Toll-like receptor 4 (TLR4) recognizes pathogen ligands and mediates signaling to initiate innate and adaptive immune responses. In this experiment, a 316 bp and 382 bp fragments of TLR4 gene named T4CRBR1 and T4CRB...Toll-like receptor 4 (TLR4) recognizes pathogen ligands and mediates signaling to initiate innate and adaptive immune responses. In this experiment, a 316 bp and 382 bp fragments of TLR4 gene named T4CRBR1 and T4CRBR2, of Chinese Holstein, Sanhe cattle, and Chinese Simmental was amplified by polymerase chain reaction (PCR), respectively. The genetic polymorphisms in the three populations were detected by Single-Strand Conformational Polymorphism (SSCP) in the first locus and by digesting the fragments with restriction endonuclease Alu I in the second one. Results showed that both alleles (A and B) of two loci were found in all the three populations and the value of polymorphism information content (PIC) indicated that these were a moderate polymorphism. Statistical results of X^2 test indicated that two polymorphism sites in the three populations fitted with Hardy-Weinberg equilibrium (P 〉 0.05). After sequencing, A-G single nucleotide polymorphism (SNP) was identified at nucleotide 4,525 in intron 1 of TLR4 gene and C-T SNP was identified at nucleotide 1,397 in exon 3 of TLR4 gene. Meanwhile, the effect of polymorphism of TLR4 gene on somatic cell score (SCS) was analyzed, the results indicated that the cattle with allele A in T4CRBR1 showed lower somatic cell score than that of allele B (P 〈 0.05). In short, the allele A might play an important role in mastiffs resistance in bovine.展开更多
AIM:To evaluate the role of endoscopic retrograde cholangiopancreatography(ERCP) in patients with sickle cell disease(SCD) .METHODS:Two hundred and twenty four SCD patients with cholestatic jaundice(CJ) had ERCP.The i...AIM:To evaluate the role of endoscopic retrograde cholangiopancreatography(ERCP) in patients with sickle cell disease(SCD) .METHODS:Two hundred and twenty four SCD patients with cholestatic jaundice(CJ) had ERCP.The indications for ERCP were based on clinical and biochemical evidence of CJ and ultrasound findings.RESULTS:Two hundred and forty ERCPs were performed.The indications for ERCP were:CJ only in 79,CJ and dilated bile ducts without stones in 103,and CJ and bile duct stones in 42.For those with CJ only,ERCP was normal in 42(53.2%) ,and 13(16.5%) had dilated bile ducts without an obstructive cause.In the remaining 22,there were bile duct stones with or without dilation.For those with CJ,dilated bile ducts and no stones,ERCP was normal in 17(16.5%) ,and 28(27.2%) had dilated bile ducts without an obstructive cause.In the remaining 58,there were bile ducts stones with or without dilation.For those with CJ and bile duct stones,ERCP was normal in two(4.8%) ,and 14(33.3%) had dilated bile ducts without an obstructive cause.In the remaining 26,there were bile duct stones with or without dilatation.CONCLUSION:Considering the high frequency of biliary sludge and bile duct stones in SCD,endoscopic sphincterotomy might prove helpful in these patients.展开更多
AIM: To evaluate the relation of cluster of differentiation 44 (CD44) expression with clinicopathological features of gastric adenocarcinoma, and also its effect on prognosis with an emphasis on the differences betwee...AIM: To evaluate the relation of cluster of differentiation 44 (CD44) expression with clinicopathological features of gastric adenocarcinoma, and also its effect on prognosis with an emphasis on the differences between intestinal and diffuse types. METHODS: From 2000 to 2006, 100 patients with gastric adenocarcinoma, who had undergone total or subtotal gastrectomy without any prior treatment, were studied. Haematoxylin & eosin (HE) staining was used for histological evaluation, including the type (Lauren's classifi cation) and grading of the tumor. The expression of CD44 in the gastric adenocarcinoma mucosa and the adjacent mucosa were determined by immunohistochemistry. The survival analysis was obtained using the Kaplan-Meier test. RESULTS: Of 100 patients, 74 (74%) patients were male. The tumors were categorized as intestinal type (78%) or diffuse type (22%). Sixty-five percent of patients were CD44-positive. CD44 expression was not detected in normal gastric mucosa. Rather, CD44 was more commonly expressed in the intestinal subtype (P = 0.002). A signifi cant relation was seen between the grade of tumor and the expression of CD44 (P = 0.014). The survival analysis showed a poor prognosis of patients with CD44-positive tumors (P = 0.008); and this was more prominent in the intestinal (P = 0.001) rather than diffuse type. CONCLUSION: Cell adhesion molecule CD44 is highly expressed in gastric adenocarcinoma. CD44 expression is correlated with a poor prognosis in patients with the intestinal type of gastric adenocarcinoma. CD44 can, therefore, be utilized as a prognostic marker for this group of patients.展开更多

