In the last decade, the stock conservation and aquaculture industry of this species have been facing severe challenges because of wild population collapse and degeneration of important economic traits. However, genes contributing to growth and immunity in L. crocea have not been thoroughly analyzed, and available molecular markers are still not sufficient for genetic resource management and molecular selection. In this work, we sequenced the transcriptome in L. eroeea liver tissue with a Roche 454 sequencing platform and assembled the transcriptome into 93 801 transcripts. Of them, 38 856 transcripts were successfully annotated in nt, nr, Swiss-Prot, InterPro, COG, GO and KEGG databases. Based on the annotation information, 3 165 unigenes related to growth and immunity were identified. Additionally, a total of 6 391 simple sequence repeats (SSRs) were identified from the transcriptome, among which 4 498 SSRs had enough flanking regions to design primers for polymerase chain reactions (PCR). To access the polymorphism of these markers, 30 primer pairs were randomly selected for PCR amplification and validation in 30 individuals, and 12 primer pairs (40.0%) exhibited obvious length polymorphisms. This work applied RNA-Seq to assemble and analyze a live transeriptome in L. crocea. With gene annotation and sequence information, genes related to growth and immunity were identified and massive SSR markers were developed, providing valuable genetic resources for future gene functional analysis and selective breeding ofL. eroeea.展开更多

Long noncoding RNAs (lncRNAs) play important roles in various biological regulatory processes in yeast, mammals, and plants. However, no systematic identification of lncRNAs has been reported in Gossypium arboreum. ...Long noncoding RNAs (lncRNAs) play important roles in various biological regulatory processes in yeast, mammals, and plants. However, no systematic identification of lncRNAs has been reported in Gossypium arboreum. In this study, the strand-specific RNA sequencing (ssRNA-seq) of samples from cotton fibers and leaves was performed, and lncRNAs involved in fiber initiation and elongation processes were systematically identified and analyzed. We identified 5,996 lncRNAs, of which 3,510 and 2,486 can be classified as long intergenic noncoding RNAs (lincRNAs) and natural antisense transcripts (IncNAT), respectively. LincRNAs and lncNATs are similar in many aspects, but have some differences in exon number, exon length, and transcript length. Expression analysis revealed that 51.9% of lincRNAs and 54.5% of lncNATs transcripts were preferentially expressed at one stage of fiber development, and were significantly highly expressed than protein-coding tran- scripts (21.7%). During the fiber and rapid elongation stages, rapid and dynamic changes in lncRNAs may contribute to fiber development in cotton. This work describes a set of lncRNAs that are involved in fiber development. The characterization and expression analysis of lncRNAs will facilitate future studies on their roles in fiber development in cotton.展开更多
A total of 8375 genic simple sequence repeat(SSR) loci were discovered from a unigene set assembled from 116282 transcriptomic unigenes in this study.Dinucleotide repeat motifs were the most common with a frequency ...A total of 8375 genic simple sequence repeat(SSR) loci were discovered from a unigene set assembled from 116282 transcriptomic unigenes in this study.Dinucleotide repeat motifs were the most common with a frequency of 65.11%,followed by trinucleotide(32.81%).A total of 4100 primer pairs were designed from the SSR loci.Of these,343 primer pairs(repeat length≥15 bp) were synthesized with an M13 tail and tested for stable amplification and polymorphism in four Pyrus accessions.After the preliminary test,104 polymorphic genic SSR markers were developed; dinucleotide and trinucleotide repeats represented 97.11%(101) of these.Twenty-eight polymorphic genic SSR markers were selected randomly to further validate genetic diversity among 28 Pyrus accessions.These markers displayed a high level of polymorphism.The number of alleles at these SSR loci ranged from 2 to 17,with a mean of 9.43 alleles per locus,and the polymorphism information content(PIC) values ranged from 0.26 to 0.91.The UPGMA(unweighted pair-group method with arithmetic average) cluster analysis grouped the 28 Pyrus accessions into two groups: Oriental pears and Occidental pears,which are congruent to the traditional taxonomy,demonstrating their effectiveness in analyzing Pyrus phylogenetic relationships,enriching rare Pyrus EST-SSR resources,and confirming the potential value of a pear transcriptome database for the development of new SSR markers.展开更多

As a result, we confirm a palynomorph assemblage of Cicatri- cosisporites-Protoconiferus. The pollen was primarily from gymnosperms, dominated especially by conifer pollen. Pterido- phyte spores were less common and some questionable angiosperm pollen occurred occasionally. The age of the palynomorph assemblage is dated as the late Valanginian or Hauterivian-Barrernian stage, the Early Cretaceous. The study applies the con- cept of Palynological Vegetation based on palynological spectra and the paleoecological characteristics of palynological taxa for the first time. Palynological vegetation type, climatic zone type, and humidity type are divided quantitatively for the Zhuanchengzi Bed in the Yixian Formation of western Liaoning. We then obtained the evolutionary trends. The results showed that the overall climate was warm and humid during the deposition period of the Zhuanchengzi Bed in the Yixian Formation. Palynological vegetation types are various and include coniferous forest, deciduous broadleaf forest, evergreen broad-leaved forest, grass, and shrubs. The local temperature changed from warm to much warmer and from a semi-humid to humid climate. Palynological vegetation types are always dominated by coniferous forest. The coexistence of deciduous broad-leaved forest, evergreen broad-leaved forest, shrubs, grass, and some xerophytic plants indicates vertical zonation and seasonal climate change The vertical vegetation types and the warm humid climate may imply a large geomorphological contrast in the Yixian Formation of western Liaoning.展开更多

The Tiaojishan Formation(mainly distributed in Beipiao, Chaoyang, Fuxin and Jianchang regions) is one of the most significant horizons for the Jurassic petrified wood in Liaoning Province. Previously, wood fossils of this formation were mainly reported in Beipiao and Chaoyang regions, whereas fossil wood record was merely known in Jianchang region. Here we describe new fossil wood specimens from the Tiaojishan Formation in Jianchang County, western Liaoning. Two fossil wood taxa, i.e., Protaxodioxylon jianchangense Tian et Wang sp. nov. and Xenoxylon peidense Zheng et Zhang were recognized on the basis of anatomical features. These fossil wood records add new data for understanding the fossil wood diversity, floral composition and palaeoclimate of the Tiaojishan Formation. The occurrence of Xenoxylon and Protaxodioxylon implies a cool temperate, wet and seasonal climate condition with interannual variations during the Middle to Late Jurassic transition in western Liaoning region.展开更多
