印迹基因UBE3A在人类卵母细胞及植入前胚胎的mRNA表达 被引量:1

目的 了解印迹基因泛素蛋白酶基因(UBE3A)在人类卵母细胞和植入前胚胎的表达状况。方法采用单细胞巢式RT-PCR,检测UBE3A在人类卵母细胞和植入前各个阶段胚胎中的mRNA表达。结果UBE3A在人类卵母细胞MⅠ、MⅡ和植入前胚胎2、4、6、8细... 目的 了解印迹基因泛素蛋白酶基因(UBE3A)在人类卵母细胞和植入前胚胎的表达状况。方法采用单细胞巢式RT-PCR,检测UBE3A在人类卵母细胞和植入前各个阶段胚胎中的mRNA表达。结果UBE3A在人类卵母细胞MⅠ、MⅡ和植入前胚胎2、4、6、8细胞期均有表达,单卵母细胞扩增成功率为83.3%,单个卵裂球细胞为72.3%,平均扩增成功率为77.9%。单个卵母细胞和单个卵裂球扩增成功率无显著性差别(P〉0.05),但单卵母细胞较单个卵裂球高。结论UBE3A在人类卵细胞和植入前胚胎均有表达,有助于研究卵母细胞成熟及胚胎生长发育,为探讨印迹基因与辅助生殖及遗传性印迹疾病的关系提供理论依据。 展开更多
