Optical-neural stimulation,which encompasses cutting-edge techniques such as optogenetics and infrared neurostimulation,employs distinct mechanisms to modulate brain function and behavior.These advanced neuromodulation techniques offer accurate manipulation of targeted areas,even selectively modulating specific neurons,in the brain.This makes it possible to investigate the cause-and-effect connections between neural activity and behavior,allowing for a better comprehension of the intricate brain dynamics towards complex environments.Non-human primates serve as an essential animal model for investigating these complex functions in brain research,bridging the gap between the basic research and clinical applications.One of the earliest optical studies utilizing optogenetic neuromodulation in monkeys was conducted in 2009.Since then,the optical-neural stimulations have been effectively applied in non-human primates.This review summarises recent research that employed optogenetics or infrared neurostimulation techniques to regulate brain function and behavior in non-human primates.The current state of optical-neural stimulations discussed here demonstrates their efficacy in advancing the understanding of brain systems.Nevertheless,there are still challenges that need to be addressed before they can fully achieve their potential. 展开更多

目的:冠状动脉慢性闭塞病变(CTO)的介入治疗是当前的热点和难点之一,当CTO病变介入治疗失败或潜在风险超过预期获益时,内膜下斑块修饰技术(SPM)可以成为一项替代性的策略选择。本研究意在探讨应用SPM技术,其实施的安全性和有效性。方法... 目的:冠状动脉慢性闭塞病变(CTO)的介入治疗是当前的热点和难点之一,当CTO病变介入治疗失败或潜在风险超过预期获益时,内膜下斑块修饰技术(SPM)可以成为一项替代性的策略选择。本研究意在探讨应用SPM技术,其实施的安全性和有效性。方法:本研究连续性入选自2015年1月至2019年12月,于我院行冠状动脉造影提示为CTO病变,并在后续手术过程中行SPM技术的患者。主要终点为30~90d内复查冠状动脉造影时的血管再通。次要终点为靶病变血运重建成功。安全性终点为围术期严重并发症发生率。同时对患者行SPM术后24个月进行门诊或电话随访,随访终点为症状驱动的靶血管再次血运重建的发生。结果:自2015年1月至2019年12月,共54例患者(54例病变)纳入本研究,其中有22例纳入改良SPM技术组,32例纳入传统的SPM技术组。主要终点为传统SPM技术组为20例(62.5%),而改良SPM技术组为20例(90.9%)(P<0.05),改良SPM技术组血管再通率显著更高。次要终点传统SPM技术完成26例(81.3%),改良SPM组完成21例(95.9%),两组差异无统计学意义。随访终点传统SPM组共有5例患者完成了因症状驱动的靶血管再次血运重建,而改良SPM组为2例,两组差异无统计学意义。结论:在尝试开通CTO病变的过程中,若开通该病变潜在的风险已经大于获益,可以采用SPM技术,在随访时再次尝试开通病变,血管再通率和血运重建成功率均较高,且长期随访显示该技术的有效性可以得到保证。 展开更多

The module for function electrical stimulation (FES) of neurons is designed for the research of the neural function regeneration microelectronic system, which is an in-body embedded micro module. It is implemented by using discrete devices at first and characterized in vitro. The module is used to stimulate sciatic nerve and spinal cord of rats and rabbits for in-vivo real-time experiments of the neural function regeneration system. Based on the module, a four channel module for the FES of neurons is designed for 12 sites cuff electrode or 10 sites shaft electrode. Three animal experiments with total five rats and two rabbits were made. In the in-vivo experiment, the neural signals including spontaneous and imitated were regenerated by the module. The stimulating signal was used to drive sciatic nerve and spinal cord of rats and rabbits, successfully caused them twitch in different parts of their bodies, such as legs, tails, and fingers. This testifies that the neural function regeneration system can regenerate the neural signals. 展开更多

Objective To analyze the interactions between different structural types of volatile oil compo-nents(VOCs)and skin lipid molecules;and investigate the mechanism of volatile oil in Chi-nese materia medica(VOCMM)as penetration enhancers.Methods In this study;210 different structural types of VOCs were selected from the VOCMM penetration enhancer database;and the molecular docking experiments were conducted with three main lipid molecules of skin:ceramide 2(CER2);cholesterol(CHL);and free fatty acid(FFA).Each VOC was docked individually with each lipid molecule.Cluster analysis was used to explore the relationship between the binding energy of VOCs and their molecular struc-tures.Nine specific pathogen-free(SPF)Sprague Dawley(SD)rats were randomly divided in-to Control;Nootkatone;and 3-Butylidenephthalide groups for in vitro percutaneous experi-ments;with three rats in each group.The donor pool solutions were 3%gastrodin;3%gas-trodin+3%nootkatone;and 3%gastrodin+3%3-butylidenephthalide;respectively.The pen-etration enhancing effects of VOCs with higher binding energy were evaluated by comparing the 12-hour cumulative percutaneous absorption of gastrodin(Q12;µg/cm²).Results(i)Most of the VOCs were non-hydrogen bonded to the hydrophobic parts of CHL and FFA;and hydrogen bonded to the head group of CER2.Among them;sesquiterpene ox-ides showed the most pronounced binding affinity to CER2.The VOCs with 2-4 rings(in-cluding carbon rings;benzene rings;and heterocycles)demonstrated stronger binding affini-ty for three skin lipid molecules compared with the VOCs without intramolecular rings(P<0.01).(ii)According to the cluster analysis;most of the VOCs that bond well to CER2 had 2-3 intramolecular rings.The non-oxygenated VOCs were bonded to CER2 in a hydrophobic manner.The oxygenated VOCs were mostly bonded to CER2 by hydrogen bonding.(iii)The results of Franz diffusion cell experiment showed that the Q12 of Control group was 260.60±25.09µg/cm2;and the transdermal absorption of gastrodin was significantly increased in Nootkatone group(Q12=5503.00±1080.00µg/cm²;P<0.01).The transdermal absorption of gastrodin was also increased in 3-Butylidenephthalide group(Q12=495.40±56.98µg/cm²;P>0.05).(iv)The type of oxygen-containing functional groups in VOCs was also an influencing factor of binding affinity to CER2.Conclusion The interactions between different types of VOCs with different structures in the VOCMM and three skin lipid molecules in the stratum corneum were investigated at the molecular level in this paper.This research provided theoretical guidance and data support for the screening of volatile oil-based penetration enhancers;and a simple and rapid method for studying the penetration-enhancing mechanism of volatile oils. 展开更多

