左金方及其组方成分对人胃癌细胞SGC-7901凋亡及膜电位的影响 被引量:10

目的比较左金方及其组方成分(黄连、吴茱萸及巴马汀、小檗碱、药根碱、吴茱萸碱、吴茱萸次碱)对人胃癌细胞SGC-7901生长的活性抑制、凋亡诱导和膜电位的影响。方法采用MTT比色法观察受试药物对SGC-7901的生长抑制情况;流式细胞仪PI单染... 目的比较左金方及其组方成分(黄连、吴茱萸及巴马汀、小檗碱、药根碱、吴茱萸碱、吴茱萸次碱)对人胃癌细胞SGC-7901生长的活性抑制、凋亡诱导和膜电位的影响。方法采用MTT比色法观察受试药物对SGC-7901的生长抑制情况;流式细胞仪PI单染亚二倍体峰分析细胞凋亡率;以及JC-1试剂盒测定细胞膜电位。结果左金方、黄连、巴马汀和小檗碱分别作用SGC-7901细胞后,均能抑制细胞活性,且呈时间依赖性;1.0 mg·m L-1左金方和对应浓度的黄连(0.857 mg·m L-1)、巴马汀(9.19μg·m L-1)、小檗碱(39.85μg·m L-1)作用于SGC-7901细胞48 h,细胞凋亡率分别为(36.40±1.59)%,(21.90±0.27)%,(9.32±2.58)%和(13.89±3.56)%,与对照组(1.69±1.91)%比较差异均有统计学意义(P<0.01),左金方与黄连、巴马汀、小檗碱组间比较差异亦具有统计学意义(P<0.01)。对应浓度的吴茱萸(0.143mg·m L-1)、药根碱(4.86μg·m L-1)、吴茱萸碱(0.17μg·m L-1)与吴茱萸次碱(0.19μg·m L-1)几乎无作用。JC-1染色检测结果显示,左金方及相应浓度的黄连、巴马汀和小檗碱作用SGC-790l细胞4 h后,可以明显引起细胞线粒体膜电位的下降,实验结果与凋亡检测结果相一致。结论左金方、黄连、巴马汀和小檗碱均能通过降低细胞线粒体膜电位,诱导胃癌细胞的凋亡,且左金方的作用优于黄连、小檗碱和巴马汀,但吴茱萸及其有效成分几乎无作用。 展开更多

目的探讨桃红四物汤组方药物主要水溶性有效单体成分(以下简称单体成分组方)对早期骨折愈合及血管内皮生长因子(VEGF)表达的量效关系。方法将90只SPF级雄性12周龄SD大鼠随机分为空白组、模型组、假手术组,单体成分组方低、中、高剂量6个... 目的探讨桃红四物汤组方药物主要水溶性有效单体成分(以下简称单体成分组方)对早期骨折愈合及血管内皮生长因子(VEGF)表达的量效关系。方法将90只SPF级雄性12周龄SD大鼠随机分为空白组、模型组、假手术组,单体成分组方低、中、高剂量6个组,每组15只。大鼠股骨干骨折模型制备后,分别于3、7、14 d 3个时相点分批处死大鼠,处死前记录大鼠一般情况及关节肿胀度,处死后通过观察大鼠血清中VEGF的浓度、显微CT(Micro-CT)成像描述、组织病理学改变等探讨单体成分组方对早期骨折愈合及VEGF表达的量效关系。结果与空白组比较,假手术组在3、7、14 d 3个时相点大鼠肢体肿胀程度不明显(P> 0.05);与假手术组比较,模型组大鼠在3、7、14 d 3个时相点大鼠肢体肿胀程度明显(P <0.01);与模型组比较,各剂量组大鼠在3、7、14 d 3个时相点肢体肿胀程度均有明显减轻,其中以单体成分组方高剂量减轻最为明显(P <0.05或P <0.01)。与空白组比较,假手术组在3、7、14 d 3个时相点大鼠血清VEGF浓度表达均有所降低(P <0.05);与假手术组比较,模型组在3、7、14 d 3个时相点大鼠血清VEGF浓度表达显著降低(P <0.01);与模型组比较,各剂量组在3、7、14 d 3个时相点大鼠血清VEGF浓度表达均有明显增加,其中以单体成分组方高剂量增加最为明显(P <0.05或P <0.01)。与模型组比较,单体成分组方各剂量组在3、7、14 d 3个时相点大鼠骨体积(BV)值、大鼠骨体积分数(BV/TV)值均有所增加,其中均以单体成分组方高剂量组增加最为明显(P <0.05或P <0.01),单体成分组方低、中剂量组间差异不明显(P> 0.05)。结论桃红四物汤主要水溶性有效单体成分组方可以减轻大鼠肢体肿胀程度,增加大鼠血清中VEGF浓度,并上调大鼠骨体积(BV)值、大鼠骨体积分数(BV/TV)值,说明桃红四物汤有效单体成分组方对大鼠VEGF的表达及早期骨折的愈合具有明显促进作用;其疗效与剂量、时相点呈正相关性。 展开更多

