METHODS: Male Sprague Dawley rats were assigned to 3 Groups: Group A, sham operated rats as controls (n = 7); Group B, acute pancreatitis induced by ductal injection with 5% sodium cholate at a volume of 1.0 mL/kg without any other treatment; Group C, after the pancreatitis was induced as in Group B, the rats were injected intravenously with 5-FU 40 mg/kg. The animals in Groups B and C were killed at 2, 6 and 24 h after operation (n = 7), and blood samples were taken for measurement of tumor necrosis factor-α (TNF-α), interleukin-1 (IL-1), interleukin-6 (IL-6) (by bioassay), and interleukin-10 (IL-10), transforming growth factor-β (TGF-β) (by ELISA). The wet weight of pancreatic tissue, serum amylase levels and white blood cells were also measured. RESULTS: Four rats in Group B and one in Group C died after pancreatitis was induced. Both pro-inflammatory cytokines (TNF-α, IL-1, IL-6) at the 2 and 6 h period and the anti-inflammatory cytokines (IL-10, TGF-β) at 24 h increased significantly (P 〈 0.05) in rats of Group B. After treatment with 5-FU, TNF-α, IL-1, and IL-6 in serum of rats of Group C were inhibited at 2 and 6 h after operation (P 〈 0.05), and IL-IO, TGF-13 were inhibited at 24 h compared to Group B (P 〈 0.05). Obvious improvements in the severity of the acute pancreatitis, including the amylase levels, wet weight of pancreatic tissue and neutrophil counts, were also observed after treatment with 5-FU. CONCLUSION: 5-FU is an anti-metabolic and immunosuppressive agent which can minimize the abnormal immune o/tokine response and relieve the pathophysiological disorders associated with experimental acute pancreatitis.展开更多

METHODS: Male Bama pigs (n = 14) treated with ketamine hydrochloride and Sumianxin Ⅱ as induc- tion drugs followed by inhalation anesthesia with 2% isoflurane, underwent 70% hepatotectomy with loss of bleeding less than 50 mL, and with hepatic pedicle occlusion for 20 min, were divided into two groups: Hydrogen-group (n = 7), the pigs with inhalation of 2% hydrogen by the tracheal intubation during major hepa- totectomy; Contrast-group (n= 7), underwent 70% hepatotectomy without inhalation of hydrogen. Hemo- dynamic changes and plasma concentrations of alanine aminotransferase (ALT), aspartate aminotransferase (AST), hyaluronic acid (HA), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and malondialdehyde (MDA) in liver tissue were measured at pre-operation, post-hepatotectomy (PH) 1 h and 3 h. The apoptosis and proliferating cell nuclear antigen (PCNA) expres- sion in liver remnant were evaluated at PH 3 h. Then we compared the two groups by these marks to evalu- ate the effect of the hydrogen in the liver injury during major hepatotectomy with the Pringle Maneuver in the swine. RESULTS: There were no significant differences in body weight, blood loss and removal liver weight be- tween the two groups. There was no significant differ- ence in changes of portal vein pressure between two groups at pre-operation, PH 30 min, but in hydrogen gas treated-group it slightly decrease and lower than its in Contrast-group at PH 3 h, although there were no significant difference (P = 0.655). ALT and AST in Hydrogen-group was significantly lower comparing to Contrast-group (P = 0.036, P = 0.011, vs P = 0.032, P = 0.013) at PH 1 h and 3 h, although the two groups all increased. The MDA level increased between the two group at PH i h and 3 h. In the hydrogen gas treated- group, the MDA level was not significantly significant at pre-operation and significantly low at PH 1 h and 3 h comparing to Contrast-group (P = 0.0005, P = 0.0004). In Hydrogen-group, the HA level was also significantly low to Contrast-group (P = 0.0005, P = 0.0005) al- though the two groups all increased at PH 1 h and 3 h. The expression of cluster of differentiation molecule 31 molecules Hydrogen-group was low to Contrast-group. However, PCNA index (%) was not statistically signifi- cant between the two groups (P = 0.802). Micropho- tometric evaluation of apoptotic index (AI) in terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling-stained tissue after hepatotectomy for 3h, the AI% level in the hydrogen was significantly low to Contrast-group (P = 0.012). There were no significant difference between Hydrogen-group and Contrast- group at pre-operation (P = 0.653, P = 0.423), but after massive hepatotectomy, the TNF-α and IL-6 levels increase, and its in Hydrogen-group was significantly low compared with Contrast-group (P = 0.022, P = 0.013, vs P = 0.016, P= 0.012), respectively. Hydro- gen-gas inhalation reduce levels of these markers and relieved morphological liver injury and apoptosis.CONCLUSION: H2 gas attenuates markedly ischemia and portal hyperperfusion injury in pigs with massive hepatotectomy, possibly by the reduction of inflamma- tion and oxidative stress, maybe a potential agent for treatment in clinic.展开更多

