From d 1 of pregnancy (E0) until postnatal d 20 (P20), maternal mice were fed experimental diets that contained 1 mg Zn/kg/day (severe zinc deficient, SZD), 5 mg Zn/kg/day (marginal zinc deficient, MZD), 30 mg Zn/kg/day (zinc adequately supplied, ZA) or 100 mg Zn/kg/day (zinc supplemented, ZS and pair-fed, PF). Brains of offspring from these dietary groups were examined at various developmental stages for expression of nestin, an intermediate filament protein found in neural stem cells and young neurons. Immunocytochemistry showed nestin expression in neural tube 10.5 d post citrus (dpc) as well as in the cerebral cortex and neural tube from 10.5 dpc to postnatal d 10 (P10). Nestin immunoreactivities in both brain and neural tube of those zinc-supplemented control groups (ZA, ZS, PF) were stronger than those in zinc-deficient groups (SZD and MZD). Western blot analysis confirmed that nestin levels in pooled brain extracts from each of the zinc-supplemented groups (ZA, ZS, PF) were much higher than those from the zinc-deficient groups (SZD and MZD) from 10.5 dpc to P10. Immunostaining and Western blots showed no detectable nestin in any of the experimental and control group brains after P20. These observations of an association between maternal zinc deficiency and decreased nestin protein levels in brains of offspring suggest that zinc deficiency suppresses development of neural stem cells, an effect which may lead to neuroanatomical and behavioral abnormalities in adults.展开更多
The semisolid slurry of the A356 aluminum alloy was prepared by self-inoculation method(SIM),the effects of melt treatment temperatures and isothermal holding parameters on water-quenched microstructures of A356 alumi...The semisolid slurry of the A356 aluminum alloy was prepared by self-inoculation method(SIM),the effects of melt treatment temperatures and isothermal holding parameters on water-quenched microstructures of A356 aluminum alloy semisolid slurry were investigated,and the solidification behavior of the remaining liquid phase(secondary solidification)was analyzed.The results indicate that the melt treatment temperature has significant effects on the final semisolid microstructures.The semisolid slurry which is suitable for the rheological forming can be produced when the melt treatment temperature is between 680 and 690°C.During the isothermal holding process,the growth rate of the primary particles conforms to the dynamic equation of Dt 3-Do3=Kt,and the coarsening rate of the primary particles is the fastest when the isothermal holding temperature is 600°C.Additionally,the isothermal holding time also has obvious effect on the secondary solidification microstructures.The secondary particles are the smallest and roundest when the isothermal holding time is 3 min.The amount of the secondary particles gradually increases with the increase of isothermal holding temperature,and the eutectic reaction therefore is confined into small intergranular areas,contributing to the compactness of the final solidified eutectic structures.展开更多

Fert...Broodstock of Verasper moseri (Jordan and Gilbert) aged 3-4 years old were selected, and reinforced cultivation was conducted to promote maturation under controlled water temperature and photoperiod conditions. Fertilized eggs were obtained by artificial fertilization, and the development of embryos, larvae and juveniles was observed continuously. The results showed that the fertilized eggs of V. moseri were spherical, with transparent yolk and homogeneous bioplasm, and had no oil globule inside. The average diameter of the eggs was 1.77+0.02 mm. The eggs of V. moseri were buoyant in water with salinity above 35. The cleavage type was typical discoidal. Young pigment cells appeared when olfactory plates began to form. Hatching occurred at 187 h after fertilization at a water temperature of 8.5℃. The newly hatched larvae, floating on the water surface, were transparent with an average total length of 4.69±0.15 mm. During the cultivation period, when the water temperature was raised from 9 to 14.5℃, 4-day old larvae showed more melanophores on the body surface, making the larvae gray in color. The pectoral fins began to develop, which enabled the larvae to swim horizontally and in a lively manner. On days 7-8, the digestive duct formed. The yolk sac was small and black. The yolk sac was absorbed on day 11. Larvae took food actively, and body length and body height clearly increased. The rudiments of dorsal and anal fin pterygiophores were discernible and caudal fin ray elements formed on day 19. On day 24, the larval notochord flexed upwards, and the rays of unpaired fins began to differentiate. Pigment cells converged on the dorsal and anal fin rays, and the mastoid teeth on the mandible appeared. On day 29, the left eyes of juveniles began to move upwards. Depigmentation began in some juveniles and they became sandy brown in color on day 37. Most juveniles began to settle on the bottom of the tank. The left eyes of juveniles migrated completely to the right side on day 50, when the average body length attained 2.5±0.18 cm, and juveniles accomplished metamorphosis to young.
