These migrants are most likely to be caught in attending to the needs of their children or their elderly parents, both requiring support. Over a period of two years, 35 older Polish migrants were interviewed and they told their stories of the migration experience. It was found that they had to make choices and were faced with quite difficult decisions on whether they were going to leave children or parents in Poland while they were forced into a position where they had to find work outside their own country due to age discrimination. Both men and women expressed feelings of guilt at leaving sick parents, and at not being able to afford to travel back to Poland more regularly to be with their families. Without effective cost or free digital technology, these families would not be able to sustain communication channels and relationships would break down. Consequently both the economic and emotional costs to these families and the communities in which they live could be very high.展开更多

This study investig...Objectives: The burn repair associated with diabetes complications showed considerable changes in the healing process and the development of alternative therapies to favor the repair is important. This study investigated the efficacy of the Casearia sylvestris in the burn repair in diabetic and non-diabetic rats. Methods: The animals were divided into four groups (n = 20): (C) non-diabetics treated with carbopol gel; (G) non-diabetic treated with C. sylvestris extract in carbopol gel; (DM-C) diabetics treated with carbopol gel; (DM-G) diabetics treated with C. sylvestris extract in carbopol gel. Burn was induced using a metal plate (2 cm diameter/120 °C/20 sec). Samples were collected on the 3rd, 7th, 14th and 21st days after the injury for histomorphometric analysis (inflammatory infiltrate, fibroblasts, blood vessels and collagen), MPO (Myeloperoxidase) and NAG (N-Acetilglicosaminidase) dosage, hydroxyproline, glycosaminoglycans, TGF-β1, VEGF, collagen I and III. Key findings: C. sylvestris decreased the inflammatory process during the experimental period in diabetic and non-diabetic animals which has been demonstrated by MPO, NAG quantification, and TGF-β1 expression. C. sylvestris also increased blood vessels and decreased VEGF expression during the studied period in non-diabetic animals. Fibroplasia and collagenase were increased in all experimental periods in the DM-G group by means of de hydroxyproline quantification, collagen I, collagen III and glycosaminoglycans. Conclusions: C. sylvestris application can modulate favorably the time of inflammation and collagenous process in the repair of burn injuries in diabetic rats.展开更多

Therefore, clarifying the mechani...Clinical therapies of pluripotent stem cells (PSCs)-based transplantation have been hindered by frequent development of terato- mas or tumors in animal models and clinical patients. Therefore, clarifying the mechanism of carcinogenesis in stem cell therapy is of great importance for reducing the risk of tumorigenicity. Here we differentiate Oct4-GFP mouse embryonic stem cells (mESCs) into neural progenitor cells (NPCs) and find that a minority of Oct4+ cells are continuously sustained at Oct4+ state. These cells can be enriched and proliferated in a standard ESC medium. Interestingly, the differentiation potential of these enriched cells is tightly restricted with much higher tumorigenic activity, which are thus defined as differentiation-resistant ESCs (DR-ESCs). Transcriptomic and epigenomic analyses show that DR-ESCs are characterized by primordial germ cell-like gene sig- natures (Dazl, Rec8, Stro8, BUmp1, etc.) and specific epigenetic patterns distinct from mESCs. Moreover, the DR-ESCs possess germ cell potential to generate Sycp3+ haploid cells and are able to reside in sperm-free spermaduct induced by busulfan. Finally, we find that TGFβ signaling is overactivated in DR-ESCs, and inhibition of TGFβ signaling eliminates the tumorigenicity of mESC-derived NPCs by inducing the full differentiation of DR-ESCs. These data demonstrate that these TGFβ-hyperactivated germ ceU-like DR-ESCs are the main contributor for the tumorigenicity of ESCs-derived target cell therapy and that inhibition of TGFβ signaling in ESC-derived NPC transplantation could drastically reduce the risk of tumor development.
