However, short-term variations (e.g. at a tidal timescale) of these organisms and their regulating factors remain unclear. We determined the time-series of fluctuations of picoplankton and viruses with tide simultaneously in flow cytometry in the Changjiang estuary during a cruise in June 2006, in which a tidal model based rectangle equation was applied. The results indicate that high cell abundances of picoplankton and viruses occurred during flood tide and low cell abundances during ebb tide. The period of the surface cell abundance variations was about 13 h, suggesting the surface cell abundances in the Changjiang estuary were largely regulated by tide. However, cell abundances in middle and bottom waters varied in different periods due to influences of tidal induced physical forces such as resuspension and stratification. Therefore, tidal action is an important factor for the diel variations of picoplankton and viruses in the Changjiang estuary.展开更多

METHODSUsing CCl<sub>4</sub> administration for eight weeks, we induced hepatic fibrosis in wild-type C57BL/6 mice and in mice with deletion of the cytoplasmic signalling domain of TF (TF<sup>§CT/§CT</sup>), deletion of PAR-2 (PAR-2<sup>-/-</sup>) and combined deletion of TF signalling domain and PAR-2 (TF<sup>§CT/§CT</sup>/PAR-2<sup>-/-</sup>). Hepatic fibrosis area was assessed by quantitative imaging of picrosirius red staining. Hepatic collagen content was assessed by hydroxyproline levels. Hepatic stellate cells (αSMA positive) and hepatic macrophages (CD68 positive) were identified by immunohistochemistry. Hepatic gene expression was determined by PCR and liver TGFβ1 content by ELISA. RESULTSCCl<sub>4</sub> treated mice with deletion of the PAR-2 gene (PAR-2<sup>-/-</sup>) and the cytoplasmic domain of TF (TF<sup>§CT/§CT</sup>) developed significantly less hepatic fibrosis, characterised by reduced liver fibrosis area and hydroxyproline content, compared to control wildtype mice treated with CCl<sub>4</sub>. The observed reduction in histological fibrosis was accompanied by a significant decrease in the hepatic content of TGFβ, the prototypic fibrogenic cytokine, as well as fewer activated hepatic stellate cells and hepatic macrophages. Deletion of the TF cytoplasmic signalling domain reduced hepatic fibrosis to levels similar to those observed in mice lacking PAR-2 signalling but combined deletion provided no added protection against fibrosis indicating a lack of mutual modulating effects that have been observed in other contexts such as angiogenic responses. CONCLUSIONTissue factor cytoplasmic domain is involved in TF-PAR-2 signalling initiating hepatic fibrosis and is a potential therapeutic target, as its deletion would not impact coagulation.展开更多

Methods: In vitro, the drug resistanc...Objective: The aim of this study was to explore the correlation between epithelial to mesenchymal transition (EMT) and chemoresistance of non-small-cell lung cancer (NSCLC). Methods: In vitro, the drug resistance index of cisplatin resistant lung adenocarcinoma cell line (A549/DDP) was detected by CCK-8 assay; the morphological change between A549/ DDP cells and lung adenocarcinoma cells (A549) was observed by phase contrast microscope; expression of EMT markers (including E-cadherin and vimentin) and resistance protein, excision repair cross-complementing 1 (ERCC1) was detected by immunocytochemistry. The expression of E-cadherin, vimentin and ERCC1 was investigated by immunohistochemistry in 120 cases of NSCLC, half of that were treated with pre-operative neoadjuvant chemotherapy (neoadjuvant chemotherapy group), and the other underwent surgery alone (simple surgery group). Results: There was a significant difference between the ICso (half maximal inhibitory concentration) of A549/DDP cells (5.20) and A549 cells (1.88) (P 〈 0.05), and the drug resistance index of A549/DDP cells was 2.77. Compared with A549 cells, A549/DDP cells increased expression of ERCC1 (P 〈 0.05). Moreover, A549/DDP cells showed morphological and phenotypic changes consistent with EMT: with spindle-shaped morphology, and decreased expression of E-cadherin and increased expression of vimentin. Immunohistochemistry showed significant positive correlation between the expression of ERCCI and vimentin (r = 0.496, 0.332, P 〈 0.05), and significant negative correlation between the ERCCI and E-cadherin (r = -0.403, -0.295, P 〈 0.05) in neoadjuvant chemotherapy group and simple surgery group. In addition, compared with simple surgery group, the expression of ERCC1 (P = 0.003) and vimentin (P = 0.004) was significantly increased, and the expression of E-cadherin was decreased in neoadjuvant chemotherapy group (P = 0.032). Cenclusion: A549/DDP cells acquired cisplatin-resistance and occurred EMT simultaneously; the phenomenon of chemoresistance and EMT was caused more easily in neoadjuvant chemotherapy group. As such, we further confirmed the close correlation between chemoresistance and EMT of NSCLC, and provided theoretical basis for the targeting therapy with EMT regulatory factor for chemoresistant NSCLC patients.展开更多

