研究以竹片为原料,经氢氧化钠蒸煮预处理后匀浆,再用次氯酸钠与氢氧化钠的混合液对匀浆液干燥物进行氧化处理,以考察处理过程对纤维素结构的影响和除杂效果。同时,利用扫描电镜(SEM)、红外光谱(FTIR)、热重分析(TG)和X射线衍射(XRD)对... 研究以竹片为原料,经氢氧化钠蒸煮预处理后匀浆,再用次氯酸钠与氢氧化钠的混合液对匀浆液干燥物进行氧化处理,以考察处理过程对纤维素结构的影响和除杂效果。同时,利用扫描电镜(SEM)、红外光谱(FTIR)、热重分析(TG)和X射线衍射(XRD)对各阶段竹材的变化进行表征。结果表明:以8%(w/v)浓度的氢氧化钠溶液蒸煮竹材30 min,6%(w/v)的次氯酸钠(NaClO)与8%的氢氧化钠(NaOH)混合液以4∶1的体积比在40℃条件下氧化处理24 h后,得到的竹纤维平均直径为10.33μm,半纤维素和木质素可被有效去除,相对结晶度为62.93%,比原材料略有提高,制得竹纤维的热稳定性相比原材料和碱处理后样品出现下降。 展开更多

The influences of chromatographic parameters on the resolution were studied to determine the optimal conditions for purification. The purified endoglucanase was obtained by gel filtration by Superdex 75 prep grade with an activity recovery of 92.8% and the purification factor 4.2. The sample volume should be below 6% of the column bed volume and the column bed height L≥12.0 cm. The optimum catalysis temperature and pH for the enzyme were 55 ℃ and 4.5—5.0, respectively. The cellulase was stable at pH ranging from 4.0 to 6.0 and temperature below 60 ℃. 展开更多

A bacterium hydrolyzing carboxymethylcellulose, isolated from Antarctic sea ice, was identified as Pseudoalteromonas sp. based on 16S rDNA gene sequences and named as Pseudoalteromonas sp. AN545. The extracellular endo-1,4-β-glucanase AN-1 was purified successively by ammonium sulfate precipitation, DEAE-Sepharose ion exchange chromatography and Sephadex G-75 gel filtration chromatography. The molecular mass of AN-1 was estimated to be 47.5 kDa utilizing SDS-PAGE and gel chromatography analysis. AN-1 could hydrolyze caboxymethylcellulose, avicel and β-glucan, but not cellobiose, xylan and p-Nitrophenyl-β-D-glucopyranoside. The optimal temperature and pH for the β-glucanase activity of AN-1 were determined to be at 30℃ and pH 6.0, respectively. AN-1 was stable at acidic solutions of pH 5.0-6.5 and temperatures below 30℃ for 1 h. Moreover, the specific activity was enhanced by Ca2+ and Mg2., and inhibited by Cu2+. The kinetic parameters Michaelis constant (Km) and maximum velocity (Vmax) of AN-1 were 3.96 mg/mL and 6.06×10-2 mg/(min.mL), respectively. 展开更多

This paper describes an innovative method for the immobilization of acylase I, which was entrapped into the CA-CTA micropore membrane. The most suitable casting solutions proportion for immobilizing the enzyme was obtained through orthogonal experiment. Properties of the enzyme membrane were investigated and compared with those of free enzyme and blank membrane. The thermal stability and pH stability of the enzyme inside the membrane were changed by immobilization. The optimum pH was found to be 6.0, which changes 1.0 unit compared with that of free acylase I. The optimum temperature was found to be about 90℃, which is higher than that of free acylase I (60℃). Experimental results showed that immobilization had effects on the kinetic parameters of acylase I. 展开更多

Deregulation of the FGF/FGFR signaling pathway has been associated with multiple development syndromes and cancers, and thus therapeutic strategies targeting FGFs and FGFR in human cancer are currently being explored.However, few studies on the FGF/FGFR pathway have been conducted in sarcoma, which has a poor outcome with traditional treatments such as surgery, chemotherapy, and radiotherapy. Hence, in the present review, we provide an overview of the role of the FGF/FGFR pathway signal in sarcoma and FGFR inhibitors, which might be new targets for the treatment of sarcomas according to recent research. 展开更多

Microscopic analysis of the skin and cuticle of wool fibers in Dubska pramenka show different qualitative parameters. The research study included different parts of the body and the samples of the wool fibers from different regions of the body. Histological description shows difference in the basic structurers of the skin in Dubska pramenka. A very pronounced thin epidermis, while dermis and hypodermis are more developed on the samples from the shoulder. Cornified flakes-cuticle of the root of the tail was of a fine structure, and comified flakes from the rump showed the features of the rough wool fibers. "Transitional form of cuticle" was dominant on the shoulder. The qualitative parameters have a significant influence on the overall quality of wool in general, and the research contributes to a greater usability value of the wool and development of livestock farming in areas in a broader sense. 展开更多

The adsorption of fibrinogen can be used as a quick indicator of surface haemocompatibility because of its prominent role in coagulation and platelet adhesion. In this work the molecular interaction between fibrinogen and a modified titanium oxide surface/platelet has been studied by quartz crystal microbalanee with dissipation (QCM-D) in situ. In order to further characterize the conformation of adsorbed fibrinogen, αC and γ-chain antibody were used to check the orientation and denaturation of fibrinogen on solid surface. QCM-D investiga- tions revealed the fibrinogen have the trend to adsorb on hydropllilic surface in a side-on orientation by positively charged αC domains, which would reduce the exposure of platelet bonding site on γ chain and enable less platelet adhesion and be activated. These obser- vations suggest that certain conformations of adsorbed fibrinogen are less platelet adhesive than others, which opens a possibility for creating a non-platelet adhesive substrates. 展开更多

The nitrogen adsorption isotherms at 77.69 K were measured for two samples of activated carbon fibers and their microstructures were investigated. Among established isotherm equations, the Dubinin-Radushkevieh equation showed the best agreement with the experimental data, while the Langmuir equation showed a large deviation when employed at low relative pressures. The MP method, t-method and as-method were used to analyze the pore size distribution. The calculated average pore widths and BET (Brunauer-Emmett-Teller) surface areas for the sample A-13 were 0.86 nm and 1 286.60 m^2/g, while for the sample A-16, they were 0.82 nm and 1 490.64 m^2/g. The sample with larger pore width was more suitable to be used as additive in chemical heat pumps, while the other one could be used as adsorbent in adsorption refrigeration systems . 展开更多
