Methods: Detecting proteins of p130Cas, PTEN and PTEN mRNA ...Objective: To research the contributions of p130Cas and PTEN signal molecules to the carcinogenesis of gastric carcinoma and the relationship between them. Methods: Detecting proteins of p130Cas, PTEN and PTEN mRNA of 76 cases normal gastric mucosa and 112 cases gastric carcinoma by immunohistochemistry EnVision method and molecular hybridization in situ method respectively. Detecting PTEN genetic mutation of 30 cases normal gastric mucosa, 7 cases early gastric cancer and 30 cases progressive gastric cancer by PCR-SSCP. Results: The expression of p130Cas protein of gastric carcinoma increased significantly than that of normal gastric mucosa (P 〈 0.05). Opposite to above, the expression of PTEN protein of gastric carcinoma group was significantly lower than that of normal gastric rnucosa group (P 〈 0.05). The expression of PTEN mRNA of gastric carcinoma group decreased obviously than normal gastric mucosa group (P 〈 0.001). Only one case exon 5 and one case exon 8 of PTEN appeared gene mutation of progressive gastric carcinoma group, the difference has no significance compared with normal gastric mucosa group and early gastric cancer group. Conclusion: The signaling molecules p130Cas and PTEN play an important role in the carcinogenesis of gastric carcinoma, and p130Cas plays the part of promoter, oppositely, maybe PTEN can inhibit it.展开更多
Schwannoma is a well-described,benign nerve sheath tumor of the soft tissue,but is rare in the gastrointestinal tract.Gastrointestinal schwannomas are often incidentally discovered as small polypoid intraluminal lesio...Schwannoma is a well-described,benign nerve sheath tumor of the soft tissue,but is rare in the gastrointestinal tract.Gastrointestinal schwannomas are often incidentally discovered as small polypoid intraluminal lesions.In this report,we describe the clinicopathologic and immunohistochemical features of a distinctive neural mucosal polyp composed of a diffuse cellular proliferation of uniform bland spindled cells in the lamina propria that entraps the colonic crypts.Immunohistochemical analysis revealed strong and diffuse positivity for the S-100 protein.To avoid confusion of these solitary colorectal polyps containing pure spindled Schwann cell proliferation in the lamina propria with neural lesions that have significant association with inherited syndromes,it is better to use the designation "mucosal Schwann hamartoma".展开更多

Methods: Gastric mucosa samples were obtained from antrum of 33...Objective: To study the relationship between nitric oxide synthase (NOS) expression in human gastric mucosa and Helicobacter pylori (H.pylori) infection. Methods: Gastric mucosa samples were obtained from antrum of 33 patients received gastroendoscopy. H.pylori infection was confirmed by Giems staining and bacteria culture under microaerophilic conditions. Expression of iNOS, eNOS and nitrotyrosine were detected by immunohistochemistry. Results: (1) The positive rate of H. pylori infection was 66.7%(22/33). (2) iNOS positive staining in inflammatory cells was detected in 77.3%(17/22) of samples with H.pylori and 27.3%(3/11) without H.pylori infection (P<0.01). (3) eNOS expression in inflammatory cells was found in 77.3%(17/22) of samples with H. pylori and 18.2%(2/11) without H.pylori infection (P<0.01). (4) Nitrotyrosine expression in inflammatory cells was observed in 59.1%(13/22) of samples with H. pylori and 54.5%(6/11) without H. pylori infection (P>0.05). (5) Moderate and severe infiltrations of inflammatory cells were found in 86.4%(19/22) of gastric biopsies with H. pylori and 9.1%(1/11) of samples without H. pylori infection (P<0.01). Conclusion: H.pylori infection might promote infiltration of mononuclear cells and macrophages in gastric mucosa and induce iNOS expression in these cells. The accumulated nitric oxide in local area may result in gastric mucosa damage.展开更多