The high speed cell converter with the switching frequency over MHz behaves as an ideal transformer, and this behavior solves the voltage imbalance issue in the multicellular converter topology. The analysis of the unregulated cell converter is conducted by using the state space averaging method, and the operation condition for the ideal transformer is specified. The behavior of the multicellular DCX using the high speed cell converters has been also analyzed, and the voltage imbalance issue among cell converters is discussed quantitatively. A prototype of a 19.2 kW 384 V-384 V multicellular DCX using sixty-four unregulated cell converters is fabricated and the validity of the analyses is verified.展开更多
OBJECTIVE: To examine 10 cases with primary cutaneous CD30-positive anaplastic large cell lymphoma (ALCL), analyze their clinical manifestations and pathological and immunohistochemical features, and improve early dia...OBJECTIVE: To examine 10 cases with primary cutaneous CD30-positive anaplastic large cell lymphoma (ALCL), analyze their clinical manifestations and pathological and immunohistochemical features, and improve early diagnosis of this disease. METHODS: We studied the morphological characteristics of primary cutaneous CD30-positive ALCL using histopathological methods. Leukocyte common antigen (LCA), CD20, CD30, CD45RO, CD68, epithelial membrane antigen (EMA), cytokeratin (CK) and HMB45 antibodies were used to determine the expression of their respective antigens from routine paraffin samples of the patients. RESULTS: Ten patients (7 men and 3 women, aged 31 to 84 years) complained of subcutaneous masses or papular eruptions over their lower trunks and extremities. Histopathologically, the lesions were composed of numerous large round or oval pleomorphic cells. The cytoplasm was usually abundant, amphophilic or basophilic, and finely vacuolated. Nuclei were commonly eccentrically localized and lobated or horseshoed in shape, and multinucleated giant cells and Reed-Sternberg-like cells were seen. Nucleoli were generally multiple and large. Of the 10 patients, tumor cells displayed positive antigen expression of CD30 in all cases, positive CD45RO in 6 cases, positive CD20 in only 1 case, but negative CD45RO and CD20 expressions in 3 cases. Two patients died at 7 weeks and 3.4 years of follow-up, respectively. CONCLUSION: Our study highlights the importance of histopathologic features and positive CD30 staining for differentiation of this disease from other malignant skin tumors.展开更多

In the present study, we aimed to evaluate the effect of pharmaco-serum of "Taoren-Honghua-herb pair" on IL-1β- induced chondrocyte degeneration in vitro. Taoren (Semen persicae) and Honghua (Safflower carthamus) were administered to the rats, and the pharmaco-serum was collected and prepared. Chondrocytes of the third passage, isolated from the rat's vertebral endplates, were treated by standard medium only (Group NC), IL-1β (Group IL) or combination of IL-1β and pharmaco-serum (Group TRHH). Cell proliferation and apoptosis were determined, and the expression of aggrecan, Col2ul, Coll0ul, IL-6 and SOX9 at the mRNA level in chondrocytes was quantified by real-time PCR. Immunohistochemistry staining of type II and X collagen and Safranine O staining were also used to evaluate the chondrocytes. Compared with the Group NC, IL-1β treatment inhibited the cell proliferation and induced the cell apoptosis (P〈0.05), and the expression of aggrecan, Col2αl and SOX9 at the mRNA level was down-regulated. In contrast, the expression of Coll0ul and IL-6 was up-regulated after IL-1β treatment (P〈0.05). Meanwhile, the immune-staining of type II collagen and Safranine O staining were decreased, while the staining of type X collagen was increased.
