目的:为了进一步了解肺癌细胞生物学行为,评价肺小标本ICM—DNA含量检测对肺癌的诊断价值。方法:选取纤维支气管镜活检肺小标本 95例,其中肺癌组 64例[包括鳞状上皮细胞癌(简称鳞癌)45例,腺癌 9例,小细胞未分化... 目的:为了进一步了解肺癌细胞生物学行为,评价肺小标本ICM—DNA含量检测对肺癌的诊断价值。方法:选取纤维支气管镜活检肺小标本 95例,其中肺癌组 64例[包括鳞状上皮细胞癌(简称鳞癌)45例,腺癌 9例,小细胞未分化癌10例],肺良性病变(炎症)组31例。将肺小标本石蜡包块以5μm连续切片,分别作HE染色和Feulgen 染色,后者进而用国际标准化 CAS-200型 图像细胞仪(ICM)检测 DNA含量。分析指标包括 DNA质量、DNA指数(DI)、增殖指数(PI)、S期百分比、核面积、直方图类型及倍体状况等。结果:肺癌组细胞核DNA质量、DI、PI、S%和核面积等指标均明显高于肺炎症组(P<0.05或P<0.01);肺癌组DNA直方图类型主要为Ⅲ型和Ⅵ型(占79.7%),未见Ⅰ型;而肺炎症组相反,无1例为Ⅲ型和Ⅵ型,多呈Ⅰ型改变(90.3%),两组之间有显著差异(P<0.01);两组的细胞核 DNA 倍体状况存在极显著差异(P<0.01),肺癌组多为异倍体,异倍体检出率达96.9%,而肺炎症组均为二倍体。以异倍体为标准,结合S期百分比,诊断肺癌的敏感性为96.7%,特异性达100%。结论:应用ICM检测纤维支气管? 展开更多

Application of Image Fusion Methods to Cell Imaging Processing

Aim To fuse the fluorescence image and transmission image of a cell into a single image containing more information than any of the individual image. Methods Image fusion technology was applied to biological cell imaging processing. It could match the images and improve the confidence and spatial resolution of the images. Using two algorithms, double thresholds algorithm and denoising algorithm based on wavelet transform,the fluorescence image and transmission image of a Cell were merged into a composite image. Results and Conclusion The position of fluorescence and the structure of cell can be displyed in the composite image. The signal-to-noise ratio of the exultant image is improved to a large extent. The algorithms are not only useful to investigate the fluorescence and transmission images, but also suitable to observing two or more fluoascent label proes in a single cell. 展开更多

Automatic interpretation of the images of colon cell biopsies requires automatic segmentation of these cells in the image obtained. The active contour method for image segmentation is a well known method for automatic detection of the cell contour. However, the application of this method on colon cell images was not effective. In this paper, the authors have proposed a new technique to reduce the analysis time needed to detect cells in a given image. This technique is based on the active contour method but now using a progressive division of the dimensions of the image to achieve convergence. The model proposed succeeded in detecting cells whose boundaries are not necessarily defined by a gradient. The initial curve can be anywhere in the image, and interior contours can be automatically detected. The developed algorithm was successfully applied on textured multispectral images of three types of cells, including benign hyperplasia (BH), intraepithelial neoplasia (IN), and carcinoma (Ca) cells. 展开更多

Objective To evaluate the effects endothelial progenitor cells (EPCs) survival in vitro of serum deprivation (SD) and hypoxia on the bone marrow Methods Rat bone marrow EPCs were exposed for 48 h to 02 deprivation, serum deprivation ( SD ) , and prolonged ( 120 h) hypoxia concomitant with serum deprivation. Cell death was assessed by Live/Dead staining and image analysis. Reaulta The EPCs death rate seemed not affected by 48 h hypoxia ( P 〉 0. 05 ), but affected by SD ( P 〈 0. 01 ). Prolonged hypoxia concomitant with SD resulted in the death of nearly all EPCs from 72 h, but this rate was remarkably reduced when with 20% fetal bovine serum( P 〈 0. 01 ). Conclusion Our findings indicate that EPCs are sensitive to hypoxia/SD stimuli, while serum may be the more important factor for EPCs survival. 展开更多