METHODS: Bot...AIM: To test the effect of the dephytinization of three different commercial infant cereals on iron, calcium, and zinc bioavailability by estimating the uptake, retention, and transport by Caco-2 cells. METHODS: Both dephytinized (by adding an exogenous phytase) and non-dephytinized infant cereals were digested using an in vitro digestion protocol adapted to the gastrointestinal conditions of infants younger than 6 too. Mineral cell retention, transport, and uptake from infant cereals were measured using the soluble fraction of the simulated digestion and the Caco-2 cells. RESULTS: Dephytinization of infant cereals significantly increased (P 〈 0.05) the cell uptake efficiency (from 0.66%-6.05% to 3.93%-13%), retention (from 6.04%-16.68% to 14.75%-20.14%) and transport efficiency (from 0.14%-2.21% to 1.47%-6.02%), of iron, and the uptake efficiency (from 5.0%-35.4% to 7.3%-41.6%) and retention (from 4.05%-20.53% to 14.45%-61.3%) of zinc, whereas calcium only cell uptake showed a significant increase (P 〈 0.05) after removing phytate from most of the samples analyzed. A positive relationship (P 〈 0.05) between mineral solubility and the cell uptake and transport efficiencies was observed. CONCLUSION: Removing phytate from infant cereals had a beneficial effect on iron and zinc bioavailability when infant cereals were reconstituted with water. Since in developing countries cereal-based complementary foods for infants are usually consumed mixed with water, exogenous phytase additions could improve the nutritional value of this weaning food.展开更多
AIM: To evaluate the efficacy and the safety of azathioprine (AZA) and buthionine sulfoximine (BSO) bylocalized application into HepG2 tumor in vivo.METHODS: Different hepatoma and colon carcinoma cell lines (HepG2, H...AIM: To evaluate the efficacy and the safety of azathioprine (AZA) and buthionine sulfoximine (BSO) bylocalized application into HepG2 tumor in vivo.METHODS: Different hepatoma and colon carcinoma cell lines (HepG2, HuH7, Chang liver, LoVo, RKO, SW-48, SW-480) were grown in minimal essencial medium supplemented with 10% fetal bovine serum and 1% antibiotic/antimycotic solution and maintained in a humidified 37 ℃ incubator with 5% CO2. These cells were pretreated with BSO for 24 h and then with AZA for different times. We examined the effects of this combination on some proteins and on cellular death. We also studied the eff icacy and the safety of AZA (6 mg/kg per day) and BSO (90 mg/kg per day) in HepG2 tumor growth in vivo using athymic mice. We measured safety by serological markers such as aminotransferases and creatine kinase.RESULTS: The in vitro studies revealed a new mechanism of action for the AZA plus BSO combination in the cancer cells compared with other thiopurines (6-mercaptopurine, 6-methylmercaptopurine, 6-thioguanine and 6-methylthioguanine) in combination with BSO. The cytotoxic effect of AZA plus BSO in HepG2 cells resulted from necroptosis induction in a mitochondrial-dependent manner. From kinetic studies we suggest that glutathione (GSH) depletion stimulates c-Jun amino-terminal kinase and Bax translocation in HepG2 cells with subsequent deregulation of mitochondria (cytochrome c release, loss of membrane potential), and proteolysis activation leading to loss of membrane integrity, release of lactate dehydrogenase and DNA degradation. Some of this biochemical and cellular changes could be reversed by N-acetylcysteine (a GSH replenisher). In vivo studies showed that HepG2 tumor growth was inhibited when AZA was combined with BSO.CONCLUSION: Our studies suggest that a combination of AZA plus BSO could be useful for localizedtreatment of hepatocellular carcinoma as in the currently used transarterial chemoembolization method.展开更多

