阿维拉霉素(avilamycin)是一种新型消化促进剂,能够明显的促进鸡和猪的生长,提高饲料的利用率。但是,在发酵生产研究中发现菌丝结团是影响阿维拉霉素生产的主要问题之一。文章主要研究了碳源、氮源、金属离子及接种量对阿维拉霉素生产... 阿维拉霉素(avilamycin)是一种新型消化促进剂,能够明显的促进鸡和猪的生长,提高饲料的利用率。但是,在发酵生产研究中发现菌丝结团是影响阿维拉霉素生产的主要问题之一。文章主要研究了碳源、氮源、金属离子及接种量对阿维拉霉素生产菌丝结团的影响。试验研究表明,菌团的形成会降低阿维拉霉素的产量,阿维拉霉素的产量与菌体生物量成正相关,与菌丝团的大小成负相关。容易被菌体利用的碳源,有利于菌丝生长,不利于菌丝体结团;容易利用的氮源和不容易利用的氮源复合不利于菌丝体结团。同时研究发现,适量钙、镁及铁离子促进菌体生长、抑制菌丝结团;过量的钙、镁、铁离子抑制菌体生长,促进菌体结团;适量的锰、钴离子有利于阿维拉霉素产生,但对菌体生长没有明显的影响,且过量的锰、钴离子抑制菌体生长,促进了菌丝团的形成,不利于阿维拉霉素的生产。 展开更多

以褐煤、肥煤和无烟煤为研究对象,进行降解时间、粒径和煤阶为因素的降解正交试验及绿孢链霉菌降解固定粒径各煤阶煤的实验;通过COD测定、元素分析和接触角测试,分析比较降解前后溶液有机物含量、煤的组成及亲水性,进而分析绿孢链霉菌... 以褐煤、肥煤和无烟煤为研究对象,进行降解时间、粒径和煤阶为因素的降解正交试验及绿孢链霉菌降解固定粒径各煤阶煤的实验;通过COD测定、元素分析和接触角测试,分析比较降解前后溶液有机物含量、煤的组成及亲水性,进而分析绿孢链霉菌对各煤阶煤降解转化的影响因素。结果表明:影响绿孢链霉菌对煤降解效果的主要因素是煤阶;绿孢链霉菌最容易利用降解褐煤生成的有机小分子物质,其次是利用降解肥煤与无烟煤生成的有机小分子物质;各煤阶煤经绿孢链霉菌降解后C元素和N元素含量相对升高,O元素和S元素含量及n(H)∶n(C)和n(O)∶n(C)值均降低;煤样经绿孢链霉菌降解后,褐煤的固液接触角变大,疏水性增强,而肥煤与无烟煤的固液接触角变小,亲水性增强,与各煤阶煤样的固液接触角由大到小的三种溶液依次为蒸馏水、矿井水和KO溶液。 展开更多

[目的]为阐明1株拮抗放线菌菌株的分类地位提供依据。[方法]以从东北地区蔬菜保护地土壤中分离的1株拮抗放线菌菌株T8-41为试材,分析其形态和培养特征、细胞壁成分和生理生化特性。经PCR扩增后,对其与相关菌株的16S rDNA进行同源性分析... [目的]为阐明1株拮抗放线菌菌株的分类地位提供依据。[方法]以从东北地区蔬菜保护地土壤中分离的1株拮抗放线菌菌株T8-41为试材,分析其形态和培养特征、细胞壁成分和生理生化特性。经PCR扩增后,对其与相关菌株的16S rDNA进行同源性分析和聚类分析。[结果]拮抗菌株T8-41属于典型的链霉菌属菌丝形态。其细胞壁中含有L-DAP和甘氨酸,细胞壁类型为I型。该菌可利用D-木糖、L-鼠李糖、D-果糖、蔗糖、棉子糖、葡萄糖、D-甘露醇和肌醇,但不利用乳糖。可以初步鉴定为绿产色链霉菌。T8-41菌株的16SrDNA全长为1 465 bp,与绿产色链霉菌相似性达99%。[结论]链霉菌T8-41属于青色类群的绿产色链霉菌,可将其定名为绿产色链霉菌T8-41(S.viridochromogenes T8-41)。 展开更多

目的通过检测环氧化酶-1(COX-1)和环氧化酶-2(COX-2)在人脑星形细胞瘤、正常脑组织中的表达,探讨COX在人脑星形细胞瘤发生发展中的作用.方法应用逆转录聚合酶链反应(RT-PCR)和免疫组化SP法检测人脑星形细胞瘤、正常脑组织中COX-1,COX-2m... 目的通过检测环氧化酶-1(COX-1)和环氧化酶-2(COX-2)在人脑星形细胞瘤、正常脑组织中的表达,探讨COX在人脑星形细胞瘤发生发展中的作用.方法应用逆转录聚合酶链反应(RT-PCR)和免疫组化SP法检测人脑星形细胞瘤、正常脑组织中COX-1,COX-2mRNA和蛋白的表达及其与肿瘤恶性程度、病理级别的关系.结果RT-PCR检测20例人脑星形细胞瘤组织COX-2mRNA均阳性表达,正常脑组织中未见表达,两者有显著性差异(P<0.0001);COX-1mRNA在20例肿瘤标本及正常脑组织中均有结构性表达.50例人脑星形细胞瘤标本免疫组化显示COX-2蛋白主要集中在肿瘤细胞胞浆内,阳性表达率为72%,正常脑组织未见阳性表达;而COX-1蛋白主要表达在正常脑组织中,肿瘤组织中为阴性表达.COX-2蛋白阳性表达强度与肿瘤病理级别有关,恶性程度较高的Ⅲ,Ⅳ级星形细胞瘤高于恶性程度较低的Ⅰ,Ⅱ级星形细胞瘤.结论COX-2mRNA及其蛋白在人脑星形细胞瘤组织中阳性表达且与肿瘤病理级别相关,说明COX-2可能在人脑星形细胞瘤的发生发展中起重要作用. 展开更多

