目的探讨血清乙肝病毒核心相关抗原(HBcr Ag)及线性化乙型肝炎病毒表面抗原(HBs Ag)对隐匿性乙型肝炎(OBI)的诊断价值。方法选择OBI患者63例(OBI组)、体检健康者150例(对照组),采用化学发光酶联免疫分析法检测两组血清HBcr Ag、线性化HB... 目的探讨血清乙肝病毒核心相关抗原(HBcr Ag)及线性化乙型肝炎病毒表面抗原(HBs Ag)对隐匿性乙型肝炎(OBI)的诊断价值。方法选择OBI患者63例(OBI组)、体检健康者150例(对照组),采用化学发光酶联免疫分析法检测两组血清HBcr Ag、线性化HBs Ag,采用荧光定量PCR法检测血清HBV DNA。采用受试者工作特征(ROC)曲线分析HBcr Ag、线性化HBs Ag二者单独及联合检测对OBI的诊断效能。结果 OBI组和对照组血清HBcr Ag阳性率分别为90.5%和10.7%,线性化HBs Ag阳性率分别为42.9%和6.0%,两组比较差异均有统计学意义( χ~2分别为125.5、42.9,P均<0.05)。相关分析显示,HBV DNA与HBcr Ag、HBV DNA、线性化HBs Ag均呈正相关(R2分别为0.471 4、0.122 2、0.086 8,P<0.05或<0.01)。ROC曲线分析显示,血清HBcr Ag、线性化HBs Ag诊断OBI的曲线下面积(AUC)分别为0.911(95%CI为0.864~0.946)、0.683(95%CI为0.616~0.745),二者联合检测诊断OBI的AUC为0.895(95%CI为0.846~0.933),血清HBcr Ag对OBI的诊断效能显著优于线性化HBs Ag(Z=6.134,P<0.05),二者联合检测并不能提高其对OBI的诊断效能(Z=0.657,P>0.05)。当血清HBcr Ag、线性化HBs Ag的最佳临界值分别为100 U/m L、0.48 IU/m L时,血清HBcr Ag诊断OBI的敏感性显著高于线性化HBs Ag( χ~2=121.4,P<0.05),特异性稍低于线性化HBs Ag( χ~2=3.034,P>0.05),二者联合检测诊断OBI的敏感性和特异性较单独检测HBcr Ag并未明显提高( χ~2分别为1.322、3.802,P均>0.05)。结论血清HBcr Ag与HBV DNA存在良好的相关性,对OBI的诊断效能优于线性化HBs Ag,但联合检测HBcr Ag、线性化HBs Ag并不能提高其诊断效能;血清HBcr Ag可单独作为诊断OBI的血清学指标。 展开更多

快速稳定地将外源基因插入杆状病毒基因组获得重组杆状病毒,是提高昆虫杆状病毒表达系统应用效率的关键技术之一。在Ac Bacmid的基础上,通过λRed同源重组技术敲除病毒基因组lef2和orf1629基因的3'端部分序列获得RDAc Bacmid-DualKo,并... 快速稳定地将外源基因插入杆状病毒基因组获得重组杆状病毒,是提高昆虫杆状病毒表达系统应用效率的关键技术之一。在Ac Bacmid的基础上,通过λRed同源重组技术敲除病毒基因组lef2和orf1629基因的3'端部分序列获得RDAc Bacmid-DualKo,并用Bsu36Ⅰ线性化之后,经重叠PCR获得5'端和3'端各含有50 bp同源臂及以绿色荧光蛋白(GFP)基因作为外源基因的表达盒片段,再将线性化杆状病毒载体片段与表达盒片段共转染sf9细胞,3~4 d即可获得重组杆状病毒rvAcBacmid-GFP。SDS-PAGE和Western blot结果表明,被rvAcBacmid-GFP感染的sf9细胞能高效表达绿色荧光蛋白。该方法通过线性化复制缺陷型杆状病毒载体和携带有同源臂及外源基因的重叠PCR片段,在胞内完成同源重组获得重组杆状病毒,避免了繁琐的病毒空斑纯化和分子克隆操作,获得的重组杆状病毒中无不稳定因子BAC及影响下游应用研究的抗性基因,为快速构建重组杆状病毒高效表达外源基因及相关研究应用提供了新的技术平台。 展开更多