METHODS: The assay is based on microencapsulation technology, wherein human tumor cells are enca...AIM: To establish a more stable and accurate nude mouse model of pancreatic cancer using cancer cell microencapsulation. METHODS: The assay is based on microencapsulation technology, wherein human tumor cells are encapsulated in small microcapsules (approximately 420 μm in diameter) constructed of semipermeable membranes. We implemented two kinds of subcutaneous implantation models in nude mice using the injection of single tumor cells and encapsulated pancreatic tumor cells. The size of subcutaneously implanted tumors was observed ona weekly basis using two methods, and growth curves were generated from these data. The growth and metastasis of orthotopically injected single tumor cells and encapsulated pancreatic tumor cells were evaluated at four and eight weeks postimplantation by positron emission tomography-computed tomography scan and necropsy. The pancreatic tumor samples obtained from each method were then sent for pathological examination. We evaluated differences in the rates of tumor incidence and the presence of metastasis and variations in tumor volume and tumor weight in the cancer microcapsules vs single-cell suspensions. RESULTS: Sequential in vitro observations of the microcapsules showed that the cancer cells in microcapsules proliferated well and formed spheroids at days 4 to 6. Further in vitro culture resulted in bursting of the membrane of the microcapsules and cells deviated outward and continued to grow in flasks. The optimum injection time was found to be 5 d after tumor encapsulation. In the subcutaneous implantation model, there were no significant differences in terms of tumor volume between the encapsulated pancreatic tumor cells and cells alone and rate of tumor incidence. There was a significant difference in the rate of successful im- plantation between the cancer cell microencapsulation group and the single tumor-cell suspension group (100% vs 71.43%, respectively, P = 0.0489) in the orthotropic implantation model. The former method displayed an obvious advantage in tumor mass (4th wk: 0.0461 ± 0.0399 vs 0.0313 ± 0.021, t = -0.81, P = 0.4379; 8th wk: 0.1284 ± 0.0284 vs 0.0943 ± 0.0571, t = -2.28, respectively, P = 0.0457) compared with the latter in the orthotopic implantation model. CONCLUSION: Encapsulation of pancreatic tumor cells is a reliable method for establishing a pancreatic tumor animal model.展开更多
OBJECTIVE A system was established to evaluate the metastasis and prognosis of oral squamous cell carcinoma by analyzing the tumor differentiation, the TNM stage, the mode of invasion, and the expression of E-cadherin...OBJECTIVE A system was established to evaluate the metastasis and prognosis of oral squamous cell carcinoma by analyzing the tumor differentiation, the TNM stage, the mode of invasion, and the expression of E-cadherin and S100A4. METHODS Squamous cell carcinoma of the oral cavity of 86 cases was the focus of our study. In this system, the histopathological grade and the histochemical patterns were estimated on a 0-3 point scale, the total points graded from 0 to 13. RESULTS The incidence of metastasis and prognosis in the cases with total points more than 8 was significantly higher than that with total points less than 7 (P 〈 0.05,χ^2= 22.0658 and P 〈 0.05, χ^2= 10.7047). The system had a significant higher specificity than that of 'DIAGS index' system (Differentiation, Invasion mode, Adhesion molecules, Glycosaminoglycan, and the Sugar chain) in the evaluation of metastasis (P 〈 0.05, u = 2.2339). Moreover, the specificity for evaluation of metastasis in the system was significantly higher than that of E-cadherin (P 〈 0.05, u = 2.4996) or S100A4 (P 〈 0.05, u = 2.4289) only. Furthermore the specificity for evaluation of unfavorable prognosis in the system was also significantly higher than that of E-cadherin (P 〈 0.05, u = 2.1313) or S100A4 only (P 〈 0.05, u = 2.0301). CONCLUSION This is a valuable evaluation system with high specificity to predict metastatic potential and prognosis of oral squamous cell carcinoma.展开更多
Monitoring the service condition of concrete structures requires the quantitative assessment of properties and corrosion rate of structural steels surrounded by concrete.A multi-cell sensor system that included a refe...Monitoring the service condition of concrete structures requires the quantitative assessment of properties and corrosion rate of structural steels surrounded by concrete.A multi-cell sensor system that included a reference electrode,a chloride content sensor,a macrocell current unit and an electrical resistance measurement unit was developed.This system provided the following important electrochemical data in the cover-zone concrete on site:open circuit potential,macrocell current from anodes to cathode,chloride profile,concrete resistance and corrosion rate of built-in anodes.The experimental results show that the macrocell current increases when the chloride content in concrete is higher.Thus,monitoring the chloride content is a good method for monitoring the corrosion state.The chloride ion content and cover depth are the key factors that affect the electrical resistance of concrete.Without considering the temperature and time,a simplified model of the instantaneous corrosion rate of steel rebar in a concrete structure based on the measured chloride contents and concrete resistance was proposed.The test results further prove the reliability of this simplified predicting model.展开更多