Methods: Thirty-two male SD rats were randomly and evenly divided into four groups: normal control group; UC control group; "Zusanli" (ST 36, EA) group; non-acupoint group. A solution containing ethanol and 2,4,6-trinitrobenzenesulfonate was instilled into the distal colon in the rat (at a dose of 100 mg/kg) to set up UC model. Non-anesthetized rats of EA group were stimulated at ST-36 by electroacupuncture once a day, while those of non-acupoint group were done at the site 0.5 cm beside ST-36. After 10 days’ treatment, all rats were sacrificed simultaneously. Colon mucosal inflammation and damage were assessed by measuring colon mass, morphologic damage score, colonic MPO activity, contents of serum interleukin-6 (IL-6), interleukin-8 (IL-8) and tumor necrosis factor-alpha (TNF-α). Morphologic damage score was examined under stereomicroscope. Colonic MPO was measured by spectrophotometric method. Serum TNF-α,IL-6 and IL-8 were determined by radioimmunoassay (RIA). Results: Both ratio of colonic mass/body mass (mC/mB) and activity of colonic MPO (μkat/g) in rats with UC were markedly increased (P<0.01 vs normal control group). Concentrations of serum TNF-α, IL-6 and IL-8 (ng/L) in rats of UC control group were significantly elevated (P<0.01 vs normal control group). In EA group, mC/mB and activity of MPO were decreased markedly compared with those of UC control group (P<0.05). Serum levels of TNF-α, IL-6 and IL-8 of ST-36 group were decreased by EA of ST-36 (P<0.01; P<0.05; P<0.05 vs UC control group). In comparison with UC control group, no significant changes were observed in the non-acupoint group. Furthermore, there were significant correlations among these parameters. Conclusion: In UC rats, serum TNF-α, IL-6 and IL-8 levels and MPO activity all increase significantly, indicating that these cytokines may cause inflammatory and immune response. EA stimulation induced downregulation on serum TNF-α, IL-6 and IL-8 levels and MPO activity may be responsible for its therapeutic effect in improving UC.展开更多

Methods: A total of 51 male Wistar rats, weighing 280 340 g , were anesthetized with chloral hydrate ( 400 mg/kg body weight) through intraperitoneal injection and fixed on a stereotaxic instrument. Severe brain injury was created in 16 rats (severe injury group) and moderate brain injury in 18 rats (moderate injury group) by a fluid percussion model, and cytokine levels of IL 1β, TNFα and IL 6 were measured with biological assay. And sham operation was made on the other 17 rats (control group). Results: In the control group, the levels of IL 1β, TNFα and IL 6 were hardly detected in the cortex of the rats, but in the ipsilateral cortex of the rats in both injury groups, they increased obviously at 8 hours after injury. The increasing degree of these cytokines had no significant difference between the two injury groups. The levels of IL 6 in the plasma of all the rats increased slightly, whereas the levels of IL 1β and TNFα were undetectable. Conclusions: The increase of IL 1β, TNFα and IL 6 levels is closely related to brain injury. The increased cytokine levels in the central nervous system are not parallel to those in the peripheral blood. It suggests that inflammatory cytokines play important roles in the secondary neural damage after brain injury.展开更多

