目的 :通过骨髓细胞移植对心肌梗死大鼠模型血管内皮生长因子及其受体表达的影响 ,探讨骨髓细胞移植改善缺血心脏功能的可能机制。方法 :利用左前降支冠状动脉结扎术制备大鼠心肌梗死模型 ,然后行异体骨髓细胞移植 ;分别于术后 1d ,3d ,... 目的 :通过骨髓细胞移植对心肌梗死大鼠模型血管内皮生长因子及其受体表达的影响 ,探讨骨髓细胞移植改善缺血心脏功能的可能机制。方法 :利用左前降支冠状动脉结扎术制备大鼠心肌梗死模型 ,然后行异体骨髓细胞移植 ;分别于术后 1d ,3d ,7d ,14d和 2 8d取材 ;利用免疫荧光和RT -PCR技术分析细胞移植对血管内皮生长因子 (VEGF)及其受体 (Flk 1)表达的影响和移植细胞的分化情况 ;通过免疫组化计算血管数量 ;应用血流动力学检测大鼠心脏功能在术后各时间点的变化。结果 :VEGF和Flk 1在移植组动物的心肌梗死区残存细胞、梗死周边区细胞以及部分移植细胞内表达。移植组VEGF和Flk 1的mRNA表达明显高于对照组 ,分别于术后 3d和 14d达到高峰 ,以后逐渐减弱。术后 7d ,14d和 2 8d移植组血管数量较同期对照组明显增加 ,移植组心脏功能较对照组明显改善。术后 14d可在部分移植细胞中检测到心肌细胞或血管内皮细胞特异性蛋白的表达。结论 :骨髓细胞移植通过上调移植细胞及受者内源性VEGF和Flk 1的表达 ,促进血管新生 ,进而改善缺血心脏功能。 展开更多

目的 探讨高龄和青年大鼠自体骨髓间质细胞移植是否能改善心功能。方法 从高龄和青年大鼠中获取骨髓间质细胞 ,部分细胞培养后克隆计数 ,其余培养时加以诱导。冠状动脉结扎后制作同源大鼠心肌梗死模型 ,3周后分为高龄细胞移植组、青... 目的 探讨高龄和青年大鼠自体骨髓间质细胞移植是否能改善心功能。方法 从高龄和青年大鼠中获取骨髓间质细胞 ,部分细胞培养后克隆计数 ,其余培养时加以诱导。冠状动脉结扎后制作同源大鼠心肌梗死模型 ,3周后分为高龄细胞移植组、青年细胞移植组和对照组。 5周后再次行心脏超声检查 ,以及检查细胞存活和疤痕区血管增生情况。结果 与青年组相比 ,高龄组骨髓间质干细胞数目以及体外诱导后结蛋白表达率明显下降。移植的高龄细胞可以在疤痕区存活 ,诱导血管新生 ,但未发现心功能改善。结论 高龄大鼠骨髓间质细胞移植未能发现可以改善梗死心肌的心功能 ,可能由于高龄骨髓间质细胞缺乏足够的干细胞数目和较低的诱导分化率。 展开更多

The aim of this study is... Cardiogenic shock is the leading cause of death among patients hospitalized with acute myocardial infarction (MI). Understanding the mechanisms for acute pump failure is therefore important. The aim of this study is to examine in an acute MI dog model whether mitochondrial bio-energetic function within non-ischemic wall regions are associated with pump failure. Anterior MI was produced in dogs via ligation of left anterior descending (LAD) coronary artery, that resulted in an infract size of about 30% of the left ventricular wall. Measurements ofhemodynamic status, mitochondrial function, free radical production and mitochondrial uncoupling protein 3 (UCP3) expression were determined over 24 h period. Hemodynamic measurements revealed a 〉 50% reduction in cardiac output at 24 h post infarction when compared to baseline. Biopsy samples were obtained from the posterior non-ischemic wall during acute infarction. ADP/O ratios for isolated mitochondria from non-ischemic myocardium at 6 h and 24 h were decreased when compared to the ADP/O ratios within the same samples with and without palmitic acid (PA). GTP inhibition of (PA)-stimulated state 4 respiration in isolated mitochondria from the non-ischemic wall increased by 7% and 33% at 6 h and 24 h post-infarction respectively when compared to sham and pre-infarction samples. This would suggest that the mitochondria are uncoupled and this is supported by an associated increase in UCP3 expression observed on western blots from these same biopsy samples. Blood samples from the coronary sinus measured by electron paramagnetic resonance (EPR) methods showed an increase in reactive oxygen species (ROS) over baseline at 6 h and 24 h post-infarction. In conclusion, mitochondrial bio-energetic ADP/O ratios as a result of acute infarction are abnormal within the non-ischemic wall. Mitochondria appear to be energetically uncoupled and this is associated with declining pump function. Free radical production may be associated with the induction of uncoupling proteins in the mitochondria. 展开更多

Subjective: To observe the effect of electroacupuncture (EA) of acupoints of the Heart Meridian and Lung Meridian on ischemic cardiac systolic ability for analyzing the relative specific relationship between the Heart Meridian and the heart. Methods: Acute myocardial ischemia (AMI) was produced by intravenous infusion of pituitrin (40 u + 5% glucose injection 500 ml, 60 drips/min) in the rabbit. Left intraventricular pressure (LVP), maximal rising velocity of LVP (dp/dt max), isovolumetric pressure (IP) and end-diastolic pressure (EDP) of the left cardiac ventricle were used as the indexes. Three points of Heart Meridian [HM, from 'Shenmen' (HT 7) to 'Lingdao' (HT 4)] and the three points of Lung Meridian [LM, from 'Taiyuan' (LU 9) to 'Lieque' (LU 7)] were punctured with filiform needles and stimulated with hand-manipulation and electrically with ZY2-1 EA Therapeutic Apparatus. 30 rabbits anesthetized with urethane (1 g/kg) were randomly and evenly divided into control group, HM group and LM group. Result-s: The effects of EA of HM points were evidently superior to those of EA of LM points in promoting the recovery of both AMI-induced decrease of LVP and dp/dtmax, and AMI-induced increase of IP and EDP. Conclusion: Acupoints of Heart Meridian has a relatively specific connection with the heart in comparison with those of Lung Meridian; and the Heart Meridian is a functional whole. 展开更多