Objective To investigate the effect of microRNA-205 reduction by antagomirs on adhesion ability of normal human corneal epithelial keratinocytes(NHCEKs).Methods Antagomir-205,complementary and inhibitory to microRNA-2...Objective To investigate the effect of microRNA-205 reduction by antagomirs on adhesion ability of normal human corneal epithelial keratinocytes(NHCEKs).Methods Antagomir-205,complementary and inhibitory to microRNA-205,was used to suppress endogenous microRNA-205 in NHCEKs.The adhesion ability of treated NHCEKs was then assessed by cell adhesion assay.Immunoblot and immunohistochemistry were conducted to determine the level of two focal adhesion-related proteins,focal adhesion kinase(FAK) and paxillin(Pax).Phalloidin staining was performed to measure the level of filamentous actin in antagomir-treated NHCEKs.Results Antagomir-205 markedly reduced the level of microRNA-205 in NHCEKs and significantly enhanced adhesion ability of NHCEKs(P<0.01).Further protein analysis validated that inhibition of mi-croRNA-205 increased the number of phosphorylated FAK and phosphorylated Pax,and decreased filamen-tous actin.Conclusion Our findings suggest that microRNA-205 has down-regulating effect on cell motility in NHCEKs.展开更多

In the s...Endophytes are beneficial microbes that are capable of promoting growth, besides protecting colonized plants against plant pathogens. These microbes are of either bacterial, fungal or actinomycetes in plants. In the study, the endophytic bacteria isolated from sugarcane with their characterization related to plant growth promotion and pathogen suppression have been reported. Roots, shoots and leaves of rooted tissue culture plantlets of sugarcane cultivars of 87A298 and 2009A107 were excised aseptically and isolated endophytic bacterial strains. The strains were identified using 16S rRNA gene sequence based homology. Molecular characterization of these strains was carried out for presence of antimicrobial genes. The results showed that the endophytes isolated from sugarcane tissue culture plantlets were of the genera Bacillus (B. amyloliquefaciens, B. subtilis, B. cereus, B. safensis, B. siamensis, B. aryabhattai, B. flexus and B. velezensis) and Paenibacillus pabuli. There were three antimicrobial peptides (AMPs) producing genes of bacilysin, bacillomycin and fengycin in B. amyloliquefaciens (SE1, SE7), B. siamensis (SE4, SE16), B. subtilis (SE2, SE3) and B. velezensis (SE15). The biochemical characterization assays showed that some of these strains could produce hydrogen cyanide (HCN), protease, cellulase and indole acetic acid (IAA). Few strains (SE1 and SE4) were phosphate solubilizers, whereas nine isolates were found to be diazotrophs. Most of the bacterial isolates were found antagonistic to Fusarium sacchari, the sugarcane wilt pathogen under in vitro conditions. Overall, the results suggested the scope and potentiality of sugarcane endophytic bacteria, isolated from tissue culture plantlets, in promoting plant growth and suppression of sugarcane pathogen.展开更多

In this study, rill erosion for...Application of simple and locally based erosion assessment methods that fit to the local condition is necessary to improve the performance and efficiency of soil conservation practices. In this study, rill erosion formation and development was investigated on the topo-sequence of three catchments (300-500 m slope length); and on agricultural fields (6 m and 24 m slope lengths) with different crop-tillage surfaces during erosive storms. Rill density and rill erosion rates were measured using rill cross section survey and close range digital photogrammetry. Rill formation and development was commonly observed on conditions where there is wider terrace spacing, concave slope shapes and unstable stone terraces on steep slopes. At field plot level, rill development was controlled by the distribution and abrupt change in the soil surface roughness and extent of slope length. At catchment scale, however, rill formation and development was controlled by landscape structures, and concavity and convexity of the slope. Greater rill cross sections and many small local rills were associated to the rougher soil surfaces. For instance, relative comparison of crop