R-apomorphine protects against 6-hydroxydopamine-induced nigrostriatal damage in rat 被引量:1

Objective The aim of the present study was not only to assess the retrograde degenerative changes in the dopaminergic neurons of the substantia nigra (SN) and ventral tegmental area (VTA) after injection of 6-hydroxydopamine (6-OHDA) into the striatum, but also to use this 6-OHDA model of Parkinson's disease to explore the possible neuroprotective effect of R-apomorphine (R-APt). Methods The partial lesion was obtained by intrastriatal administration of 6-OHDA. R- APt administration (10 mg/kg, s.c.) started 15 min prior to lesioning and continued daily for another 22 days post surgery. Testing was carried out 5 weeks after lesioning. We investigated the histology and associated behavior and neurochemical changes. Structural and functional deficits were quantified by tyrosine hydroxylase (TH) / Nissl-staining cell number counting, striatal dopamine (DA) content determination and amphetamine-induced rotation analysis. Results R-APt- treatment attenuated the amphetamine-induced ipsiversive rotation 5 weeks after the lesion induction. R-APt administra- tion for 22 days significantly reduced the size of the lesion at the level of the SN from 50% (control group) to 69%. Moreover, the cell shape resembled that observed in the intact animals. R-APt treatment significantly increased the number of cells in both the lesion and the intact sides of VTA by 60%, suggesting selective neurotrophic effect of R-APt in this area. Finally, R-APt-treatment significantly attenuated the 6-OHDA-induced striatal DA depletion and normalized dihydroxyphenylacetic acid (DOPAC)/DA ratios. Conclusion We conclude that R-APt has neuroprotective and pos- sible neurotrophic effect on a striatal lesion with 6-OHDA, suggesting that this drug may have rescuing properties in patients with early stage Parkinson's disease. These effects are more pronounced in VTA and enhance with duration of treatment. 展开更多

Penetration therapy with head electrical acupuncture was administered in treatment. Normal group, sham-operation group, model group and penetration therapy group were set up. (1)lmmunohistochemical (IHC) method was used to test the morphology and count of positive cell of tyrosine hydroxylase (TH). (2)RT-PCR technology was used to detect the expression of nestin mRNA of neural stem cell (NSC). Results (1)Compared with model group, in pene- tration therapy group, the expressions of TH-positive neurons in immune response were increased in areal density (AD), numerical density (ND) and integrating optic density (P〈0.05). (2)Compared with model group, in penetration therapy group, the expression of nestin mRNA was increased (P〈0. 05). It has been well established that the recovery ability of central nervous system (CNS) is very poor in adult mammals. As a result, CNS trauma generally leads to severe and persistent functional deficits. Thus, the investigation in this field becomes a "hot spot". Up to date, accumulating evidence supports the hypothesis that the failure of CNS neurons to regenerate is not due to their intrinsic inability to grow new axons, but due to their growth state and due to lack of a permissive growth environment. Therefore, any successful approaches to facilitate the regeneration of injured CNS axons will likely include multiple steps: keeping neurons alive in a certain growth-state, preventing the formation of a glial scar, overcoming inhibitory molecules present in the myelin debris, and giving direction to the growing axons. This brief review focused on the recent progress in the neuron regeneration of CNS in adult mammals. 展开更多

Planarian is among the simplest animals that possess a centralized nervous system (CNS), and its neural regenera- tion involves the replacement of cells lost to normal 'wear and tear' (cell turnover), and/or injury. In this review, we state and discuss the recent studies on molecular control of neural regeneration in planarians. The spatial and temporal expression patterns of genes in intact and regenerating planarian CNS have already been described relatively clearly. The bone morphogenetic protein (BMP) and Wnt signaling pathways are identified to regulate neural regeneration. During neural regeneration, conserved axon guidance mechanisms are necessary for proper wiring of the nervous system. In addition, apoptosis may play an important role in controlling cell numbers, eliminating unnecessary tissues or cells and remodeling the old tissues for regenerating CNS. The bilateral symmetry is established by determination of anterior-posterior (A-P) and dorsal-ventral (D-V) patterns. Moreover, neurons positive to dopamine, serotonin (5-HT), and gamma-aminobutyric acid (GABA) have been detected in planarians. Therefore, planarians present us with new, experimentally accessible contexts to study the molecular actions guiding neural regeneration. 展开更多
