目的 :探讨新辅助化疗对胃癌患者细胞免疫功能及胃癌细胞凋亡与增殖的影响。方法 :进展期胃癌患者 6 0例 ,随机分为 3组 (每组 2 0例 ) ,A组为单纯手术 ,B组为LF +手术 ,C组为LOF +手术。分别应用免疫荧光法检测T细胞亚群 ,核素释放法测... 目的 :探讨新辅助化疗对胃癌患者细胞免疫功能及胃癌细胞凋亡与增殖的影响。方法 :进展期胃癌患者 6 0例 ,随机分为 3组 (每组 2 0例 ) ,A组为单纯手术 ,B组为LF +手术 ,C组为LOF +手术。分别应用免疫荧光法检测T细胞亚群 ,核素释放法测定NK细胞活性 ,TUNEL法和免疫组织化学染色法检测胃癌细胞的凋亡 (AI)与增殖 (PI)及二者之比 (AI/PI)。结果 :B、C组与A组AI、PI差异具有极显著意义 ,P <0 0 1 ;AI/PI差异有显著意义 ,P <0 0 5。化疗前、后及术后 3组T细胞亚群的百分率差异无显著意义 ,P >0 0 5。化疗后及术后 ,B、C组较A组NK细胞活性明显降低 ,P <0 0 5 ,P <0 0 1。结论 :新辅助化疗可明显促进胃癌细胞凋亡并抑制其增殖 ,对手术期患者的T细胞亚群免疫功能无明显影响 。 展开更多

目的:探讨降钙素对兔膝骨性关节炎关节软骨细胞凋亡及NO含量的影响。方法:将36只健康大耳白兔随机分为空白对照组(A组)、模型组(B组)和CT治疗组(C组),B、C两组采用改良Hulth法复制膝骨性关节炎模型,A组除切开皮肤和关节腔外,不做其他处... 目的:探讨降钙素对兔膝骨性关节炎关节软骨细胞凋亡及NO含量的影响。方法:将36只健康大耳白兔随机分为空白对照组(A组)、模型组(B组)和CT治疗组(C组),B、C两组采用改良Hulth法复制膝骨性关节炎模型,A组除切开皮肤和关节腔外,不做其他处理,从术后第6周开始C组每天皮下注射CT 5u/kg,A、B组给与等量生理盐水皮下注射,连续注射20d后处死动物。观察降钙素对兔膝骨性关节炎关节软骨病理形态、关节软骨细胞凋亡情况及NO含量的影响。结果:与A组相比较,B组JP和Mankin’s评分明显升高,关节软骨损伤明显加重,血清及关节液NO含量和软骨细胞凋亡百分数均明显升高;与B组比较,C组JP评分和Mankin’s评分明显降低,软骨损伤明显减轻,血清及关节液NO含量和软骨细胞凋亡百分数均明显降低。结论:CT可能通过降低NO含量而减轻KOA的软骨损伤,对KOA起治疗作用。 展开更多

Taxol production of Taxus chinensis (Pilger) Rehd. var. mairei (Lemee et Lévl.) Cheng et L. K. Fu induced by oligosaccharide from Fusarium oxysporum f. vasinfectum (Atkinson) Snyder et Hansen was studied in suspension cultures, and it was found that oligosaccharide triggered cell apoptosis. Under transmission electron microscope the following morphological changes were observed: cell shrinkage, condensation of cytoplasm, nuclear fragmentation, and the increase of high electron density bodies in vacuole in great quantity. In oligosaccharide_treated cells, agarose gel electrophoresis revealed that DNA was digested into oligonucleosomal fragments that were times of 200 bp appearing as DNA ladders. Control cells were in normal physiological state, they were intact, abundant in organelle and with integral nucleus DNA, and the rate of taxol biosynthesis in these cells was very low. After the oligosaccharide to the culture system, the defense system of cells was elicited and the secondary metabolism was strengthened, i.e. phenolics were accumulated in the medium, the activity of polyphenol oxidase (PPO) was increased quickly and secondary wall of cells was thickened. The activity of L _phenylalanine ammonia lyase (PAL), the critical enzyme of the phenylpropanoid pathway, was increased promptly 1 h after elicitation. The rate of taxol production was improved sharply and the maximal taxol concentration at 72 h was six times that of control. Appearance of cell apoptosis was accompanied with the highest concentration of taxol in suspension cultures. 展开更多

