络治法对STZ诱导的糖尿病大鼠视网膜NF-κB表达的影响 被引量:4

目的观察络治法对链佐脲菌素(STZ)诱导的糖尿病大鼠视网膜组织中核转录因子κB(NF-κB)表达的影响,探讨络治法对糖尿病视网膜病变微血管损伤的保护作用及作用机制。方法 60只雌性Wistar大鼠随机分成正常组、模型组、羟苯磺酸钙对照药物... 目的观察络治法对链佐脲菌素(STZ)诱导的糖尿病大鼠视网膜组织中核转录因子κB(NF-κB)表达的影响,探讨络治法对糖尿病视网膜病变微血管损伤的保护作用及作用机制。方法 60只雌性Wistar大鼠随机分成正常组、模型组、羟苯磺酸钙对照药物组、络治组四组。除正常组不做处理外,其它各组均采用STZ腹腔注射制作糖尿病大鼠模型。给药24周后每组大鼠取右眼做眼球壁石蜡切片,行NF-κB免疫组织化学染色,计算机图像定量分析灰度值。左眼取视网膜组织,Western blot法检测视网膜中NF-κB p65蛋白含量。结果免疫组化结果显示络治组视网膜中NF-κB表达明显低于模型组及对照组(P<0.05),灰度值与空白组大体相当(P>0.05)。Western blot结果显示络治组NF-κB p65表达较模型组及对照组显著降低(P<0.05)。结论络治法可明显抑制视网膜组织中NF-κB的活化,甚至接近正常大鼠水平。这可能是络治法对糖尿病大鼠视网膜微血管病变发挥防治作用的重要机理之一。 展开更多

目的观察络治法对链佐脲菌素(STZ)诱导的糖尿病大鼠视网膜毛细血管消化铺片中血管细胞黏附分子-1(VCAM-1)表达的影响,探讨络治法对糖尿病视网膜病变微血管损伤的保护作用及机制。方法将60只雌性Wistar大鼠随机分成空白组、模型组、对照... 目的观察络治法对链佐脲菌素(STZ)诱导的糖尿病大鼠视网膜毛细血管消化铺片中血管细胞黏附分子-1(VCAM-1)表达的影响,探讨络治法对糖尿病视网膜病变微血管损伤的保护作用及机制。方法将60只雌性Wistar大鼠随机分成空白组、模型组、对照组及络治组。除空白组不做处理外,其余各组均制作糖尿病大鼠模型并按方案给药。给药24周后每组大鼠取双侧眼球做视网膜血管消化铺片,右眼PAS染色做形态学观察,左眼免疫荧光染色法测定VCAM-1在血管壁中的表达。结果 PAS染色结果显示,络治组糖尿病大鼠视网膜毛细血管在管径粗细、局部扩张程度、扭曲聚集程度等方面较对照组及模型组有明显改善,且络治组大鼠毛细血管管壁周细胞的丢失明显减少,组间比较有统计学差异(P<0.05)。免疫荧光染色显示,络治组视网膜血管壁中VCAM-1呈较少的表达,组间比较有统计学差异(P<0.05)。结论络治法能够抑制VCAM-1在糖尿病视网膜毛细血管壁中的表达,有效减轻高血糖引起的视网膜微血管损伤,改善糖尿病视网膜微血管的结构与机能。 展开更多

观察益气扶正、养血通络外治疗法治疗气血两虚型青盲临床效果。方法 选取2022年05月-2023年03月本院门诊及住院72例气血两虚型青盲患者为样本,随机数字表法分组,均单眼发病,眼周与全身穴位注射黄芪注射液纳入治疗组,眼周与全身穴位针刺... 观察益气扶正、养血通络外治疗法治疗气血两虚型青盲临床效果。方法 选取2022年05月-2023年03月本院门诊及住院72例气血两虚型青盲患者为样本,随机数字表法分组,均单眼发病,眼周与全身穴位注射黄芪注射液纳入治疗组,眼周与全身穴位针刺纳入对照组,青盲治疗周期20d,治疗10d停止1周,对比青盲患者视力恢复情况、眼底视网膜恢复情况、视野恢复情况。结果 治疗组疗效86.1%较对照组75%高,存在统计学差异(P<0.05);治疗组视野平均缺损和平均光敏度均优于对照组,存在统计学差异(P<0.05)。结论 选用益气扶正、养血通络外治疗法治疗气血两虚型青盲疗效确切,作用直接。 展开更多

AIM: In the present study, antibody and peripheral blood mononuclear cells (PBMC) proliferative responses against hepatitis C virus (HCV) antigens were evaluated in HCV chronically infected patients. METHODS: Paired serum and PBMC samples were taken six months apart from 34 individuals, either treated or not, and tested by enzyme-linked immunosorbent assay (ELISA) and carboxyfluorescein succinimidyl ester staining. RSULTS: Over 70% of the patients showed specific IgG and IgM against capsid, E1 and NS3, while HVR-1 was recognized by half of the patients. An increase in the levels of the anti-capsid IgM (P = 0.027) and IgG (P = 0.0006) was observed in six-month samples, compared to baseline. Similarly, a significantly higher percent of patients had detectable IgA reactivity to capsid (P = 0.017) and NS3 (P = 0.005) after six months, compared to baseline. Particularly, IgA against structural antigens positively correlated with hepatic damage (P = 0.036). IgG subclasses evaluation against capsid and NS3 revealed a positive recognition mediated by IgG1 in more than 80% of the individuals. On the contrary, less than 30% of the patients showed a positive proliferative response either of CD4+ or CD8+ T cells, being the capsid poorly recognized. CONCLUSION: These results confirm that while the cellular immune response is narrow and weak, a broad and vigorous humoral response occurs in HCV chronic infection. The observed correlation between IgA and hepatic damage may have diagnostic significance, although it warrants further confirmation. 展开更多

Due to the substantial role of bridges in transportation networks and in accordance with the limited funding for bridge management, remediation strategies have to be prioritised. A conservative bridge assessment will result in unnecessary actions, such as costly bridge strengthening or repairs. On the other hand, any bridge maintenance negligence and delayed actions may lead to heavy future costs or degraded assets. The accuracy of decisions developed by any manager or bridge engineer relies on the accuracy of the bridge condition assessment which emanates from visual inspection. Many bridge rating systems are based on a very subjective procedure and are associated with uncertainty and personal bias. The developing condition rating method described herein is an important step in adding more holism and objectivity to the current approaches. Structural importance and material vulnerability are the two main factors that should be considered in the evaluation of element structural index and the causal factor as the representative of age, environment, road class and inspection is implemented as a coefficient to the OSCI (overall structural condition index). The AHP (analytical hierarchy process) has been applied to evaluate the priority vector of the causal parameters. 展开更多
