复方痛风康对高尿酸血症模型大鼠肾功能的保护作用及影响IL-1β、TNF-α、TGF-β_1的研究 被引量:17

目的:探讨以清热利湿化瘀散结法创制的中药复方痛风康对高尿酸血症模型大鼠血尿酸、肾功能、肾组织病理改变的影响,以及对肾脏组织中白细胞介素1(IL-1β)、肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)表达的影响。方法:将雄性S... 目的:探讨以清热利湿化瘀散结法创制的中药复方痛风康对高尿酸血症模型大鼠血尿酸、肾功能、肾组织病理改变的影响,以及对肾脏组织中白细胞介素1(IL-1β)、肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)表达的影响。方法:将雄性SD大鼠随机分为空白组、模型组、苯溴马隆组及复方痛风康高、中、低剂量组,共6组。除空白组外,其余各组均以腺嘌呤和酵母膏灌胃,同时各给药组灌胃给予相应的药物,空白组和模型组给予等体积蒸馏水,连续21 d。末次给药后,测定大鼠血清尿酸(UA)、尿素氮(BUN)、肌酐(Scr)水平;放射免疫法测定大鼠肾脏组织IL-1β、TNF-α、TGF-β1的含量;制作肾组织病理切片,光镜下观察肾组织超微病理学的改变。结果:模型组动物血清UA、BUN、Scr水平升高,与空白组、苯溴马隆组比较,差异有统计学意义(P<0.05)。痛风康高剂量组UA含量明显降低,与空白组比较,差异有统计学意义(P<0.01)。用药各组与模型组相比BUN、Scr含量均明显降低,差异有统计学意义(P<0.05)。模型组肾组织IL-1β、TNF-α、TGF-β1含量与空白组比较,显著升高(P<0.01)。苯溴马隆组、痛风康高、中、低剂量组大鼠肾组织IL-1β、TNF-α、TGF-β1含量较模型组均有所降低。其中痛风康高剂量组大鼠肾组织IL-1β、TNF-α含量与模型组相比,差异有统计学意义(P<0.01,P<0.05)。肾脏病理变化以模型组损伤最为明显,各给药组均有不同程度的改善。结论:以清热利湿化瘀散结法创制的中药复方痛风康,在降低高尿酸血症大鼠血清尿酸的同时,可降低炎性因子含量,促使抗炎症因子的产生,从而减轻肾组织炎性反应,对肾脏功能和形态的损伤具有一定的保护作用。 展开更多

LDP术治疗UPJO肾积水患儿的疗效及对肾功能的影响

目的探讨腹腔镜离断式肾盂输尿管成形术(LDP)治疗肾盂输尿管连接部梗阻(UPJO)肾积水患儿的疗效及对肾功能的影响。方法回顾性选取2019年1月~2022年1月就诊于宁夏回族自治区人民医院的85例UPJO肾积水患儿,按照手术方法分为观察组(43例,给... 目的探讨腹腔镜离断式肾盂输尿管成形术(LDP)治疗肾盂输尿管连接部梗阻(UPJO)肾积水患儿的疗效及对肾功能的影响。方法回顾性选取2019年1月~2022年1月就诊于宁夏回族自治区人民医院的85例UPJO肾积水患儿,按照手术方法分为观察组(43例,给予LDP)和对照组(42例,给予开放肾盂成形术)。对比两组手术指标(手术时间、住院费用、术中出血量、肾周引流总量、术后镇痛剂用量、住院时间)、肾功能、肾形态和并发症发生情况。结果观察组术中手术时间和住院费用与对照组比较,显著增多(P<0.05);术中出血量、肾周引流总量、术后镇痛剂用量和住院时间与对照组比较,均显著减少(P<0.05)。术后两组肾小球滤过率(GFR)较术前均显著升高,血肌酐(Scr)水平较术前均显著降低(P<0.05)。但术后两组GFR和Scr水平比较,差异无统计学意义(P>0.05)。观察组术后并发症发生率显著低于对照组(P<0.05)。术后两组肾体积均较术前显著减小(P<0.05),肾实质厚度均较术前显著升高(P<0.05)。但两组肾体积和肾实质厚度比较,差异无统计学意义(P>0.05)。结论LDP术治疗UPJO肾积水患儿的疗效显著,促进肾功能、肾脏形态恢复,减少术后并发症。 展开更多

Effect of heme oxygenase-1 on renal function in rats with liver cirrhosis 被引量:8

AIM: To investigate the role of heme oxygenase-1 (HO-1) in pathogenesis of experimental hepatorenal syndrome (HRS). METHODS: Rats were divided into liver cirrhotic group, zinc protoporphyrin IX (ZnPP) treatment group, cobalt protoporphyrin (CoPP) treatment group and sham group. Biliary cirrhosis was established by bile duct ligation in the first three groups. Rats in the ZnPP and CoPP treatment groups received intraperitoneal injection of ZnPP and CoPP, respectively, 24 h before sample collection. Expression of HO-1 mRNA in kidney was detected by reverse-transcription polymerase chain reaction, while protein expression was determined by immunohis-tochemical analysis. Hematoxylin and eosin staining was performed to observe liver cirrhosis and renal structure. Renal artery blood flow, mean arterial pressure and portal vein pressure, 24 h total urinary volume, serum and urine sodium concentrations, and creatinine clearance rate (Ccr) were also measured.RESULTS: The HO-1 mRNA and protein expression levels in kidney, 24 h total urinary volume, renal artery blood flow, serum and urine sodium concentration and Ccr were lower in cirrhotic group than in sham group (P < 0.05). However, they were significantly lower in ZnPP treatment group than in cirrhotic group and significantly higher in CoPP treatment group than in cirrhotic group (P < 0.05). CONCLUSION: Low HO-1 expression level in kidney is an important factor for experimental HRS. 展开更多

Objective:To investigate effects of Zibu Shenjing Fang(滋补肾精方) on growth and development of the mouse with insufficiency of kidney-essence and the mechanism.Methods:Total 50 mice were randomly divided into a normal group,a model group,a Jingui Shenqi Wan(金匮肾气丸) group,a Zibu Shenjing Fang high dose group and a Zibu Shenjing Fang low dose group,10 mice in each group.The kidney-essence insufficiency mouse model was established by use of threat-injuring the kidney combined with over-fatigue.At the same time of modeling,the mice in the model group were intragastrically administrated with saline 20 mL·kg-1·d-1,in the Jingui Shenqi Wan group with suspension of the Jingui Shenqi Wan 2.7 g·kg-1·d-1,in the Zibu Shenjing Fang high dose group with Zibu Shenjing Fang 20 g·kg-1·d-1 and in the Zibu Shenjing Fang low dose group with Zibu Shenjing Fang 10 g·kg-1·d-1,for 21 consecutive days.The general state was observed,the body weight was weighted,and serum growth hormone(GH) and insulin-like growth factor-1(IGF-1) contents were detected.Results:Compared with model group,Zibu Shenjing Fang groups and Jingui Shenqi Wan group could improve manifestation of the mouse with kidney-essence insufficiency,increase body weight of the mouse and serum GH and IGF-1 contents,especially in the high dose group.Conclusion:Zibu Shenjing Fang gives play to the function of tonifying the kidney and replenishing essence through regulating GH and IGF-1 levels,so as to influence growth and development of the mouse. 展开更多