AIM: To explore the expression and replication of hepatitis B virus (HBV) DNA in primary duck hepatocytes (PDHs).METHODS: Complete HBV genome was transfected into PDHs by electroporation (transfected group, 1.19×1012copies of linear HBV DNA/1×107 PDHs). After 1-5 d of transfection, HBsAg and HBeAg in the supernatant and lysate of PDHs were measured with the IMX System.Meanwhile, replicative intermediates of HBV DNA were analyzed by Southern blotting and Dot blotting. PDHs electroporated were used as control group.RESULTS: HBsAg in the hepatocyte lysates of transfected group was 15.24 (1 d), 14.55 (3 d) and 5.13 (5 d; P/N values, positive≥2.1) respectively. HBeAg was negative (<2.1). Both HBsAg and HBeAg were negative in the supernatant of transfected group. Dot blotting revealed that HBV DNA was strongly positive in the transfected group and negative in the control group. Southern blot analysis of intracellular total DNA indicated that there were relaxed circular (rc DNA), covalently closed circular (ccc DNA), and single-stranded (ss DNA) HBV DNA replicative intermediates in the transfected group, there was no integrated HBV DNA in the cellular genome. These parameters were negative in control group.CONCLUSION: Expression and replication of HBV genes can occur in hepatocytes from non-mammalian species.HBV replication has no critical species-specificity, and yet hepatic-specific regulating factors in hepatocytes may be essential for viral replication. 展开更多

Objective To explore the contrast-enhanced ultrasonographic features for quantitative assessment of hepatic fibrosis. Methods 86 patients with chronic viral hepatitis B were enrolled in this study from March 2007 to August 2009. The patients were classified into 5 groups (S0-S4) according to fibrosis stage evaluated with ultrasound guided liver biopsy. New contrast-enhanced ultrasonography (CEUS) features including area under the time-intensity curve (TIC) of portal venous phase/hepatic arterial phase (Qp/Qa) and intensity of portal venous phase/hepatic arterial phase (Ip/Ia) were used to detect the blood supply ratio (portal vein/hepatic artery) in each group. Arrival time of portal vein trunk (Tp) and decreasing rate of TIC (β ) were also analyzed. Results Qp/Qa and Ip/Ia decreased from S0 to S4, while Tp and β increased. These 4 features were significantly correlated with the degree of fibrosis (P<0.001) and were significantly different among the five groups (P<0.001). Sensitivity and specificity of Ip/Ia were 80% and 86% for groups ≥S1, 75% and 86% for groups ≥ S2, 71% and 84% for groups ≥ S3, and 76% and 80% for group S4, respectively. Sensitivity and specificity of Qp/Qa were 70% and 88% for groups ≥ S1, 80% and 76% for groups ≥ S2, 74% and 70% for groups ≥ S3, and 81% and 95% for group S4, respectively. Conclusion Ip/Ia and Qp/Qa could be adopted as reliable, non-invasive features for quantitative assessment of hepatic fibrosis. 展开更多

The peritrophic membrane plays an important role in the defense system of the arthropod gut. The digestive tract is considered one of the major tissues targeted by white spot syndrome virus (WSSV) in shrimp. In this study, the nucleotide sequence encoding peritrophin-like protein of Litopenaeus vannamei (LvPT) was amplified from a yeast two-hybrid library of L. vannamei. The epitope peptide of LvPT was predicted with the GenScript OptimumAntigenTM design tool. An anti-LvPT polyclonal antibody was produced and shown to specifically bind a band at -27 kDa, identified as LvPT. The LvPT protein was expressed and its concentration determined. LvPT dsRNA (4 pg per shrimp) was used to inhibit LvPT expression in shrimp, and a WSSV challenge experiment was then performed with reverse gavage. The pleopods, stomachs, and guts were collected from the shrimp at 0, 24, 48, and 72 h post-infection (hpi). Viral load quantification showed that the levels of WSSV were significantly lower in the pleopods, stomachs, and guts of shrimp after LvPT dsRNA interference than in those of the controls at 48 and 72 hpi. Our results imply that LvPT plays an important role during WSSV infection of the digestive tract. 展开更多

In this paper, we consider two nonlinear models for viral infection with humoraL immu- nity. The first model contains four compartments; uninfected target cells, actively infected cells, free virus particles and B cells. The second model is a modification of the first one by including the latently infected cells. The incidence rate, removal rate of infected cells, production rate of viruses and the latent-to-active conversion rate are given by more general nonlinear functions. We have established a set of conditions on these general functions and determined two threshold parameters for each model which are sufficient to determine the global dynamics of the models. The global asymptotic stability of all equilibria of the models has been proven by using Lyapunov theory and applying LaSalle's invariance principle. We have performed some numerical simulations for the models with specific forms of the general functions. We have shown that, the numerical results are consistent with the theoretical results. 展开更多

