With the help of measuring ...Fluorescence spectra of native purple bacterial reaction center (RC) and bacterial pheophytin (Bphe) replaced RCs were obtained from 600 nm to 900 nm with a selective excitation at 597 nm. With the help of measuring the fluorescence from bacterial chlorophyll, bacterial pheophytin and plant pheophytin, the corresponding components in the RCs are classified for fluorescence emission. Results showed that pheophytin substitution influences the composition of fluorescence spectra. Therefore, four, three and two components were obtained from fluorescence spectra of native RC, Bphe B_replaced RC and Bphe A,B _replaced RC, respectively. Fluorescence components are well correlated to the binding of plant pheophytin. The decay of excited state of primary electron donor P in different RCs was also studied by measuring the fluorescence decay at 686.4, 674.1 and 681.1 nm, respectively. The decaying kinetics changed in different RCs, indicating that pheophytin replacement influenced the energy transduction and primary photochemical reaction in purple bacterial reaction centers.展开更多

The authors' observations also displayed a continuous arrangement process of nucleolar DNA, i.e., the extranucleolar DNA entered FC through the nucleolar organizer region (NOR) channel, then extended to the periphery of FC or to the border between FC and dense fibrillar component (DFC), and distributed along the periphery of FC. The mechanism of the proton_transfer_coupled electron transfer (PT_ET) reactions between the menaquinone Q A (MQ 1) and ubiquinone Q B (UQ 1) in the bacterial photosynthetic reaction center of Rhodopseudomona vi...The mechanism of the proton_transfer_coupled electron transfer (PT_ET) reactions between the menaquinone Q A (MQ 1) and ubiquinone Q B (UQ 1) in the bacterial photosynthetic reaction center of Rhodopseudomona viridis was studied by using the B3LYP/6_31G(d) method. The changes of standard Gibbs free energy ΔG 0 of all possible reactions followed the ET reaction (1) were calculated. The results indicated that: (1) according to the ΔG 0 values of corresponding reactions, UQ 1 could not accept two electrons from MQ - 1 continually without the coupled proton transfer reactions. Because of ΔG 0 2b 0, ΔG 0 3b 0 and ΔG 0 4b 0, the corresponding PT_ET reactions could take place along with reactions (2b), (3b) and (4b) sequentially; (2) on the gaseous condition, the first and second transferred protons (H +(1) and H +(2)) from the surrounding amino acid residues or water molecules will combine with the oxygen No.7 and oxygen No.8 of UQ 1, respectively. On the condition of protein surroundings (by SCRF model, ε =4.0), the results are converse but the energy difference between the combination of H +(1) and H +(2) with UQ - 1 is quite small. Using agreen fluorescent protein-calmodulin fusion protein as a living cell probe, we have previously found that calmodulin isassociated with the initiation and progression of cytokinesis. In this study, we further studied the effect of calmodulinon the repositioning of the centriole and subsequent cell cycle progression. When activity of calmodulin is inhibited, theregression of the centriole from the intercellular bridge to the cell center is blocked, and thus the completion of celldivision is repressed and two daughter cells are linked by longer cell bridge in perturbed cells. W7 treatment duringcytokinesis also results in unfinished cytokinesis and stopped G1 phase. These results suggest that calmodulin activity isrequired for centriole repositioning and can affect the completion of cytokinesis and cell cycle progression.展开更多
AIM: To elucidate the relationship between the frequency of core mutations and the clinical activity of hepatitis B virus (HBV)-related liver disease and to characterize the amino acid changes in the core region of HB...AIM: To elucidate the relationship between the frequency of core mutations and the clinical activity of hepatitis B virus (HBV)-related liver disease and to characterize the amino acid changes in the core region of HBV.METHODS: We studied 17 Chinese patients with chronic hepatitis B according to their clinical courses and patterns of the entire core region of HBV.RESULTS: Amino acid changes often appeared in the HBV core region of the HBV gene in patients with high values of alanine aminotransferase (ALT) or with the seroconversion from HbeAg to anti-HBe. The HBV core region with amino acid changes had high frequency sites that corresponded to HLA Ⅰ/Ⅱ restricted recognition epitopes reported by some investigators.CONCLUSION: The core amino acid changes of this study occur due to influence of host immune system. The presence of mutations in the HBV core region seems to be important for predicting the clinical activity of hepatitis B in Chinese patients.展开更多
Background The neutrophil-to-lymphocyte (N/L) ratio has been associated with poor prognosis in patients with heart failure, but it has not been compared with N-terminal pro-brain natriuretic peptide (NT-proBNP) in...Background The neutrophil-to-lymphocyte (N/L) ratio has been associated with poor prognosis in patients with heart failure, but it has not been compared with N-terminal pro-brain natriuretic peptide (NT-proBNP) in elderly patients with chronic heart failure (CHF). We sought to make this comparison. Methods A total of 1355 elderly patients with CHF were analyzed. A multivariate logistic regression model was used to analyze the variables associated with atrial fibrillation (AF). Cox regression analysis was used to assess the multivariable rela- tionship between the N/L ratio, NT-proBNP level, and subsequent major cardiovascular events (MCE). Results In the multiple logistic regression analysis, the N/L ratio was demonstrated as a risk factor for AF in elderly patients with CHF [odds ratio (OR): 1.079, 95% confi- dence interval (CI): 1.027-1.134, P = 0.003]. The median follow-up period was 18 months. In a multivariable model using tertiles of both variables, the highest tertile of the N/L ratio was significantly associated with MCE [hazard ratio (HR): 1.407, 95% CI: 1.098-1.802, P = 0.007] compared with the lowest tertile. Similarly, the highest NT-proBNP tertile was also significantly associated with MCE (HR: 1.461, 95% CI: 1.104-1.934, P- 0.008). Conclusions In elderly patients with CHF, the N/L ratio is one of the important risk factors for AF and it is an inexpensive and readily available marker with similar independent prognostic power to NT-proBNP. The risk of MCE increases 1.407-fold when the N/L ratio is elevated to the highest tertile.展开更多

