γ射线照射对两种组织修复细胞的直接效应与W_(11)-a_(12)的作用 被引量:6

目的 探讨电离辐射是否对成纤维细胞和血管内皮细胞的增殖游走功能具有直接抑制作用及其抑制程度 ,以及W1 1 a1 2 的促愈作用。方法 采用培养的单层 3T3细胞 (小鼠胚胎FBs株 )和培养的单层ECV30 4细胞 (人脐静脉内皮细胞株 )划痕伤... 目的 探讨电离辐射是否对成纤维细胞和血管内皮细胞的增殖游走功能具有直接抑制作用及其抑制程度 ,以及W1 1 a1 2 的促愈作用。方法 采用培养的单层 3T3细胞 (小鼠胚胎FBs株 )和培养的单层ECV30 4细胞 (人脐静脉内皮细胞株 )划痕伤口模型进行离体照射 ,观察痕隙闭合速度。结果 在 6Gy照射后培养的单层 3T3细胞与单层L ECV30 4细胞划痕“伤口”闭合明显延缓。单层 3T3细胞在伤后 10h对照组伤口完全闭合 ,而照射组仅闭合 77% ;照射组单层ECV30 4细胞在伤后 12h仅为对照组的 83 6 %。W1 1 a1 2 用药组对单层 3T3细胞与ECV30 4细胞“划痕”伤口的闭合均有促进作用。结论 放射损伤对成纤维细胞与血管内皮细胞的增殖游走有直接抑制作用 ,这是伤口难愈的重要原因之一。促进伤口细胞游走和增殖是W1 1 a1 2 作用的一个重要途径。 展开更多

贝复剂治疗褥疮30例临床体会

Mesenchymal stem cells (MSCs) have great potential for treating various diseases, especially those related to tissue damage involving immune reactions. Various studies have demonstrated that MSCs are strongly immunosuppressive in vitro and in vivo. Our recent studies have shown that un-stimulated MSCs are indeed incapable of immunosuppression; they become potently immunosuppressive upon stimulation with the supernatant of activated lymphocytes, or with combinations of IFN-γ, with TNF-α, IL-1α or IL-1β. This observation revealed that under certain circumstances, inflammatory cytokines can actually become immunosuppressive. We showed that there is a species variation in the mechanisms of MSC-mediated immunosuppression: immunosuppression by cytokine-primed mouse MSCs is mediated by nitric oxide (NO), whereas immunosuppression by cytokine-primed human MSCs is executed through indoleamine 2, 3-dioxygenase (IDO). Additionally, upon stimulation with the inflammatory cytokines, both mouse and human MSCs secrete several leukocyte chemokines that apparently serve to attract immune cells into the proximity with MSCs, where NO or IDO is predicted to be most active. Therefore, immunosuppression by inflammatory cytokine-stimulated MSCs occurs via the concerted action of chemokines and immune-inhibitory NO or IDO produced by MSCs. Thus, our results provide novel information about the mechanisms of MSC-mediated immunosuppression and for better application of MSCs in treating tissue injuries induced by immune responses. 展开更多

Objective: To testify the effect of the stem cells derived from the widely distributed fat tissue on repairing full-thickness hyaline cartilage defects. Methods: Adipose-derived stem cells (ADSCs) were derived from adipose tissue and cultured in vitro. Twenty- seven New Zealand white rabbits were divided into three groups randomly. The cultured ADSCs mixed with calcium alginate gel were used to fill the full-thickness hyaline cartilage defects created at the patellafemoral joint, and the defects repaired with gel or without treatment served as control groups. After 4, 8 and 12 weeks, the reconstructed tissue was evaluated macroscopically and microscopically. Histological analysis and qualitative scoring were also performed to detect the outcome.Results: Full thickness hyaline cartilage defects were repaired completely with ADSCs-derived tissue. The result was better in ADSCs group than the control ones. The microstructure of reconstructed tissue with ADSCs was similar to that of hyaline cartilage and contained more cells and regular matrix fibers, being better than other groups. Plenty of collagen fibers around cells could be seen under transmission electron microscopy. Statistical analysis revealed a significant difference in comparison with other groups at each time point (t=4.360, P〈0.01). Conclusion: These results indicate that stem cells derived from mature adipose without induction possess the ability to repair cartilage defects 展开更多

Since 1980s, the rapid development of tissue engineering and stem cell research has pushed regenerative medicine to a new fastigium, and regenerative medicine has become a noticeable research field in the international biology and medicine. In China, about 100 million patients need repair and regeneration treatment every year, while the number is much larger in the world. Regenerative medicine could provide effective salvation for these patients. Both Chinese Academy of Sciences and Chinese Academy of Engineering have made roadmaps of 2010-2050 and 2011-2030 for regenerative medicine. The final goal of the two roadmaps is to make China go up to leading position in most research aspects of regenerative medicine. In accord with this strategy, the government and some enterprises have invested 3-5 billion RMB (0.5-0.8 billion USD)for the research on regenerative medicine. In order to push the translation of regenerative medicine forward -- from bench to bedside, a strategic alliance has been established, and it includes 27 top-level research institutes, medical institutes, colleges, universities and enterprises in the field of stem cell and regeneration medicine. Recently the journal, Science, has published a special issue-Regenerative Medicine in China, consisting of 35 papers dealing with stem cell and regeneration, tissue engineering and regeneration, trauma and regeneration and bases for tissue repair and regenerative medicine. It is predicated that a greater breakthrough in theory and practice of regenerative medicine will be achieved in the near future (20 to 30 years). 展开更多
