Occurrence and Antibiotic Susceptibility Profile of Streptococcus spp. Isolated from Ewe's Milk

The purpose of this study was to determine the occurrence and antimicrobial resistance of Streptococcus spp. isolated from fresh ewe's milk and consequently to draw conclusions concerning the health condition of the animal flocks and the current trends of antibiotic use in small ruminants. A total of 77 mountainous sheepfolds were investigated representing 10% of all transhumant sheep and goats flocks in the area. Duplicate milk samples were collected from January to February transferred to the laboratory and analyzed for the presence of Streptococcus spp. by using the serial dilution method and plating on the surface of Slanetz and Bartley agar. The isolates were tested against nine antimicrobial agents with the Kirby-Bauer disk diffusion method. The results showed that Streptococcus spp. occurred in 84.4% of the samples and in counts ranged between 1.8 log CFU/mL and 4.6 log CFU/mL. Concerning susceptibility, only 1.3% of the isolates were resistant to all antimicrobials while 27.3% were fully susceptible. Resistance to cefepime was the most common (93.1%) observation followed by resistance to ampicillin (82.1%), ciprofloxacine (58.3%) and meropenem (51.9%). The isolates were vancomycin susceptible at a rate of 83.6%. Multidrug resistance to at least three antibiotics was observed to be 22.1% of Streptococcus spp.. Given the prevalence of Streptococcus spp. and the elevated resistance of isolates to various antibiotics it is concluded that further examinations and even interventions are required in order to pertain the appropriate animal health and milk quality. 展开更多

Twenty-two lactic acid bacteria (LAB) strains isolated from Argentinean goat dairy products were evaluated for its biochemical properties and esterase activities relevant to flavor development. Streptococcus thermophilus (UNSE314), Lactobacillus (L.) delbrueckii subsp, bulgaricus (UNSE309), L. rhamnosus (UNSE308), L. plantarum (UNSE287, UNSE316, UNSE317) and Pediococcus pentosaceus (UNSE315) strains presented high acidifying activity. All strains tested metabolized citrate and produced diacetyl-acetoin in goat milk. Based on these results, 10 strains with the best performance in diverse technological properties were selected to determine esterolytic activity. In all evaluated strains, esterase specific activity (ESA) was detected on ct-naphthyl (ct-NA) acetate and 13-naphthyl ([3-NA) acetate, propionate, eaprylate and ct-NA butyrate. No activity was detected on [3-NA laurate. The highest values were detected when using a-NA instead of fI-NA derivatives as substrate. In Pediocoecus strains, wide variability in ESA were observed, which were species- and strain-specific. These results allow us to select strains with biochemical properties and esterase activities to design starter and adjunct cultures that contribute to flavor development during cheese ripening, thus preserving the typical organoleptic characteristics of Argentinean goat cheeses. 展开更多

Ghrelin O-acyltransferase(GOAT), a member of MBOATs family, is essential for octanoylation of ghrelin, which is required for active ghrelin to bind with and activate its receptor. GOAT is expressed mainly in the stomach, pancreas and hypothalamus. Levels of GOAT are altered by energy status. GOAT contains 11 transmembrane helices and one reentrant loop. Its invariant residue His-338 and conserved Asn-307 are located in the endoplasmic reticulum lumen and cytosol respectively. GOAT contributes to the regulation of food intake and energy expenditure, as well as glucose and lipids homeostasis. Deletion of GOAT blocks the acylation of ghrelin leading to subsequent impairment in energy homeostasis and survival when mice are challenged with high energy diet or severe caloric restriction. GO-Co A-Tat, a peptide GOAT inhibitor, attenuates acyl-ghrelin production and prevents weight gain induced by a medium-chain triglycerides-rich high fat diet. Further, GO-Co A-Tat increases glucose-induced insulin secretion. Overall, inhibition of GOAT is a novel strategy for treatment of obesity and related metabolic disorders. 展开更多
