In this work, a peptide mineralized Au cluster probe was applied as a new simplified strategy to both recognize and inhibit a single bacteria species of Staphylococcus aureus(S. aureus) simultaneously. The probes are composed of peptides and Au clusters. Moreover, the peptides specifically target S. aureus cells and the Au clusters provide fluorescent imaging and have an antibacterial effect. Xiangxi flavor vinegar(XV) is one of Hunan Province's traditional fermented vinegars.It is produced from herb,rice,and spring water with spontaneous liquid-state fermentation techniques.In this study,we investigate...Xiangxi flavor vinegar(XV) is one of Hunan Province's traditional fermented vinegars.It is produced from herb,rice,and spring water with spontaneous liquid-state fermentation techniques.In this study,we investigated the antioxidant property of XV by analyzing its antioxidant compounds,its free radical scavenging property in vitro and in vivo,and its effects on antioxidant enzyme activity and apoptosis in Caenorhabditis elegans.The results showed that XV is rich in antioxidants.In particular,ligustrazine reached 6.431 μg/ml.The in vitro 2,2-diphenyl-1-picrylhydrazyl free radical(DPPH*),hydroxyl radical(*OH),and superoxide anion radical(O2^(*-)) scavenging rates of XV were 95.85%,97.22%,and 63.33%,respectively.The reactive oxygen species(ROS) content in XV-treated C.elegans decreased significantly(P0.01) compared to the control group.Giutathione peroxidase(GSH-Px),superoxide dismutase(SOD),and catalase(CAT) activities were remarkably increased(P0.01) in C.elegans after XV treatment.In addition,XV could upreguiate CED-9 protein expression and downreguiate CED-3 protein expression in C.elegans.These results prove that XV is rich in antioxidants and scavenges radicals in vitro efficiently.XV inhibits apoptosis in C.elegans probably by scavenging ROS and increasing the activities of its antioxidant enzymes.展开更多
Chronic lymphocytic leukaemia (CLL) is a rare blood cancer that always relapses as refractory disease and eventually leads to death. To date, therapeutic options for CLL patients are scarce and there is an urgent ne...Chronic lymphocytic leukaemia (CLL) is a rare blood cancer that always relapses as refractory disease and eventually leads to death. To date, therapeutic options for CLL patients are scarce and there is an urgent need to develop novel chemotherapeutics that are both effective and safe. Gold-containing compounds induce a lethal oxidative and endoplasmic reticulum stress response in cultured and primary CLL cells via inhibition of thioredoxin reductase (TrxR). However, traditional gold-containing medicines have revealed side effects during clinical applications. Therefore, safer gold-containing drugs are needed to overcome this challenge. In this study, a novel peptide templated gold cluster Au2sSv9 was synthesized and its therapeutic effect on CLL cells was evaluated. This nanocluster could induce cell apoptosis in MEC-1 cells in a dose-dependent manner which correlated with the uptake amount of clusters in cells. As expected, increasing intracellular reactive oxidative species (ROS) in MEC-1 cells was exhibited with the increase of cluster dosage. Further analyses demonstrated the underlying mechanism that the nan- oclusters suppress the activity ofTrxR1, increase the level of intracellular ROS, destroy the mitochondrial membrane potential and finally trigger the mitochondrial apoptotic pathway in MEC-1 cells. Furthermore, the direct interaction between Au2sSv9 clusters and TrxRl was confirmed for the first time by isothermal titration calorimetry. These findings explored the preclinical efficacy and potential mech- anism of gold clusters in CLL therapy and provided a fundamental reference for the development of other novel gold-containing chemotherapeutics to treat CLL.展开更多
