Brassica campestris ssp. chinensis, also known as non-heading Chinese cabbage, is an important vegetable widely distributed in southern China. High temperature is the most common adversity factor in vegetable production, because Brassica campestris ssp. chinensis is a thermophilic vegetable, which can't well grow at high temperature. In summer and autumn, high temperature stress would prevent Brassica campestris ssp. chinensis from growing and result in poor quality of its seedlings, seriously influencing yield and quality of the vegetable in later period. In this paper, present situation and latest advances in heat resistance research of Brassica campestris ssp. chinensis in recent years were reviewed at home and abroad. The research tendency in Bassica campestris ssp. chinensis was also described, providing reference for breeding of heat-resistant Brassica campestris ssp. chinensis. 展开更多

[ Objective] The study was aimed to determine the biological characteristics of Rhabditis (Oscheius) sp. isolated from a rhizosphere soil of alfalfa field in Hailar City, Hulunbuir League, Inner Mongolia Autonomous Region. [ Method] The last instar larvae of Galleria mellonella and Tenebrio molitor were used as insect hosts. The infectivity of nematode was determined, and its heat tolerance ability was measured by using water bath method. [Result] The infectivity of the nematodes was lower at the dosages of 10, 20 and 40 IJs/larva, but higher at the dosages of 80,160, 32.0 and 640 IJs/larva. The corrected mortality (CM) of G. mellonella at 96 h and T. molitor at 120 h were both above 90%. The nematode infectivity was highest at the dosage of 80 IJs/larva when the hosts' CM reached 100% after 120 h. About 25% nematode individuals could survive in 36℃ water bath for 2 d in the densities of 1 000 -5 000 IJ/ml, after 6 d, there were still 3% -9% individuals survived, until 9 d the nematodes died totally. A hundred percent of the nematode individuals died in 38℃ and 40 ~C water bath after 6 h and 2 h, respectively. [Conclusion] Rhabditis ( O. ) sp. has a higher potential to cause the host mortality and represents heat tolerance up to 36℃. 展开更多

Effects of High Temperature on Activities of Some Protective Enzymes in Mononychellus tanajoa

[Objective] Mononychellus tanajoa is a mite speices recently invaded into China in 2008. Temperature is one of the most important ecological factors affecting the growth and reproduction of M. tanajoa. The objective of the current study was to reveal the effects of high temperature incubation on the activities of some protective enzymes in M. tanajoa at different growth stages. The results would contribute to the understanding of the adaptable distribution of M. tanajoa after its invasion into China, the mechanisms in its invasion, diffusion and ecological adaptation, and the monitor- ing, early warning and effective prevention of its damage. [Method] Six protective enzymes, Le. polyphenol oxidase (PPO), peroxidase (POD), ascorbate oxidase (AsA- POD), catalase (CAT), superoxide dismutase (SOD) and esterase (EST), were cho- sen to study their activities after the mites at different growth stages were incubated at a extremely high temperature of 42 ~C for a certain period of time up to 24 h. The activities were measured by spectrophotometric endpoint assay method. [Results] Enzyme activities in M. tanajoa were affected by the high temperature incubation. However, differences in enzyme activity changes were found among different protec- tive enzymes and among different growth stages of M. tanajoa. Activities of PPO, POD, AsA-POD and CAT were significantly increased in the larval mites and female adult mites of M. tanajoa. CAT activity was significantly decreased in protonymph and deutonymph of M. tanajoa. Activities of PPO, POD and AsA-POD in protonymph and deutonymph showed no obvious difference from the control. [Conclusion] The activity changes of some protective enzymes in M. tanajoa following high-temperature treatment are part of its anti-stress reaction mechanism. In mite protonymph and deutonymph, activities of PPO, POD and AsA-POD are similar to the untreated con- trol which may be associated with the thermostability of M. tanajoa. It is concluded that, the long-time stress of extreme temperature may result in the increase of the thermostability of mite individuals, the enhancement of the population thermal stability and subsequently lead to rapid expansion of the population. 展开更多

With tissue cultured-seedlings of Rhododendron chrysanthum Pall. in Changbai Mountain as test material, the effect of EMS treatment on the heat toler- ance of R. chrysanthum, as well as the possible heat tolerance mechanism, was in- vestigated in this study so as to lay a foundation for the popularization, application and germplasm resource innovation of R. chrysanthum. The results showed that when the leaves of the tissue cultured seedlings were treated with medium with 0.1% of EMS for 2 d, the survival rate of the leaves was 48.92%, thus the con- centration of EMS was close to the semi-lethal dose, and the EMS concentration and treatment time could be used as screening conditions for heat-tolerant mutants. The plantlets regenerated from EMS-treated leaves were treated with different tem- peratures, and related physiological indicators were determined. The results showed that the chlorophyll content, MDA content, Pro content, SOD activity, POD activity and soluble sugar content of regeneration plantlets in the treatment group were sig- nificantly different from those in the control group. At the same temperature, the chlorophyll content, soluble sugar content, Pro content, SOD activity and POD ac- tivity were significantly higher and the MDA content was significantly lower in the treatment group compared with the control. In conclusion, EMS treatment improved the heat tolerance of tissue-cultured R. chrysanthum seedlings. 展开更多
