Methods Eb was administered to Kunming mice (dosage, 0.1 g·kg^(-1)·d^(-1)) intragastrically for 7 successive days. The contents of selenium in heart, liver, spleen, kidneys, lungs, stomach, brain, muscle, and bone were determined by fluorometric method on the eighth day. MTT assay was used to study tumor growth inhibition of Eb in vitro, and lymphocyte transformation, hemolysin formation and phagocytosis assay were used to study its immunocompetence. Results After 7 days′ administration of Eb, the tissue contents of sele-(nium) in liver, spleen, lungs, kidneys, and bone of mice increased, especially those in liver and spleen increased significan-tly, compared with controls; but no significant changes of such contents were found in muscle, heart, brain, and stomach. Eb demonstrated inhibitory effects on human Bel-7402, BGC-823, and Calu-3 cancer cell lines in vitro. Eb also showed ability to enhance lymphocyte transformation and serum hemolysin formation in vitro and increase the phagocytosis of macrophages. Conclusion The validated antitumor and immunostimulatory activities of Eb suggest a hypothesis that Eb may behave as a biological response modifier when used as an antitumor agent. Eb is worthy of further study in developing a new antineoplastic and immunity enhancing agent in the light of its antitumor activity, immunocompetence and specific distribution in liver, lungs, kidneys, bone, and spleen.展开更多

At that time few researches on it have been conducted due to less popularity of the products even among the researching field. Since then some of the bio-functional works of the black garlic gradually appeared along with gaining of its popularity. Representative bio-activities reported up to now are: (1) enhancement of anti-tumor potency in the mouse tumor model; (2) immune system enforcement, and suppressive act against allergic symptoms caused by decreasing IL-4 cytokine production; (3) bacteria killing potency; (4) anti-oxidant activity; (5) reduction of carcinogenic potency; (6) lowering cholesterol level and preventive work against obesity; (7) controlling high blood pressure; (8) beneficial effect against diabetics; (9) regenerative effect to the burned skin (Clinical study case) et al. On the black garlic business (marketing), the Aomori Black Garlic Union (Association) had been established in Aomori Prefecture to offer the citizen high quality, high safety and high security black garlic (products) aiming at contribution for health improvement. All union members are now exerting themselves to the most to attain the high quality production of the black garlic. The quality proving certification seal will be issued to the products that passed the quality approval test by the third party inspections to place it on the their products. Initiation and progression of hepatocellular carcinoma (HCC) is intimately associated with a chronically diseased liver tissue. This diseased liver tissue background is a drastically different microenvironment from ...Initiation and progression of hepatocellular carcinoma (HCC) is intimately associated with a chronically diseased liver tissue. This diseased liver tissue background is a drastically different microenvironment from the healthy liver, especially with regard to immune cell prevalence and presence of mediators of immune function. To better understand the consequences of liver disease on tumor growth and the interplay with its mi- croenvironment, we utilized two standard methods of fi- brosis induction and orthotopic implantation of tumors into the inflamed and fibrotic liver to mimic the liver condition in human HCC patients. Compared to non-diseased con- trols, tumor growth was significantly enhanced under fi- brotic conditions. The immune cells that infiltrated the tumors were also drastically different, with decreased numbers of natural killer cells but greatly increased num- bers of immune-suppressive CDllb^+ Gr1^hi myeloid cells in both models of fibrosis. In addition, there were model- specific differences: Increased numbers of CD11b^+ mye- loid cells and CD4^+ CD25^+ T cells were found in tumors in the bile duct ligation model but not in the carbon te- trachloride model. Induction of fibrosis altered the cytokine production of implanted tumor cells, which could have far- reaching consequences on the immune infiltrate and its functionality. Taken together, this work demonstrates that the combination of fibrosis induction with orthotopic tumor implantation results in a markedly different tumor mi- croenvironment and tumor growth kinetics, emphasizing the necessity for more accurate modeling of HCC pro- gression in mice, which takes into account the drastic changes in the tissue caused by chronic liver disease.展开更多