目的:制备以生脉散中有效成分人参皂苷Rg1、人参皂苷Rb1、麦冬皂苷DT-13及五味子醇甲为配基的亲和介质。方法:采用4个有效成分与环氧活化琼脂糖凝胶6B(EAS 6B)偶联,制备亲和介质。建立HPLC-DAD对4个有效成分同时在线分离与检测,以偶联... 目的:制备以生脉散中有效成分人参皂苷Rg1、人参皂苷Rb1、麦冬皂苷DT-13及五味子醇甲为配基的亲和介质。方法:采用4个有效成分与环氧活化琼脂糖凝胶6B(EAS 6B)偶联,制备亲和介质。建立HPLC-DAD对4个有效成分同时在线分离与检测,以偶联率为指标,通过L9(34)正交试验考察反应温度,反应时间和投料比对偶联反应的影响,优化制备工艺条件。结果:4个成分均可与EAS 6B发生偶联,最佳制备工艺为反应温度25℃,反应时间24 h,添加10倍量生脉散有效成分组方。人参皂苷Rg1、人参皂苷Rb1、麦冬皂苷DT-13及五味子醇甲亲和介质的偶联率分别为27.4%,22.3%,50.0%,24.5%。结论:成功制备了以生脉有效成分组为配基的亲和介质,为进一步识别该方的作用靶点提供技术手段,同时也为制备类似结构中药复方的亲和介质提供实验依据。 展开更多

Quadratic Discrimination Function (QDF) is commonly used in speech emotion recognition, which proceeds on the premise that the input data is normal distribution. In this paper, we propose a transformation to normalize the emotional features, emotion recognition. Features based on prosody then derivate a Modified QDF (MQDF) to speech and voice quality are extracted and Principal Component Analysis Neural Network (PCANN) is used to reduce dimension of the feature vectors. The results show that voice quality features are effective supplement for recognition, and the method in this paper could improve the recognition ratio effectively. 展开更多

Authenticated skip list is an important data structure used in cloud data integrity verification. How to get the membership proof of the element in authenticated skip list efficiently is an important part of authentication. Kaouthar Blibech and Alban Gabillon proposed a head proof and a tail proof algorithms for the membership proof of elements in the authenticated skip list. However, the proposed algorithms are uncorrelated each other and need plateau function. We propose a new algorithm for computing the membership proof for elements in the authenticated skip list by using two stacks, one is for storing traversal chain of leaf node, the other is for storing authentication path for the leaf. The proposed algorithm is simple and effective without needing plateau function. It can also be applicable for other similar binary hash trees. 展开更多

