目的利用单细胞转录组测序(scRNA-seq)在单细胞水平系统描绘人早期红系发育的细胞图谱及分子轨迹。方法通过对人妊娠第36天CS15后期的胚胎卵黄囊组织scRNA-seq数据进行主成分分析(PCA);聚类分析以及拟时序分析;观察标记基因在不同细胞... 目的利用单细胞转录组测序(scRNA-seq)在单细胞水平系统描绘人早期红系发育的细胞图谱及分子轨迹。方法通过对人妊娠第36天CS15后期的胚胎卵黄囊组织scRNA-seq数据进行主成分分析(PCA);聚类分析以及拟时序分析;观察标记基因在不同细胞群体中的表达情况;预测红系细胞发生时间和发育路径。结果在CS15的卵黄囊的不同细胞群体中,红细胞及其祖细胞、前体细胞等的标记基因存在差异性表达,不同细胞的发生时序不同,且卵黄囊内的红细胞存在一定的异质性。CS15的卵黄囊内存在类型丰富的细胞,可能包括血管内皮细胞、造血前体细胞、粒细胞、不同状态的红细胞以及单核细胞等。结论推测人早期红系发育路径大致为血管内皮细胞分化产生造血前体细胞,进一步分化成为红髓祖细胞,最终产生巨核细胞和红细胞。 展开更多

在人-小鼠异种移植模型中提高人红细胞产生效率条件的优化

本研究旨在探讨在建立人-小鼠异种移植模型中提高人红细胞产生效率的条件。本研究从三个方面分别进行条件优化:①受体免疫缺陷小鼠品系选择,分别对NOD/SCID和NOD/SCID/IL2rγnull两种小鼠品系进行移植,考察移植后不同品系小鼠嵌合率和... 本研究旨在探讨在建立人-小鼠异种移植模型中提高人红细胞产生效率的条件。本研究从三个方面分别进行条件优化:①受体免疫缺陷小鼠品系选择,分别对NOD/SCID和NOD/SCID/IL2rγnull两种小鼠品系进行移植,考察移植后不同品系小鼠嵌合率和人红细胞发育情况;②考察照射剂量和照射方法对小鼠生存率的影响;③优化人造血干细胞培养条件及感染方法,检测重建率及红细胞产生率。结果表明,移植后4周检测小鼠骨髓细胞显示,NOD/SCID/IL2rγnull小鼠人CD45比例可达(51.4±13.9)%,并能检测到人红细胞(5.98±3.46)%;利用X射线分2次对小鼠进行总剂量2.5 Gy的照射,照射后小鼠生存率可以达到100%;利用新鲜脐带血分离的CD34+细胞,不经过冻融和分选过程,在分离后72 h内完成转染和移植过程,移植后嵌合率可达(51.4±13.9)%,并且人红细胞产生的效率为(5.98±3.46)%。结论:利用X射线分两次照射NOD/SCID/IL2rγnull小鼠后通过尾静脉注射2×105个细胞,新鲜分离的人脐带血CD34+细胞在移植前缩短体外培养时间(≤72 h),有利于提高免疫缺陷小鼠体内人红细胞产生效率。 展开更多

Non-monophyly of Rhacophorus rhodopus,Theloderma and Philautus albopunctatus Inferred from Mitochondrial 16S rRNA Gene Sequences 被引量:6

Mitochondrial gene fragments of 16S rRNA gene of four species (Rhacophorus rhodopus, R. reinwardtii, Philautus albopunctatus and P. rhododiscus ) from 11 populations were sequenced in this study. Homologous sequences of R. bipunctatus, Theloderma asperum, T. corticale and Buergeria japonica were obtained by screening the GenBank database. After excluding all gaps and ambiguous positions, aligned sequences were 500 bp in length with 115 variable sites and 92 parsimony-informative sites. Using B .japonica as an outgroup, phylogenetic relationships were analyzed using Bayesian inference, maximum parsimony and maximum likelihood methods. Our results indicated that neither of R. rhodopus and P. albopunctatus were monophyletic at the species level. The population of R. rhodopus from Hainan Island was more close to R. bipunctatus than to populations of R. rhodopus from Yunnan Province. Furthermore, the populations of R. rhodopus from Yunnan Province can be divided into two main lineages. Theloderma corticale and P.rhododiscus were clustered together and T. asperum was nested in P.albopunctatus. We considered that P. albopunctatus Liu and Hu, 1962, was the synonymy of T. asperum Boulenger, 1886, and suggested removing P. rhododiscus from Philautus into the genus Theloderma. 展开更多