Methods: SD rats were randomized into a normal group, a model group, a herbs-partitioned moxibustion group, and a sulfasalazine (SASP) group. The rats in the herbs-partitioned group were treated with herbs-partitioned moxibustion at Tianshu (ST 25) and Qihai (CV 6), and those in the SASP group were treated by intragastric administration. After interventions, HE staining and light microscope were adopted in observing the histopathological changes of rat's colon, and immunohistochemical methods for detecting the expressions of KGF-1, KGF-2, and IL-6 proteins in rat's colon. Results: Compared with the model group, the rats' colons in the herbs-partitioned moxibustion group and the SASP group were histopathologically improved; compared with the normal group, the expressions of KGF-1, KGF-2, and IL-6 proteins increased significantly in the model group (P〈0.05); after intervened by herbs-partitioned moxibustion and SASP respectively, the expressions of KGF-1, KGF-2, and IL-6 proteins in rat's colon were decreased markedly (P〈0.05). Conclusion: Both herbs-partitioned moxibustion and SASP can down-regulate the expressions of KGF-1, KGF-2, and IL-6 proteins in rat's colon, which might be one of the mechanisms of herbs-partitioned moxibustion and SASP in treating UC.展开更多

Methods:The study included 33 patients (25 females and 8 males; mean age 46 years) with body mass index (BMI) values of〉40 kg/m2. Evaluations of CRP, IL-6, TNF-α, lipid profile, HOMA-IR, and fasting glucose were carried out before (baseline data) and three weeks after the treatment. The control group consisted of 20 healthy volunteers (15 females and 5 males) with a mean age of 39 years and BMI values of≤24.9 kg/m2. Results:In the blood of patients with morbid obesity we found significantly elevated levels of CRP, TNF-α, triglycerides, HOMA-IR and fasting glucose, but a de-creased level of high density lipoprotein (HDL)-cholesterol, compared with the healthy individuals. The treatment resulted in about a 9.4%reduction in body weight from 122.5 to 111.0 kg and a significant decrease in the concen-tration of CRP, but no change in TNF-αor IL-6. HOMA-IR was significantly reduced. Conclusions:The decrease in CRP level without changes in TNF-α or IL-6 concentrations after the low-calorie diet and balneological treatment, suggests that an essential amount of adipose tissue must be removed before proper adipocyte function is restored. The decrease in HOMA-IR indicates an improvement in insulin sensitivity, which is beneficial in obese patients.展开更多

Methods Thirty-nine cases undergoing LC were randomly divided into a CAGA group and a GA group. The CAGA group was treated with electroacupuncture at Hegu (合谷- LI 4), Neiguan (内关PC 6), Zusanli (足三里ST 36) and Yanglingquan (阳陵泉 GB 34) for 15-30 minutes followed by the general anesthesia, and the continuous electroacupuncture stimulation was given till the operation finished. The GA group was treated with simple general anesthesia. Changes of T cell subsets, tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6) were observed before anesthesia induction, and 2 hours, 1st and 3 rd day after operation, and the adverse reaction after operation was recorded. Results At 2 hours after operation, the percentages of CD3 and CDs in both groups were significantly lower than those before anesthesia induction (all P〈0.05), and the percentage of CD4 in the GA group decreased significantly (P〈0.05), while the percentage of CD: did not significantly change and CD4/CD8 increased significantly in the CAGA group (P〈0.05). At 3 days after operation, the level of TNF-α in the ACGA group decreased significantly as compared with that before anesthesia induction (P〈0.05). The cases with nausea after operation in the CAGA group were significantly less than those in the GA group (P〈0.05). Conclusion Acupuncture combined with general anesthesia has a little effect on immune function in patients of LC with less adverse reactions.展开更多

文摘Aim: To investigate the regulatory effects of electroacupuncture (EA) at "Zusanli" (ST 36) on proinflammatory cytokines and myeloperoxidase (MPO) activity in rats with ulcerative colitis (UC), and further elucidate the underlying mechanism of EA on UC. EA stimulation induced downregulation on serum TNF-α, IL-6 and IL-8 levels and MPO activity may be responsible for its therapeutic effect in improving UC.

