MethodBased on the mechanisms of liquid impinger and filtration sampler, a double concentration aerosol sampler was designed for collecting Mhp aerosol. Firstly, the collection was performed in a closed environment full of artificial aerosol of Mhp. Secondly, collection efficiency was detected by real-time PCR. Thereafter, the clinical feasibility of the designed equipment was tested by collecting aerosol samples in different pig herds. In one assay, the samples were collected at different times from one pig house challenged with Mhp. In another assay, the samples was collected from the delivery room, nursery and fattenning house of a MPS outbreak farm as well as a Mhp infection positive pig farm without obvious clinical symptoms. All the aerosol samples were then detected by real-time PCR or nested PCR. ResultThe collection efficiency of the designed bioaerosol sampler was (37.04±6.43) %, Mhp could be detected 7 d after intratracheal challenge with pneumonic lung homogenate suspension. Aerosol samples of 11 pig houses from the two Mhp positive pig farms with or without clinical symptoms all showed a positive result of PCR, the positivity rate was 100%. ConclusionA high sensitive collecting and detecting technology of aerosol was successfully established, which can be applied to clinical detection of Mhp in aerosol.展开更多
Pulmonary arterial pressure (PAP) was increased obviously in rats after 3 days of normobaric hypoxic exposure and reached a maximum at 14 days of hypoxia. It remained at the same level during prolonged hypoxic expo...Pulmonary arterial pressure (PAP) was increased obviously in rats after 3 days of normobaric hypoxic exposure and reached a maximum at 14 days of hypoxia. It remained at the same level during prolonged hypoxic exposure of up to 21 days. Right ventricular weight (RV/LV +S) and hydroxyproline (HP) content in the pulmonary artery began to increase at day 7. HP content had increased much faster than the relative rate of increase of PAP after 14 days, but HP content in the thoracic aorta showed no change. The relative proportion of type Ⅰto Ⅲ collagen increased singnificantly, and compliance of the pulmonary vessels obviously decreased. All parameters returned to the normal range within 14 days after recovery from hypoxia, except for HP content as expressed per vessel. 764-3 treatment obviously attenuated most of the changes caused by hypoxia, though it had no effect on compliance of the pulmonary vessels. It is suggested that collagen, especially type Ⅰcollagen, accumulation may play an important role in maintaining pulmonary hypertension. 764-3 has certain protective effects and may be useful in the treatment of early chronic obstructive pulmonary disease.展开更多

It is believed that macrophages in areas of chronically inflamed lung play a key role in fibrotic response. Therefore, we investigated the effects of alveolar macrophage (AmΦ) conditioned media from interstitial lung disease (ILD) patients on lung fibroblast proliferation and procollagen mRNA expression. After stimulating with AmΦ conditioned media from ILD patients, the fibroblast proliferation increased 71. 4 % compared with the control, but for media from bronchial carcinoma (BC) patients, it just increased 14. 3%. There is a significant difference between the two groups (P<0. 05). The procollagen a1 (Ⅰ) mRNA in fibroblasts stimulated with AmΦ conditioned media from ILD patients was increased 21. 3 %, and a1 ( Ⅲ) was 37. 2% higher than control (P<0.05). It increased 6. 8% and 12. 8% for media from BC patients respectively, but there was no difference when compared to the control.
