Methods TGFβ1 gene was obtained by polymerase chain reactions (PCR). The upstream of TGFβ1 contained restriction enzyme site of EcoR Ⅰ, and the restriction enzyme site of Sal Ⅰ was at the downstream of TGFβ1. Using the multiple cloning sites (MCS) in plasmid AAV and the corresponding contained restriction enzyme site in PCR product of TGFβ1, TGFβ1 gene was subcloned into AAV. The recombinant plasmid AAV-TGFβ1 was detected by restriction enzyme digestion and DNA sequencing. Then, AAV-TGFβ1 virus was packaged and TGFβ1 expression mediated by AAV was detected by immunofluence analysis in H293 cells. AAV transfection rate to NP cells was evaluated with AAV-PEGF. After NP cells were respectively transfected by AAV-TGFβ1 virus and AV-TGFβ1 virus, proteoglycan synthesis was detected and compared by using Antonopulos methods. Results DNA sequencing revealed that the PCR-amplified TGFβ1 gene was consistent with NCBI Gene Bank. The recombinant plasmid was proved to be constructed successfully by restriction enzyme digestion. AAV could be transfected into NP cells and mediate an efficient expression of TGFβ1 protein. AV-TGFβ1 virus could quickly enhance the proteoglycan synthesis of the NP cells, but its biological effect was transient. AAV-TGFβ1 virus could enhance stably proteoglycan synthesis. Conclusion AAV-TGFβ1 virus was successful constructed and enhanced stably proteoglycan synthesis of NP cells.展开更多

And the ecological function of nutrient salts such as nitrogen (mg/L), phosphorus (rag/L) as well as silicon (rag/L) in different consistency on the population growth of H. circularisquama was investigated. The results showed that overtop or too low consistency of nutrient salts both delayed the population multiplication of the red tide algae, and different nutrient factors as nitrogen, phosphorus and silicon all had significant effects on the population amplification of H.circularisquama (p〈0.01). The optimal concentration of nitrogen, phosphorus and silicon in the culture medium for the algae propagation and growth were 10 mg/L, 1-5 mg/L and 0 mg/L respectively.展开更多
Current arable land and increasing food demand necessitates the practice of double and multiple cropping systems with inclusion of ultra-fast maize hybrids, which are characterized by smaller size, fewer leaves per pl...Current arable land and increasing food demand necessitates the practice of double and multiple cropping systems with inclusion of ultra-fast maize hybrids, which are characterized by smaller size, fewer leaves per plant, lower leaf area and fewer self-shading problems, under irrigation. In this context, a field experiment was conducted for two successive cropping seasons 2008/2009 to 2009/2010 at Kenilworth Experimental Station to evaluate the effect of row spacings and plant density on growth. Three row spacing (0.225, 0.45 and 0.90 m) and five plant densities (5, 7.5, 10, 12.5 and 15 plants m^-2) were used. Treatments were combined in a factorial combination and laid out in a completely randomized design with replications consisting of five single plants randomly selected from each treatment for destructive sampling. Growth factors reacted differently to row spacing by plant density. At crop establishment, growth indicators were not significantly affected by either main effects or a combination thereof. However, at the end of the vegetative phase, almost all growth indicators reached a maximum and were significantly affected by treatment interactions. Growth analysis showed that there was an interaction effect of row spacing by plant density on plant height, dry matter (DM) accumulation, leaf area index (LAI), crop growth rate (CGR) and net assimilation rate (NAR) of maize. OBJECTIVE:To investigate the clinical effects of Weining Granules on gastric precancerous lesions(GPLs).METHODS:120 patients with GPLs were randomly assigned in a 1:1 ratio to receive Weining Granules(trial group) or ...OBJECTIVE:To investigate the clinical effects of Weining Granules on gastric precancerous lesions(GPLs).METHODS:120 patients with GPLs were randomly assigned in a 1:1 ratio to receive Weining Granules(trial group) or the comparator Weifuchun tablets(control group) for 6 months.Outcomes were compared between the two groups including:overall response;gastroscopically-determined response;pathologically-confirmed response;eradication of Helicobacter pylori(HP);microvessel density(MVD) in the gastric mucosa;expression of vascular endothelial growth factor(VEGF);interleukin 2(IL-2);interleukin 6(IL-6);T lymphocyte subsets;immunoglobulins;symptom scores;quality of life(QOL);and adverse reactions.RESULTS:Patients in the trial group had a significantly higher(P<0.05) overall response rate(81.7%) as compared with those in the control group(63.3%).Relative to treatment with Weifuchun tablets treatment with Weining Granules resulted in a significant improvement(P<0.05) in the scores for gastric pain,distension and stuffiness in the hypochondrium,and anorexia.As compared with the tablets the Granules were associated with a significantly higher overall gastroscopically-determined response rate(78.3%;P<0.05).Pathological examination of tissue samples indicated that 61.7% of patients receiving the granules were cured with an overall response rate of 75.5%;these rates were significantly higher than in the control group(P<0.05).In comparison with patients receiving the tablets,those given the granules were significantly more likely to have their HP eradicated(75.0% vs.51.4%;P<0.05).Improvements in MVD,VEGF,CD4+,CD4+/CD8+,IL-2,IL-6 and IgG were significantly greater with the Weining Granules than with the Weifuchun tablets(P<0.05 or P<0.01).After follow-up of 1 year,17.5% of patients in the trial group relapsed as compared with 39.5% in the control group(P<0.05).Relative to the control group,the trial group showed significantly greater improvements in physical,psychological and social relationships,and in environmental domains(P<0.05 or P<0.01).No significant adverse reactions were observed during treatment.CONCLUSION:The Weining Granules appear to be effective in improving the gastric precancerous state and the main symptoms,in inhibiting angiogenesis,enhancing immune function and QOL,and in reducing 1-year relapses.In addition,this preparation seems to be associated with a low risk of adverse events,making it a safe and efficacious option for the treatment of GPLs.展开更多
