This article focuses on the current underlying of molecular mechanisms of the peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) mediated pathway and discuss possible therapeutic benefits of inc...This article focuses on the current underlying of molecular mechanisms of the peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) mediated pathway and discuss possible therapeutic benefits of increased mitochondrial biogenesis in compensating for mitochondrial dysfunction and ameliorating aging and aging-related diseases. PGC-1α is the master transcription regulator that stimulates mitochondrial biogenesis, by upregulating nuclear respiratory factors and mitochondrial transcription factor A, leading to increased mitochondrial DNA replication and gene transcription. PGC-1α also regulates cellular oxidant-antioxidant homeostasis by stimulating the gene expression of superoxide dismutase-2, catalase, glutathione peroxidase 1, and uncoupling protein. Recent reports from muscle-specific PGC-1α overexpression underline the benefit of PGC-1α in muscle atrophy and sarcopenia, during which PGC-1α enhanced mitochondrial biogenic pathway and reduced oxidative damage. Thus, PGC-1α seems to have a protective role against aging associated skeletal muscle deterioration.展开更多

A total of 640...This experiment was conducted at the poultry farm, poultry research station, state board of agriculture research, ministry of agriculture to study the effect of supplemental anise seed on broiler diets. A total of 640 of one-day old unsexed broiler chicks (Cobb) were used in this study. They were randomly distributed to four treatments. Anise seeds were supplemented at the levels of 0.2%, 0.4% and 0.6% compared with the control group for 49 days. The results showed: there was a significant (P 〈 0.05) increase in the rate of body weight for the two treatments 0.6% and 0.4% of the anise seeds compared with 0.2% and control treatments, body weight gain take the same direction, like in body weight, the highest treatment was 0.6% followed by treatment 0.4% as well as the feed consumption has increased in these treatments than in 0.2% and control treatments, so about feed conversion ratio the treatment 0.6% of the anise seeds recorded the best feed conversion ratio compared with other treatments. Significant decreased in mortality and significant increased in production index by increasing the level of anise seeds in feed. Dressing percentage increased significantly for all treatments using anise seeds and treatment 0.6% recorded highest dressing percentage, from the other hand percentage of abdominal fat decreased significantly by increasing anise seed levels and no significant difference in the percentages of liver, gizzard and heart were recorded in this study. About carcass cuts, the weights of primary cuts (thigh, drum stick, breast) relative to body weight recorded significant increase in the treatments using anise seeds at the expense of secondary cuts (neck, wings, back). There was a significant decline (P 〈 0.05) for total bacterial (E. coli and Staphylococci) in small intestine in all level of anise seeds (0.2%, 0.4%, 0.6%) in ration while the decline of two types of bacteria in colon recorded in 0.4% and 0.6% treatments compared with 0.2~/0 and control treatments. Conclude from this study the possibility of using anise seeds at all levels used in the experiment and the best level that can be used to get the best performance of the positive results of broiler chickens is 0.6%.展开更多

Moreover, the accumulation of damaged mitochondria is...Mitochondrial dysfunction in aged skeletal muscle has been mainly attributable to mtDNA deletions, uncoupling in oxidative phosphorylation, leading to ATP deficit. Moreover, the accumulation of damaged mitochondria is associated with imperfect autophagocytosis and declined biogenesis of efficient mitochondria. Mitochondria-associated apoptotic signaling is involved in the loss of fibers and atrophy of the remaining fibers by ROS, TNF-α, and denervation, leading to sarcopenia. Current investigations have indicated that exercise, especially resistance exercise, has extensively preventive effects on sarcopenia. Whether caloric restriction in aged animals is suitable for aged people remains to be further clinlically experimented. Additionally, it will be interesting to develop effectively supplementary and alternative medical methods associated with Chinese tonifying herbs to prevent or reverse sarcopenia in the future because of their efficacy and less frequent side effects.展开更多

In this study, biotransformation of 1 was investig...Malabaricone C (1), isolated from the seeds ofMyristicafragrans Houtt., belongs to a kind of diarylnonanoid compounds that are only found in Myristicaceae till now. In this study, biotransformation of 1 was investigated using rat hepatic microsomes for the first time and the main biotransformation product was elucidated as malabaricone B (2) according to the spectroscopic data. Further evaluation on human gastric cancer cell lines showed that the cytotoxic effects of malabaricone C and its metabolite malabaricone B were comparable to those of vinorelbine, with the values of IC50 of (42.62±3.10) and (19.80±1.70) μg/mL on NCI-N87, and (22.94±1.33) and (19.60±2.21) μg/mL on MGC803, respectively. Statistical analysis revealed that malabaricone B had significantly stronger cytotoxicity than the parent compound (P〈0.01 on NCI-N87 and P〈0.05 on MGC803), which may indicate a bioactivation of malabaricone C by hepatic microsomes. These results suggest that malabaricone C has a simple biotransformation pathway by hepatic microsomes and provide valuable information for further investigation on both the parent compound and its biotransformation product as anti-gastric cancer agents or lead compounds.展开更多