Most studies of erythropoiesis have been performed either with the virally transformed MEL cells or with the scarce erythroblasts within unhomogeneous cell populations.MEL cells are the Friend virus transformed mouse erythroleukemia cells and have lost the capacity to respond to erythropoietin.Moreover,the research in the field of erythropoiesis has been severely hampered by the paucity of erythroblasts obtained from chick embryos,human,mouse,rat fetal liver,mouse spleen,or the bone marrow.In our paper,the fetal blood from newly slaughtered pregnant sheep meets the needs of studies as abundant staring biological materials at the nucleotide level and also reflects in vivo proliferative and differential status of erythroblasts.Appling the technique of mRNA differential display with minor modification,we directly compared cDNA fragmerts from the fetal blood erythroblasts and adult pregnant sheep leukocytes by PCR on DNA sequencing gels and revealed two differentially expressing bands.One is a novel gene fragment, which has homology with UV-sensitive cone opsin or rhodopsin by examinating the predicted amino acid sequence.Since the blood cells are sensitive to radiation,it can be safely inferred that this fragment participate in the process of cytocidal effect of the radiation.The other fragment is glucose induced gene /elongation factor 2 3′end noncoding region,which was first cloned from glucose induced bovine aortic smooth muscle cells.Using Northern blot hybridization method,authentic specific expression was confirmed. 展开更多

Due to differences in morphology, ecology and biology, the phylogenesis of Coniferopsida and Taxaceae remains controversial, and the phylogenetic relationship of Pseudotaxus is yet to be confirmed. Three rDNA regions, ITS1, 5.8S rDNA, and ITS2, were prepared for 77 species belonging to seven families of Coniferopsida, including Taxaceae. Maximum parsimony(MP)and Neighbor-joining(NJ) analyses were performed. Results showed that:(1) the Coniferopsida species formed three branches:(Taxaceae, Cephalotaxaceae, Cupressaceae, Taxodiaceae),(Araucariaceae, Podocarpaceae), and(Pinaceae);(2) Taxaceae included Taxus, Pseudotaxus, Amentotaxus, and Torreya; and(3) Pseudotaxus belonged to Taxaceae, with a support rate of 97%in MP analyses and 99% in NJ analyses. 展开更多

目的构建红系发育相关基因(erythoid develop associated gene,EDAG-1)原核表达系统并制备其单克隆抗体,以进一步研究 EDAG-1调控细胞增殖、分化及凋亡的分子机制,探讨其临床应用前景。方法本实验选用 pGEX-4T-3载体,构建含 GST 标签的 ... 目的构建红系发育相关基因(erythoid develop associated gene,EDAG-1)原核表达系统并制备其单克隆抗体,以进一步研究 EDAG-1调控细胞增殖、分化及凋亡的分子机制,探讨其临床应用前景。方法本实验选用 pGEX-4T-3载体,构建含 GST 标签的 pGEX-4T-3-EDAG-1原核表达载体,经过对表达体系进行优化,IPTG 诱导,获得较高效率的 GST-EDAG-1融合蛋白表达,纯化该蛋白,对 BALB/C 小鼠进行免疫。将免疫后小鼠的脾脏细胞和来源于同种系小鼠的 SP2/0骨髓瘤细胞(次黄嘌呤鸟嘌呤磷酸核糖转移酶缺陷型和胸腺核苷激酶缺陷型)采用 PEG 介导的融合方式进行细胞融合实验,通过 HAT 培养基对融合细胞进行初步筛选,进而通过 ELISA 实验对所获得杂交瘤细胞进行抗体表达验证。结果成功构建了含 GST 标签的pGEX-4T-3-EDAG-1原核表达载体,并表达出较高纯度的 EDAG-1蛋白,获得表达 EDAG-1抗体的单克隆杂交瘤细胞。通过对单克隆抗体的初步实验表明,所获单克隆抗体具有很好的特异性,较低的背景和较好的稳定性。结论通过原核表达途径制备 EDAG-1单克隆抗体是可行的,所获单克隆抗体具有很好的特异性,较低的背景和较好的稳定性。 展开更多