Conclusions: The increase of IL 1β, TNFα and IL 6 levels is closely related to brain injury. The increased cytokine levels in the central nervous system are not parallel to those in the peripheral blood. It suggests that inflammatory cytokines play important roles in the secondary neural damage after brain injury.

Conclusion: Both herbs-partitioned moxibustion and SASP can down-regulate the expressions of KGF-1, KGF-2, and IL-6 proteins in rat's colon, which might be one of the mechanisms of herbs-partitioned moxibustion and SASP in treating UC.
文摘Objective:In this study,we investigated the changes in peripheral blood inflammatory factors and intestinal flora in acquired immune deficiency syndrome(AIDS)and human immunodeficiency virus(HIV)-positive individuals(AIDS/HIV patients),and explored the relationships among intestinal flora, peripheral blood inflammatory factors, and CD4^+T lymphocytes. Methods:Thirty blood and stool samples from an AIDS group and a control group were collected. The levels of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)were determined by enzyme-linked immunosorbent assay(ELISA),and the number of CD4^+T lymphocytes by a FACSCount automated instrument.Quantitative real-time polymerase chain reaction(qRT-PCR)was used to determine the messenger RNA(mRNA)levels of Bifidobacterium,Lactobacillus,Escherichia coli,Enterococcus faecalis, and Enterococcus faecium.Correlations among intestinal flora, inflammatory factor levels, and CD4^+T lymphocyte values were evaluated using the Spearman correlation coefficient. Results:The levels of TNF-αand IL-6 in the AIDS group were higher than those in the control group, while the number of CD4^+T lymphocytes was lower. The amounts of Bifidobacterium and Lactobacillus in the AIDS group were significantly lower than those in control group, while the amounts of E.coli,E.faecalis,and E.faecium were much higher. The amounts of Bifidobacterium and Lactobacillus were negatively correlated with the content of TNF-αand IL-6 and the CD4^+T lymphocyte count, while those correlations were reversed for E.coli,E.faecalis,and E.faecium.Conclusions:The intestinal microbiota of AIDS/HIV patients were disordered, and there was a correlation between the amount of intestinal flora and the number of CD4^+T lymphocytes and the levels of TNF-αand IL-6.

文摘Objective:The aim of this study was to evaluate the concentrations of C-reactive protein (CRP), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and the degree of homeostasis model assessment-insulin resistance (HOMA-IR) in patients with morbid obesity exposed to a three-week low-calorie diet and balneotherapy. The decrease in HOMA-IR indicates an improvement in insulin sensitivity, which is beneficial in obese patients.

文摘Objective To compare effects of combined acupuncture and general anesthesia (CAGA) and general anesthesia (GA) on immune function in patients of laparoscopic cholecystectomy (LC) in peri-operative period. Conclusion Acupuncture combined with general anesthesia has a little effect on immune function in patients of LC with less adverse reactions.

文摘Postherpetic neuralgia(PHN) is a severe sequela of herpes zoster(HZ).Until now, only age and pain severity were considered predisposing factors for the development of PHN.We evaluated 49 patients with acute phase HZ,10 of whom developed PHN(Group A) and 39 of whom did not develop PHN(Group B).Twenty-five healthy volunteers similar in age and gender distribution to the study group were recruited as controls(Group C).Numbers of serum CD3+(pan-T lymphocytes),CD4+(helper/inducer),and CD8+(suppressor/cytotoxic) lymphocytes were decreased significantly in Groups A and B relative to the control group, but there were no statistical differences between Groups A and B.Interleukin(IL)-1β,IL-6,tumor necrosis factor(TNF)-α,IL-8,and IL-10 were significantly elevated in Groups A and B relative to Group C.IL-6 was significantly higher in Group A than in Group B,and was significantly positively correlated with pain severity scored on a visual analog scale. Therefore,we suggest that the inflammatory response, especially that of IL-6,in the acute phase of HZ may be associated with hyperalgesia and the development of PHN.
