Some of them were kept more than 15 years. Results Cell livability rate arrived at 80%~90% after anabiosis. 通过离子对反相高效液相色谱 (IP RPLC)同时检测细胞内的核糖核苷三磷酸及脱氧核糖核苷三磷酸现已成为可能 ,采用这种方法检测了 16种正常及肿瘤细胞内的ADP、CTP、dCTP、GTP、UTP、dGTP、dTTP、ATP、dATP等核苷酸的含量 ,并进行了简要... 通过离子对反相高效液相色谱 (IP RPLC)同时检测细胞内的核糖核苷三磷酸及脱氧核糖核苷三磷酸现已成为可能 ,采用这种方法检测了 16种正常及肿瘤细胞内的ADP、CTP、dCTP、GTP、UTP、dGTP、dTTP、ATP、dATP等核苷酸的含量 ,并进行了简要的对比及方法学验证。采用 6 %三氯乙酸分离提取样品 ,并用 5mol l碳酸钾中和后进行HPLC分析。采用Waters 6 0 0E 4 86色谱系统 ,SymmetryC1 8(3 5 μm ,4 6× 15 0mm)分析柱及Nova -PakC1 8SentryGuardColumn进行色谱分析 ,柱温 2 7℃。流动相由两种缓冲液组成 ,采用梯度洗脱 ,流速 1 0ml min。缓冲液A包含 10mmol l氢氧化四丁基胺 ,10mmol lKH2 PO4及 0 2 5 %甲醇 ,并用HCl调pH至 6 9;缓冲液B包括 5 6mmol l氢氧化四丁基胺 ,5 0mmol lKH2 PO4及 30 %甲醇 ,并用NaOH调pH至 7 0。流动相开始时为 6 0 %A及 40 %B ,30min达 40 %A及 6 0 %B ,此比例一直持续到 6 0min。采用各标准品的水溶液作标准曲线 (r均大于 0 99)。各物质测定的精密度 (CV %)平均为日内 0 9%,日间 5 0 %。各物质的检测限 (pmol)分别为 1 39(ADP)、4 32(CTP)、15 5 (dCTP)、2 38(GTP)、4 4 2 (UTP)、9 4 5 (dGTP)、14 6 (dTTP)、2 4 4(ATP)、11 8(dATP)。该测定方法的平均回收率亦可达到 99 5 % 展开更多

Objective To report a case of the implantation of thyroid hyperplastic or neoplastic tissue after endoscopic thyroidectomy and discuss this complication in aspects of prevalence, pathogenesis, protection, and therapies. Methods A systematic search of literature from the PubMed database was conducted for identifying eligible studies on implantation of thyroid hyperplastic or neoplastic cells after endoscopic thyroid surgery. Results Overall, 5 reported cases on patients suffering from endoscopic thyroid surgery with implantation of thyroid hyperplastic or neoplastic cells were included in the systematic review. Conclusions Unskilled surgeons, rough intraoperative surgical treatment, scarification or rupture of tumor, contamination of instruments, chimney effect, aerosolization of tumor cells may be associated with the implantation after endoscopic thyroidectomy. To minimize the risk of such complication, we should be more meticulous and strict the endoscopic surgery indications. 展开更多

Objective: The aim of this study was to use lung cancer targeting binding polypeptide ZS-9 to screen cDNA library of human lung cancer and obtain ZS-9 specific ligand to confirm tumor marker of non small-cell lung cancer. Methods: Artificially synthesize biotin labeled peptide ZS-9, anchored ZS-9 in the enzyme label plate coupled by avidin, used ZS-9 as probe to screen cDNA library of human lung cancer, after screening, obtained bacteriophage clone specifically binding with anchored polypeptide ZS-9. Extracted plasmid of bacteriophage and performed sequencing after amplified by PCR. Results: It was demonstrated by bioinformatic analysis on the sequence of ligand binded by lung cancer specific peptide ZS-9 that the ligand was the cytoskeletal protein periplakin on the surface of lung cancer cells, suggesting that periplakin might be a new marker for non-small-cell lung cancer in lung cancer. Conclusion: Use specific lung cancer binding peptide to screen new tumor marker periplakin in lung cancer and further studies on its biologic functions in genesis and development of lung cancer are still needed. 展开更多