Objective] This study aimed to determine the effects of medium composi-tion and fermentation condition on the production of antibiotics from an actinomycete antagonistic to Penicil ium viridicatum, which was isolated from soil and identified primarily as Streptomyces anandi . [Method] Single-factor experiments and orthogonal array designs were used to optimize the medium composition and fermentation con-ditions. [Result] Culture medium composition and fermentation conditions showed sig-nificant influence on the antibiotic production from S. anandi . The optimum culture medium consisted of soluble starch 10 g/L, soybean cake powder 15 g/L, KH2PO4 0.5 g/L and MgSO4·7H2O 0.1 g/L. The optimized fermentation conditions were tem-perature 30 ℃, initial pH value 7.7, culture medium amount 40 ml/250 ml flask, and fermentation time 144 h. The titer of fermentation broth reached 452.7 U/ml when the S. anandi was cultured under the optimal fermentation medium and conditions. [Conclusion] The study may lay the foundation on further isolation and purification of the antibiotic from S. anandi JXNU02. 展开更多

In order to evaluate the impact of green production introduced by the manufacturer and study the competitive strategy between two retailers,four strategic models among diverse retailers,i.e.,Cournot,Stackelberg and two cases of collusion,are constructed.The optimal decision is obtained by the game theory and numerical analysis.The results indicate that the manufacturer's marginal profitability is no less than that of the two retailers.However,due to the manufacturer's green cost,the downstream profitability of the supply chain may exceed the upstream manufacturer's profitability.The retailers'Cournot behavior causes the manufacturer to acquire the highest profitability and set the highest green level.However,there is an opposite result in collusion.Two retailers will choose Cournot pricing when the consumer's green sensitivity is higher than a threshold.When consumers are more sensitive to the price difference of products,retailers prefer collusion,and then choose the Stackelberg behavior.Manufacturers'profitability will increase as competition among retailers intensifies.Proper competition allows two duopolistic retailers to acquire a higher profitability. 展开更多

A gene (NANOC-D6D) encoding a desaturase that removes two hydrogen atoms from fatty acids at delta 6 position was isolated from a cDNA library of Nannochloropsis oculata (Droop) D. J. Hibberd (Eustigmatophyceae). The unicellular marine microalga N. oculata synthesizes rich long chain polyunsaturated fatty acids (LCPUFAs), including eicosapentaenoic acid (20:5n-3, EPA). The deduced protein contains 474 amino acids that fold into 4 trans-membrane domains. The neighbor-joining phylogenetic tree indicates that NANOC-D6D is phylogenetically close to the delta-6 fatty acid desaturase of marine microalgae such as Glossomastix chrysoplasta, Thalassiosira pseudonana, and Phaeodactylum tricornutum. The gene was expressed in Saccharomyces cerevisiae INVScl to verify the substrate specificity of NAN OC-D6D. Our results suggest that the recombinant NANOC-D6D simultaneously desaturates linoleic acid (LA) and a-linolenic acid (ALA). 展开更多

A Web Service Framework for Environmental and Carbon Footprint Monitoring in Construction Supply Chains

With the growing environmental concerns, green supply chain management (GSCM) is gaining significant attention in the construction industry. Tracking and monitoring the environmental effects brought forth by the participating members along a supply chain is important to GSCM. The GreenSCOR model developed by the Supply Chain Council provides a generic framework for measuring the total carbon footprint and environmental footprint in a supply chain. The model is based on the Supply Chain Operations Reference (SCOR) model, which represents a supply chain network in a hierarchically structured manner. This paper describes the GreenSCOR framework and its potential application to the construction industry. This paper also presents a web services approach to incorporate the GreenSCOR model to the implementation of collaborative information systems. Each process element in the SCOR model is represented and delivered as individual web service units, which can be reused and integrated using standard web services technologies. The service units are combined and managed in a prototype web service collaborative framework, called SC Collaborator, which is designed and developed for supporting construction supply chain management. An illustrative example is presented to demonstrate the implementation of the GreenSCOR-based SC Collaborator framework. 展开更多

RNA silencing is a conserved mechanism found ubiquitously in eukaryotic organisms.It has been used to regulate gene expression and development.In addition,RNA silencing serves as an important mechanism in plants' defense against invasive nucleic acids,such as viruses,transposons,and transgenes.As a counter-defense,most plants,and some animal viruses,encode RNA silencing suppressors to interfere at one or several points of the silencing pathway.In this study,we showed that Pns12 of RGDV (Rice gall dwarf virus) exhibits silencing suppressor activity on the reporter green fluorescent protein in transgenic Nicotiana benthamiana line 16c.Pns12 of RGDV suppressed local silencing induced by sense RNA but had no effect on that induced by dsRNA.Expression of Pns12 also enhanced Potato virus X pathogenicity in N.benthamiana.Collectively,these results suggested that RGDV Pns12 functions as a virus suppressor of RNA silencing,which might target an upstream step of dsRNA formation in the RNA silencing pathway.Furthermore,we showed that Pns12 is localized mainly in the nucleus of N.benthamiana leaf cells. 展开更多