With its long-term empirical clinical practice and increasing number of health benefits reported,Chinese Materia Medica(CMM)is gaining increasing global acceptance.Importantly,the identification of chemical constituents in vitro and exposed forms in vivo is a prerequisite for understanding how CMM formulae prevent and treat diseases.This review systematically summarizes the exciting and magical journey of CMM components from compound formulae to where they fight,the possible structural transformation of CMM components in vitro and in vivo,and their pharmacological contribution.When a decoction is prepared,significant chemical reactions are observed,including degradation and production of polymers and self-assembling supramolecules,leading to the construction of a component library with diverse decoction structures.After ingestion,compounds pass through the intestinal and blood-brain barriers and undergo a more wonderful journey involving the gut microbiota,microbial enzymes,and endogenous drug-metabolizing enzymes(mainly liver enzymes).At this stage,they are modified and assembled into novel and complex compounds,such as newly generated metabolites,conjugates,and self-assembling superamolecules.This review might provide a strategic orientation to explore the active compounds of CMM formulae in vivo. 展开更多

Coal preparation is an integral part of the coal commodity supply chain. This stage of post-mining, pre-utilization beneficiation uses low-cost separation technologies to remove unwanted mineral matter and moisture which hinder the value of the coal product. Coal preparation plants typically employ several parallel circuits of cleaning and dewatering operations, with each circuit designed to optimally treat a specific size range of coal. Recent innovations in coal preparation have increased the efficiency and capac- ity of individual unit operations while reinforcing the standard parallel cleaning approach. This article, which describes the historical influences and state-of-the-art design for the various coal preparation unit operations, is organized to distinguish between coarse/intermediate coal cleaning and fine/ultrafine coal cleaning. Size reduction, screening, classification, cleaning, dewatering, waste disposal unit operations are particularly highlighted, with a special focus on the LI.S. design philosophy. Unquestionably,aircraft industry plays an important part in both economy and security of a nation.However,few of articles have dealt with the spatial development of this industry sector.The purpose of this article is to analyze the dynamics of agglomeration and diffusion of aircraft industry.In a historical view,with some spatial analysis methods,this research discusses the characteristics and patterns of aircraft industry's spatial organization and evolution,globally,nationally and regionally respectively.We find out there is a ‘Matthew effect' in aircraft industry of the world,and the spatial evolution of the industry is consistent with the nation's industrialization process.Then,it concludes that the main agglomeration forces consist of capital,talents,technology and cluster's advantages,and the main diffusion forces include comparative advantages,cost and risk sharing,emerging markets,development policy for backward regions and military requirements.All the factors can be divided into market forces making the spatial development of aircraft industry normal and non-market forces making that irregular.In particular,lessons from the USA and France are expected to be conducive to the rise of China's aircraft industry in the future. 展开更多

Comparison of non-unitary and generalized unitary scattering theories is done by means of nuclear monodromy (equivalence of Schrodinger and Maxwell time-independent equations), tunneling and radioactivity. Radioactivity is important part of physics and our life. Its importance stretches from medicine as far as to war strategies. We present theoretical approach to achieve better understanding of the radioactive decay when modified quantum theory is applied. It can be done by updating existing codes to understand better construction of the world and terms and conditions of our existence. The theory modifications are strictly connected with the unimodular M matrix and Wronskian matrices (i.e. their determinants named Wronskians) which create underpinning of so called monodromy being two track wave-function evolution. 展开更多

The results showed that:( 1) The co-seismic displacement of Yutian station,about 54 km from the epicenter,is the most obvious,particularly in the EW component,with a change of about 52.5 ± 11mm,which is more than three times the mean-square error of calculating precision.( 2) In the Yutian reference station,the biggest variation in the EW component appeared within 1 minute after the earthquake.( 3) The change in the NS component is not great. 展开更多

A novel layered method was proposed to solve the problem of Web services composition.In this method,services composition problem was formally transformed into the optimal matching problem of every layer,then optimal matching problem was modeled based on the hypergraph theory,and solved by computing the minimal transversals of the hypergraph.Meanwhile,two optimization algorithms were designed to discard some useless states at the intermediary steps of the composition algorithm.The effectiveness of the composition method was tested by a set of experiments,in addition,an example regarding the travel services composition was also given.The experimental results show that this method not only can automatically generate composition tree whose leaf nodes correspond to services composition solutions,but also has better performance on execution time and solution quality by adopting two proposed optimization algorithms. 展开更多