tillage practices have showed that faba-bean tillage management was more susceptible to seasonal rill erosion followed by Teff and wheat tillage surfaces under no cover condition. Surface roughness and landscape structures played a net decreasing effect on the parallel rill network development. This implies that spatial and temporal variability of the rill prone areas was strongly associated with the nature and initial size of surface micro-topography or tillage roughness. Thus, it is necessary to account land management practices, detail micro-topographic surfaces and landscape structures for improved prediction of rill prone areas under complex topographic conditions. Application of both direct rill cross section survey and close range digital photogrammetric techniques could enhance field erosion assessment for practical soil conservation improvement.展开更多

文摘AIM: To test the effect of the dephytinization of three different commercial infant cereals on iron, calcium, and zinc bioavailability by estimating the uptake, retention, and transport by Caco-2 cells. METHODS: Both dephytinized (by adding an exogenous phytase) and non-dephytinized infant cereals were digested using an in vitro digestion protocol adapted to the gastrointestinal conditions of infants younger than 6 too. Mineral cell retention, transport, and uptake from infant cereals were measured using the soluble fraction of the simulated digestion and the Caco-2 cells. RESULTS: Dephytinization of infant cereals significantly increased (P 〈 0.05) the cell uptake efficiency (from 0.66%-6.05% to 3.93%-13%), retention (from 6.04%-16.68% to 14.75%-20.14%) and transport efficiency (from 0.14%-2.21% to 1.47%-6.02%), of iron, and the uptake efficiency (from 5.0%-35.4% to 7.3%-41.6%) and retention (from 4.05%-20.53% to 14.45%-61.3%) of zinc, whereas calcium only cell uptake showed a significant increase (P 〈 0.05) after removing phytate from most of the samples analyzed. A positive relationship (P 〈 0.05) between mineral solubility and the cell uptake and transport efficiencies was observed. CONCLUSION: Removing phytate from infant cereals had a beneficial effect on iron and zinc bioavailability when infant cereals were reconstituted with water. Since in developing countries cereal-based complementary foods for infants are usually consumed mixed with water, exogenous phytase additions could improve the nutritional value of this weaning food.

文摘AIM: To evaluate the efficacy and the safety of azathioprine (AZA) and buthionine sulfoximine (BSO) bylocalized application into HepG2 tumor in vivo.METHODS: Different hepatoma and colon carcinoma cell lines (HepG2, HuH7, Chang liver, LoVo, RKO, SW-48, SW-480) were grown in minimal essencial medium supplemented with 10% fetal bovine serum and 1% antibiotic/antimycotic solution and maintained in a humidified 37 ℃ incubator with 5% CO2. These cells were pretreated with BSO for 24 h and then with AZA for different times. We examined the effects of this combination on some proteins and on cellular death. We also studied the eff icacy and the safety of AZA (6 mg/kg per day) and BSO (90 mg/kg per day) in HepG2 tumor growth in vivo using athymic mice. We measured safety by serological markers such as aminotransferases and creatine kinase.RESULTS: The in vitro studies revealed a new mechanism of action for the AZA plus BSO combination in the cancer cells compared with other thiopurines (6-mercaptopurine, 6-methylmercaptopurine, 6-thioguanine and 6-methylthioguanine) in combination with BSO. The cytotoxic effect of AZA plus BSO in HepG2 cells resulted from necroptosis induction in a mitochondrial-dependent manner. From kinetic studies we suggest that glutathione (GSH) depletion stimulates c-Jun amino-terminal kinase and Bax translocation in HepG2 cells with subsequent deregulation of mitochondria (cytochrome c release, loss of membrane potential), and proteolysis activation leading to loss of membrane integrity, release of lactate dehydrogenase and DNA degradation. Some of this biochemical and cellular changes could be reversed by N-acetylcysteine (a GSH replenisher). In vivo studies showed that HepG2 tumor growth was inhibited when AZA was combined with BSO.CONCLUSION: Our studies suggest that a combination of AZA plus BSO could be useful for localizedtreatment of hepatocellular carcinoma as in the currently used transarterial chemoembolization method.

This article reviews the current modalities employed for the diagnosis of HCC, including serum markers, radiological techniques and histological evaluation, and summarises international guidelines for the diagnostic approach to HCC.