METHODS: In this pilot, prospective, multicenter randomized trial, 20 ...AIM: To evaluate the efficacy of 5 compared to :tO granulocyteaphaeresis sessions in patients with active steroid-dependent ulcerative colitis. METHODS: In this pilot, prospective, multicenter randomized trial, 20 patients with moderately active steroid-dependent ulcerative colitis were randomized to 5 or 10 granulocyteaphaeresis sessions. The primary objective was clinical remission at wk 17. Secondary measures included endoscopic remission and steroid consumption.RESULTS: Nine patients were randomized to 5 granulocyteaphaeresis sessions (group 1) and 11 patients to 10 granulocyteaphaeresis sessions (group 2). At wk 17, 37.5% of patients in group 1 and 45.45% of patients in group 2 were in clinical remission. Clinical remission was accompanied by endoscopic remission in all cases. Eighty-six percent of patients achieving remission were steroid-free at wk 17. Daily steroid requirements were significantly lower in group 2. Eighty-nine per cent of patients remained in remission during a one year follow-up. One serious adverse event, not related to the study therapy, was reported. CONCLUSION: Granulocyteaphaeresis is safe and effective for the treatment of steroid-dependent ulcerative colitis. In this population, increasing the number of aphaeresis sessions is not associated with higher remission rates, but affords a significant steroid-sparing effect.展开更多

The central composit...The response surface methodology(RSM) combined with bioassays was employed to optimize the extraction process of crude fucose-containing sulphated polysaccharides(c FCSP) from Sargassum fusiforme. The central composite design(CCD) was used with four variables, five levels, and four responses. The four variables were p H value of hydrochloric acid solution, extraction temperature(℃), ratio of liquid to raw material(m L g^(-1)), and extraction time(h), respectively. Chemical and bioassay indices were used in combination as the response parameters, which included the yield of c FCSP, fucose content, proliferation rate of spleen cells, and lipopolysaccharide-induced proliferation of splenocytes. The experimental data were fitted to a second-order polynomial equation using multiple regression analysis, and examined using the appropriate statistical methods. The best extraction conditions were as follows: the p H value of hydrochloric acid solution was 3.50; the extraction temperature was 100℃; the ratio of liquid to raw material was 15.00 m L g^(-1) and the extraction time was 2.50 h. The experimental yield was close to the predicted from the model. The extract could promote spleen lymphocyte proliferation, especially the lipopolysaccharide-induced lymphocyte proliferation in vitro, which suggested that its immunomodulatory effect on B lymphocytes. Therefore, c FCSP extracted from S. fusiforme could be utilized as an immunostimulant in functional foods and pharmaceutical industry in future.展开更多
Objective:The aim of this study was to investigate the clinical,radiographic and histiopathologic features of multicentric giant cell tumor of bone.Methods:All the clinical data of twenty tumors in nine patients of mu...Objective:The aim of this study was to investigate the clinical,radiographic and histiopathologic features of multicentric giant cell tumor of bone.Methods:All the clinical data of twenty tumors in nine patients of multicentric giant cell tumor that underwent surgical treatment in our department from 1990 to 2010 were retrospectively reviewed,which included three males and six females.The patients ranged from 15 to 45 years at diagnosis,with an average age of 22.3 years.Three of the patients were younger than twenty years of age.Most tumors arose in long bones,especially around the knee.Radiographically,the tumors in long bones usually manifested as expansive lytic lesions involving the metaphysis and extending into the epiphysis.Three tumors in three patients were confined to the metaphysis,and one tumor exhibited bone-forming lesions.All tumors were treated with curettage or resection.Results:The typical "giant cell" could be found in the oncologic examination in all cases.In some areas,such as the fibrohistiocytic regions,reactive bone forming and aneurysmal bone cyst-like changes could be found.Follow-up averaged 3.5 years,ranging from 6 months to 12 years.There was a recurrence of three tumors,and one patient died of pulmonary metastasis.Conclusion:Multicentric giant cell tumor occur often in younger patients than do solitary giant cell tumor.They are frequently present around the knee,and confined to the metaphysis.Each tumor arose independently,rather than being in multiple sites of metastatic lesion that develop from a single tumor.The risk of recurrence depends on the type of surgery that is performed.展开更多