目的探讨原发性肝癌组织中三种凋亡相关基因:XIAP、Smac及Caspases-3的表达及意义。方法应用免疫组化方法检测67例原发性肝癌组织中XIAP、Smac及Caspases-3的表达情况。结果XIAP、Smac及Caspases-3在正常肝组织中的阳性率分别为(5.9&... 目的探讨原发性肝癌组织中三种凋亡相关基因:XIAP、Smac及Caspases-3的表达及意义。方法应用免疫组化方法检测67例原发性肝癌组织中XIAP、Smac及Caspases-3的表达情况。结果XIAP、Smac及Caspases-3在正常肝组织中的阳性率分别为(5.9±1.0)%、(90.1±0.8)%和(87.7±1.1)%,在肝癌组织中的阳性率分别为(89.7±0.7)%、(53.21±1.3)%和(49.8±0.9)%,三者在不同性质组织问差异均有统计学意义(t=7.78,5.37,6.01,P〈0.05);肝癌组织中XIAP与Caspases-3的表达呈负相关(r=-0.69,P〈0.05),Smac与Caspases-3的表达呈正相关(r=0.67,P〈0.05)。结论XIAP、Smac通过影响Caspases-3的表达拮抗细胞凋亡过程,参与原发性肝癌的发生、发展。 展开更多

OBJECTIVE: To investigate the effects of Tangnaikang(TNK), a mixture of five herbal plant extracts,on inflammation-mediated insulin resistance andβ-cell apoptosis in SHR.Cg-Leprcp/NDmcr(SHR-cp)rats.METHODS: Seven-week-old SHR-cp rats were randomly divided into a control(CON) group and a TNK(3.24 g/kg) group. Wistar-Kyoto rats at the same age were used as the normal control group.After 7 weeks of continuous intragastric administration of TNK, the glucose metabolic status and insulin sensitivity of the rats were evaluated by assessing fasting serum glucose(FBG), the oral glucose tolerance test(OGTT), fasting serum insulin(FINS),and the insulin sensitivity index(ISI). Serum tumor necrosis factor-α(TNF-α), interleukin-6(IL-6), C-reactive protein(CRP) and adiponectin were measured via enzyme-linked immunosorbent assays.Macrophage infiltration and apoptosis in adipose tissues were detected through F4/80 immunohistochemistry and the terminal deoxynucleotidyl transferase d UTP nick end labeling(TUNEL) assay. Islet morphology and β-cell apoptosis were investigated using double immunofluorescence staining and the TUNEL assay. The expression of cytokine genes in adipose tissue, the liver, and the pancreas was detected in real-time polymerase chain reaction assays. The expression and phosphorylation levels of insulin signaling-, inflammation-, and β-cell survival-related proteins in the liver and pancreas of SHR-cp rats were detected by western blotting.RESULTS: TNK(3.24 g/kg) treatment significantly decreased body weight, FBG and FINS; improved impaired glucose tolerance; increased the ISI; reduced serum levels of TNF-α, CRP and IL-6; and increased serum adiponectin. The m RNA expression of inflammatory markers was markedly reduced in the liver, pancreas, and adipose tissue. F4/80-and TUNEL-positive cells in adipose tissues were decreased, as was the number of TUNEL-positiveβ-cells. The phosphorylation of c-Jun N-terminal kinase and that of insulin receptor substrate-1 at serines 307 and 1101 was significantly decreased.In the pancreas, the expression of nuclear factor kappa-light chain-enhancer of activated B cells-p65 was significantly decreased, and phosphoinositide3-kinase and IRS-2 were significantly increased.CONCLUSION: TNK was able to improve insulin resistance and β-cell apoptosis in SHR-cp rats, which might be associated with its ability to relieve the overall and local metabolic inflammatory responses observed in SHR-cp rats. 展开更多