文摘AIM: To establish a more stable and accurate nude mouse model of pancreatic cancer using cancer cell microencapsulation. METHODS: The assay is based on microencapsulation technology, wherein human tumor cells are encapsulated in small microcapsules (approximately 420 μm in diameter) constructed of semipermeable membranes. We implemented two kinds of subcutaneous implantation models in nude mice using the injection of single tumor cells and encapsulated pancreatic tumor cells. The size of subcutaneously implanted tumors was observed ona weekly basis using two methods, and growth curves were generated from these data. The growth and metastasis of orthotopically injected single tumor cells and encapsulated pancreatic tumor cells were evaluated at four and eight weeks postimplantation by positron emission tomography-computed tomography scan and necropsy. The pancreatic tumor samples obtained from each method were then sent for pathological examination. We evaluated differences in the rates of tumor incidence and the presence of metastasis and variations in tumor volume and tumor weight in the cancer microcapsules vs single-cell suspensions. RESULTS: Sequential in vitro observations of the microcapsules showed that the cancer cells in microcapsules proliferated well and formed spheroids at days 4 to 6. Further in vitro culture resulted in bursting of the membrane of the microcapsules and cells deviated outward and continued to grow in flasks. The optimum injection time was found to be 5 d after tumor encapsulation. In the subcutaneous implantation model, there were no significant differences in terms of tumor volume between the encapsulated pancreatic tumor cells and cells alone and rate of tumor incidence. There was a significant difference in the rate of successful im- plantation between the cancer cell microencapsulation group and the single tumor-cell suspension group (100% vs 71.43%, respectively, P = 0.0489) in the orthotropic implantation model. The former method displayed an obvious advantage in tumor mass (4th wk: 0.0461 ± 0.0399 vs 0.0313 ± 0.021, t = -0.81, P = 0.4379; 8th wk: 0.1284 ± 0.0284 vs 0.0943 ± 0.0571, t = -2.28, respectively, P = 0.0457) compared with the latter in the orthotopic implantation model. CONCLUSION: Encapsulation of pancreatic tumor cells is a reliable method for establishing a pancreatic tumor animal model.

文摘OBJECTIVE A system was established to evaluate the metastasis and prognosis of oral squamous cell carcinoma by analyzing the tumor differentiation, the TNM stage, the mode of invasion, and the expression of E-cadherin and S100A4. METHODS Squamous cell carcinoma of the oral cavity of 86 cases was the focus of our study. In this system, the histopathological grade and the histochemical patterns were estimated on a 0-3 point scale, the total points graded from 0 to 13. RESULTS The incidence of metastasis and prognosis in the cases with total points more than 8 was significantly higher than that with total points less than 7 (P 〈 0.05,χ^2= 22.0658 and P 〈 0.05, χ^2= 10.7047). The system had a significant higher specificity than that of 'DIAGS index' system (Differentiation, Invasion mode, Adhesion molecules, Glycosaminoglycan, and the Sugar chain) in the evaluation of metastasis (P 〈 0.05, u = 2.2339). Moreover, the specificity for evaluation of metastasis in the system was significantly higher than that of E-cadherin (P 〈 0.05, u = 2.4996) or S100A4 (P 〈 0.05, u = 2.4289) only. Furthermore the specificity for evaluation of unfavorable prognosis in the system was also significantly higher than that of E-cadherin (P 〈 0.05, u = 2.1313) or S100A4 only (P 〈 0.05, u = 2.0301). CONCLUSION This is a valuable evaluation system with high specificity to predict metastatic potential and prognosis of oral squamous cell carcinoma.

文摘Monitoring the service condition of concrete structures requires the quantitative assessment of properties and corrosion rate of structural steels surrounded by concrete.A multi-cell sensor system that included a reference electrode,a chloride content sensor,a macrocell current unit and an electrical resistance measurement unit was developed.This system provided the following important electrochemical data in the cover-zone concrete on site:open circuit potential,macrocell current from anodes to cathode,chloride profile,concrete resistance and corrosion rate of built-in anodes.The experimental results show that the macrocell current increases when the chloride content in concrete is higher.Thus,monitoring the chloride content is a good method for monitoring the corrosion state.The chloride ion content and cover depth are the key factors that affect the electrical resistance of concrete.Without considering the temperature and time,a simplified model of the instantaneous corrosion rate of steel rebar in a concrete structure based on the measured chloride contents and concrete resistance was proposed.The test results further prove the reliability of this simplified predicting model.

