of the CO1 and 16srRNA genes respectively. These indicated that there was no species or subspecies differentiation between purple and tea-green crabs, meaning that they belonged to the same species, Portunus trituberculatus.展开更多
Ionic skins that demonstrate great advantages in the mechanical properties and multiple sensory capabilities are regarded as an attractive candidate to mimic functions of human skin.However,human skin is vulnerable to...Ionic skins that demonstrate great advantages in the mechanical properties and multiple sensory capabilities are regarded as an attractive candidate to mimic functions of human skin.However,human skin is vulnerable to be damaged under long-time sunlight irradiation,and most of the current ionic skins also lack a protection against harmful ultraviolet and infrared lights.Herein,this work develops a multifunctional ionic skin based on ionic conductive and light-managing hydrogels via a facile one-step locally confined polymerization.It is mechanically adaptable,able to modulate light in the broadband solar spectrum,and protect human skin from the harmful ultraviolet and infrared lights.Moreover,without complicated processing,the ionic skin enables human-machine interactions via wireless and optical camouflaged Morse codes.We believe this work will promote the development of smart wearable devices with multiple customizable functions.展开更多