OBJECTIVE: To explore the relationship between Renying pulse (carotid) augmentation index (AI) and Cunkou pulse condition in different blood pres- sure groups, and the clinical significance of Reny- ing and Cunkou pulse parameters to reflect vascu- lar function. METHODS: Eighty-six patients with essential hyper- tension (EH) and 52 individuals with normal blood pressure (control group) between and January 2012 were included September 2010 this study. Reny- ing pulse AI was examined by a new diagnostic tool (ALOKA ProSound Alpha 10) --wave intensity (Wl) that is calculated as the product of the deriva- tives of the simultaneously recorded blood pres- sure changes (dP/dt) and blood-flow-velocity changes (dU/dt), while Cunkou pulse condition was detected by DDMX-100 Pulse Apparatus inboth EH and control groups. A multifactorial corre- lation analysis was performed for data analysis. RESULTS: After adjusting for potential confound- ing variables, in the EH group, AI was positively cor- related with ts, w2/t (rts=0.225, P〈0.05; rw2/t=0.230, P〈 0.05) and negatively correlated with hs, hs/hl and w2 (rhs=- 0.393,P〈0.01 ;rhs/l=- 0.444, P〈0.01 ;rw2=- 0.389, P〈0.01). In the control group, AI was positively cor- related with t3, t4, ts and w, (rt3=0.595, P〈0.01; r,4= 0.292, P〈0.05; rt5=0.318, P〈0.05; rw1=0.541, P〈0.01) and negatively correlated with h1, h2, h3, Ad and A (rh1= - 0.368, P〈0.05; rh2= - 0.330, P〈0.05; rh3= - 0.327, P〈 0.05; rAd=- 0.322, P〈0.05; rA=- 0.410, P〈0.01). In the total sample group (EH plus control group, n= 138), AI was positively correlated with t, ts, w1 and w, (rt=0.257, P〈0.01; rt5=0.266, P〈0.01; rw1=0.184, P〈 0.05; rw/t=0.210, P〈0.05) and negatively correlated with hs, hs/hl, w2 and Ad (rhs= - 0.230, P〈0.01; rh5/h1= - 0.218, P〈0.05; rw2= - 0.267, P〈0.01; rAd= - 0.246, P〈0.01). Multiple linear regression analysis was car- ried out to model the relationship (F=7.887, P〈 0.001).CONCLUSION: Renying pulse AI can effectively pre- dict arterial stiffness in synchrony with the manifes- tations of Cunkou pulse in elderly patients with hy- pertension. We consider constant con- trols and thereafter by incorporating the theory of Volterra-Lyapunov stable matrices into the classical method of Lyapunov functions, we present an approach for global stability analysis of HIV/AIDS. The analysis and results presented in this paper make building blocks toward a comprehensive study and deeper understanding of the funda- mental mechanism in HIV/AIDS. Human immunodeficiency virus (HIV) is a lenti-virus (a member of the retrovirus family) that causes acquired immunodeficieney syndrome (AIDS), a critical condition in humans in which progressive failure of the immune system allows life-threatening opportunis- tic infections. Over the past few years HIV has been spreading rapidly in the popula- tion. Almost, everyday there are thousands of new human cases of HIV infection being recorded in the world and these occur in almost every country of the world. However, the spread of HIV is relatively faster in the developing countries as compared to developed countries because developing countries have limited resources. Worldwide, 70% of HIV infections in the adults have been transmitted through heterosexual contact and vertical transmission accounts for more than 90~ of global infection in infants and children. In this paper, we propose a nonlinear mathematical model to study the spread of HIV by considering transmission of disease by heterosexual contact and vertical transmission. A stage structured model is proposed and analyzed by considering the total population variable and dividing the whole population under consideration into three stages: chil- dren, adults and old. Also, in this paper it is assumed that the rates of recruitment are different in different groups of population. Equilibria of the model and their stability are also discussed. Using the stability theory of differential equations and computer simu- lation, it is shown that due to the increase in the awareness of the disease in the adult class the total infective population decreases in the region under consideration. 展开更多