文摘Objective To investigate the effect of microRNA-205 reduction by antagomirs on adhesion ability of normal human corneal epithelial keratinocytes(NHCEKs).Methods Antagomir-205,complementary and inhibitory to microRNA-205,was used to suppress endogenous microRNA-205 in NHCEKs.The adhesion ability of treated NHCEKs was then assessed by cell adhesion assay.Immunoblot and immunohistochemistry were conducted to determine the level of two focal adhesion-related proteins,focal adhesion kinase(FAK) and paxillin(Pax).Phalloidin staining was performed to measure the level of filamentous actin in antagomir-treated NHCEKs.Results Antagomir-205 markedly reduced the level of microRNA-205 in NHCEKs and significantly enhanced adhesion ability of NHCEKs(P<0.01).Further protein analysis validated that inhibition of mi-croRNA-205 increased the number of phosphorylated FAK and phosphorylated Pax,and decreased filamen-tous actin.Conclusion Our findings suggest that microRNA-205 has down-regulating effect on cell motility in NHCEKs.
文摘Endophytes are beneficial microbes that are capable of promoting growth, besides protecting colonized plants against plant pathogens. These microbes are of either bacterial, fungal or actinomycetes in plants. In the study, the endophytic bacteria isolated from sugarcane with their characterization related to plant growth promotion and pathogen suppression have been reported. Roots, shoots and leaves of rooted tissue culture plantlets of sugarcane cultivars of 87A298 and 2009A107 were excised aseptically and isolated endophytic bacterial strains. The strains were identified using 16S rRNA gene sequence based homology. Molecular characterization of these strains was carried out for presence of antimicrobial genes. The results showed that the endophytes isolated from sugarcane tissue culture plantlets were of the genera Bacillus (B. amyloliquefaciens, B. subtilis, B. cereus, B. safensis, B. siamensis, B. aryabhattai, B. flexus and B. velezensis) and Paenibacillus pabuli. There were three antimicrobial peptides (AMPs) producing genes of bacilysin, bacillomycin and fengycin in B. amyloliquefaciens (SE1, SE7), B. siamensis (SE4, SE16), B. subtilis (SE2, SE3) and B. velezensis (SE15). The biochemical characterization assays showed that some of these strains could produce hydrogen cyanide (HCN), protease, cellulase and indole acetic acid (IAA). Few strains (SE1 and SE4) were phosphate solubilizers, whereas nine isolates were found to be diazotrophs. Most of the bacterial isolates were found antagonistic to Fusarium sacchari, the sugarcane wilt pathogen under in vitro conditions. Overall, the results suggested the scope and potentiality of sugarcane endophytic bacteria, isolated from tissue culture plantlets, in promoting plant growth and suppression of sugarcane pathogen.

文摘Application of simple and locally based erosion assessment methods that fit to the local condition is necessary to improve the performance and efficiency of soil conservation practices. In this study, rill erosion formation and development was investigated on the topo-sequence of three catchments (300-500 m slope length); and on agricultural fields (6 m and 24 m slope lengths) with different crop-tillage surfaces during erosive storms. Rill density and rill erosion rates were measured using rill cross section survey and close range digital photogrammetry. Rill formation and development was commonly observed on conditions where there is wider terrace spacing, concave slope shapes and unstable stone terraces on steep slopes. At field plot level, rill development was controlled by the distribution and abrupt change in the soil surface roughness and extent of slope length. At catchment scale, however, rill formation and development was controlled by landscape structures, and concavity and convexity of the slope. Greater rill cross sections and many small local rills were associated to the rougher soil surfaces. For instance, relative comparison of crop tillage practices have showed that faba-bean tillage management was more susceptible to seasonal rill erosion followed by Teff and wheat tillage surfaces under no cover condition. Surface roughness and landscape structures played a net decreasing effect on the parallel rill network development. This implies that spatial and temporal variability of the rill prone areas was strongly associated with the nature and initial size of surface micro-topography or tillage roughness. Thus, it is necessary to account land management practices, detail micro-topographic surfaces and landscape structures for improved prediction of rill prone areas under complex topographic conditions. Application of both direct rill cross section survey and close range digital photogrammetric techniques could enhance field erosion assessment for practical soil conservation improvement.
