目的探讨造血因子重组人粒细胞集落刺激因子(recombinant human granulocyte colony stimulating factor;rhG-CSF)、重组人白细胞介素11(recombinant human interleukin 11;rhIL11)和白细胞介素2(Interleukin 2;IL2)组合方案对急性放射...目的探讨造血因子重组人粒细胞集落刺激因子(recombinant human granulocyte colony stimulating factor;rhG-CSF)、重组人白细胞介素11(recombinant human interleukin 11;rhIL11)和白细胞介素2(Interleukin 2;IL2)组合方案对急性放射性消化系统损伤的治疗作用。方法将60Coγ射线照射损伤后的比格狗分为单纯照射损伤组、对症治疗组和细胞因子综合治疗组,通过对各主要消化器官的组织病理学观察,分析3组的治疗效果。结果单纯照射组动物在15d内全部死亡,而经治疗的动物存活时间均超过45d,从病理切片上观察发现,细胞因子(rhG-CSF、rhIL11和IL2)综合治疗组动物的胃、小肠、大肠、肝脏及胰腺的修复较对症治疗组明显。结论细胞因子综合治疗方案对放射性消化系统损伤有明显的疗效。展开更多

METHODS: mRNA levels o...AIM: To identify the role of herbal compound 861 (Cpd 861) in the regulation of mRNA expression of collagen synthesis- and degradation-related genes in human hepatic stellate cells (HSCs). METHODS: mRNA levels of collagen types I and III, matrix metalloproteinase 1 (MMP-1), matrix metalloproteinase 2 (MMP-2), membrane type-1 matrix metalloproteinase (MT1-MMP), tissue inhibitor of metalloproteinase 1 (TIMP-1), and transforming growth factor β1 (TGF-β1) in cultured-activated HSCs treated with Cpd 861 or interferon-γ, (IFN-γ,) were determined by real-time PCR. RESULTS: Both Cpd 861 and IFN-γ reduced the mRNA levels of collagen type Ⅲ, MMP-2 and TGF-β1. Moreover, Cpd 861 significantly enhanced the MMP-1 mRNA levels while down-regulated the TIMP-1 mRNA expression, increasing the ratio of MMP-1 to TIMP-1 to (6.3 + 0.3)- fold compared to the control group. CONCLUSION: The anti-fibrosis function of Cpd 861 may be mediated by both decreased interstitial collagen sythesis by inhibiting the transcription of collagen type Ⅲ and TGF-β1 and increased degradation of these collagens by up-regulating MMP-1 and down-regulating TIMP-1 mRNA levels.展开更多

METHODS: Collect and analyze the 23 sample of umbilical blood. RESULTS:(1) The combination of SCF+Flk2/Flt3 ligand and(FL)±TPO can amplify CD34,+,CD34+Thy 1+,CD34+CD33+and LTC IC in umbilical blood effectively and rapidly.(2) After add solubility type (sIL 6R)of IL 6 and IL 6 receptor into this combination, the stem cell or progenitor proliferated prominently;When add IL 6 alone(without sIL 6R), the content of CD34+cells and LTC IC didn’t increased obviously in early stage, but the amplification of CFU Mix and BFU E was not prominent as CFU GM. (3)Detect apoptosis rate of cells by FITC Annexin Ⅴlabeled by membranous change in early stage of cellular apoptosis. After add Flt3L and /or IL 6+sIL 6R, the Annexin Ⅴpositive cells decreased from 15.2%~19.1%to 2.8%~3.5%. The present study was undertaken to observe the effect of hypoxia on DNA synthesis and c-myc gene expression between newbor...The neonate is particularly susceptible to the development of hypoxic pulmonary hypertension. The present study was undertaken to observe the effect of hypoxia on DNA synthesis and c-myc gene expression between newborn calf and adult bovine PASMC in vilro. DNA synthesis measured by 3H- TdR incorporation was increased after hypoxic challenge for 24h. Hypoxia enhanced the increment in 3H-TdR incorporation induced by EGF. Northern blot analysis revealed that PASMC cultured in both normoxia and hypoxia expressed c- myc gene transcript of 2. 2Kb ,but there is a higher 2. 2Kb mRNA expression in hypoxic PASMC than that in normoxia. We speculate that newborn calf PASMC exhibited potential response to hypoxia than adult,which was augmented by EGF. Enhanced c-myc gene expression may lead to a great understanding of the mechanism of PASMC growth in the development of pulmonary hypertension.展开更多
