The analysis of the results showed that during the breakdown process of laminar-turbulent transition, the modification of the mean flow profile by the disturbances led to a remarkable change in its stability characteristics, manifested in the significant enlargement of the linear unstable zone and the maximum amplification rate, and led to that many more disturbances were excited and enhanced rapidly, correspondingly the turbulent energy also increased rapidly, and the mean flow profiles evolved swiftly from laminar to turbulent. It was also found that if the oblique waves in the initial disturbances did not form symmetrical pairs, the subsequent span-wise mean velocities would, in general, be nonzero due to nonlinear interaction, which would have a great effect on the stability characteristics and also implied that the turbulence obtained by direct numerical simulation might not be fully a random process.展开更多

Methods: The core region of fused hirudin was the C-terminal tail of hirudin(hirudin_ 53-64),which could bind to the anion binding exosite (ABE) of thrombin.Arg-Pro-Pro-Gly-Phe(RPPGF) amino acid sequence,a metabolite of bradykinin, was added to the N-terminus of hirudin_ 53-64.It bound to the active site of thrombin.Additionally,Arg-Gly-Asp(RGD)amino acid sequence,an inibitor of glycoprotein Ⅱb/Ⅲa( GP Ⅱb/Ⅲa) receptor, was linked to C-terminus of hirudin_ 53-64.This 26-animo acid-fused hirudin peptide was artificially synthesized, purified and analysed. Results: Fused hirudin peptide significantly lengthened the activated partial thromboplastin time(APTT),thrombin time(TT)and prothrombin time(PT) and inhibited the amidolytic activity of thrombin. The ADP-induced platelet aggregation was markedly inhibited by fused hirudin peptide. Conclusion: Fused hirudin peptide has activity of antithrombin as well as antiplatelet. Therefore bifunctional anticoagulation peptide has capacity to target various components of haemostatic process and may become more powerful antithrombosis agent.展开更多

Based on the axis-symmetric FEM model of the GMA driving magnetic field was computed. And the field distribution for different input currents and variant curves of magnetic flux density along the axis were determined by using FEM. Magnetic flux density of the GMM actuator was practically measured under different input currents. The experiment of output displacement and frequency response of the GMM actuator was carried out under typical working conditions. The experiment results show that the GMA for nozzle flapper servo valve has bigger output displacement and quick response speed. And theoretical basis was presented to further introduce the GMA nozzle flapper valve into two stage electro-hydraulic servooo valve.展开更多

The tension of the colonic smooth muscle was examined by smooth muscle spontaneous contractile experiments with both ends of the smooth muscle strip tied with a silk thread. Intracellular recordings were used to assess electrical field stimulation(EFS)-induced inhibitory junction potentials(IJP) on the colonic smooth muscle. Western blot analysis was used to examine the expression levels of ICCs and PDGFRα in the colonic smooth muscle.RESULTS Treatment with NG-nitro-L-arginine methyl ester hydrochloride(L-NAME) significantly increased the CMMC frequency and spontaneous contractions, especially in the proximal colon, while treatment with MRS2500 increased only distal CMMC activity and smooth muscle contractions. Both CMMCs and spontaneous contractions were markedly inhibited by NPPB, especially in the proximal colon. Accordingly, CyPPA sharply inhibited the distal contraction of both CMMCs and spontaneous contractions. Additionally, the amplitude of stimulationinduced nitric oxide(NO)/ICC-dependent slow IJPs(sIJPs) by intracellular recordings from the smooth muscles in the proximal colon was larger than that in the distal colon, while the amplitude of electric field stimulationinduced purinergic/PDGFRα-dependent fast IJPs(fIJPs) in the distal colon was larger than that in the proximal colon. Consistently, protein expression levels of c-Kit and anoctamin-1(ANO1) in the proximal colon were much higher, while protein expression levels of PDGFRα and small conductance calcium-activated potassium channel 3(SK3) in the distal colon were much higher.CONCLUSION The ICCs are mainly distributed in the proximal colon and there are more PDGFRα+ cells are in the distal colon, which generates a pressure gradient between the two ends of the colon to propel the feces to the anus.展开更多

The steel sheeting supports the wet concrete of a cast-in-situ reinforced or post-tensioned concrete slab and, after the concrete sets, acts as external reinforcement. In this type of slab, longitudinal shear failure between the concrete and the steel sheeting is the most common type of failure at the ultimate load stage. Design codes require the experimental evaluation of the longitudinal shear capacity of each type of steel decking using full-scale tests. This paper presents the results of the short-term testing up to failure of two types of profiled steel decking that are commonly used in the construction industry in Australia. Fourteen full-scale, simply-supported slabs were tested in four-point bending with shear spans of either span/4 or span/6. Four slabs were tested at age of 28 days and the other 10 slabs were subjected to drying shrinkage and various levels of sustained loads for a period of at least 6 months prior to testing to failure. The effects of creep and drying shrinkage on the load carrying capacity and deformation of the slabs at ultimate loads are presented and discussed. The bond-slip relationship of each slab is determined from the test data and the values of maximum longitudinal shear stress calculated using different methods are described and compared.展开更多
