The properties of the proton pumping activity of H+-ATPases in tonoplast vesicles were studied by acridine orange fluorescent quenching measured at 22 degreesC. The proton pumping activity of ATPase was ATP-dependent with apparent Michaelis-Menten Constant (K-m) for ATP about 0.65 mmol/L. The optimal pH for H+-ATPases activity was 7.5. The proton pumping activity of H+-ATPase could be initiated by some divalent cations, Mg2+ being highly efficient, much more than Fe2+; and Ca2+, Cu2+ and Zn2+ were inefficient under the experimental condition. The proton translocation could be stimulated by halide anions, with potencies decreasing in the order Cl- > Br- > I- > F-. The proton pumping activity was greatly inhibited by N-ethylmaleimide (NEM), N, N'-dicyclohexylcarbodiimide (DCCD), NO3- and Bafilomycin A(1), but not by orthovanadate and azide. These results demonstrated that the H+-ATPase in the tonoplast of Populus euphratica belonged to vacuolar type ATPase. This work was the first time that tonoplast-enriched vesicles were isolated from Populus euphratica cells.展开更多
The taxonomic status of long-tailed field mouse (Apodemus orestes) is still uncertain up to now.Some scholars regarded it as a subspecies or a synonym of dragon field mouse (A.draco),while other scholars considered it...The taxonomic status of long-tailed field mouse (Apodemus orestes) is still uncertain up to now.Some scholars regarded it as a subspecies or a synonym of dragon field mouse (A.draco),while other scholars considered it a valid species.In the study,five specimens of adult long-tailed field mouse,dragon field mouse and Chevrier's field mouse (A.chevrieri) (as a contrast) were chosen respectively.After treated,with the eyepiece micrometer in the inverted microscope,we made measurements of the widths of hairs and medullas and then calculated the medullary indexes of straight guard hairs from around the mouth,head,back,abdomen and forelimb of each species.The results indicate that,viewing at each of the five parts and the mixture of the five parts,there are no significant differences between A.orestes and A.draco,Significant differences can be found between A.orestes & A.chevrieri and A.draco & A.chevrieri though.The standpoint that A.orestes was listed as a valid species is not supported by this study.展开更多
A total of 31 wheat varieties(lines) released in different years in Huanghuai Area were selected as materials so as to study the carotenoids content,lipoxygenase activity, and some other quality traits of wholemeal....A total of 31 wheat varieties(lines) released in different years in Huanghuai Area were selected as materials so as to study the carotenoids content,lipoxygenase activity, and some other quality traits of wholemeal. Results showed that carotenoid content and other quality traits had highly significant differences among varieties. Carotenoids content was significantly positive correlated with watersoluble pentosan and yellowness. Carotenoids content was significantly negative correlated with peak viscosity, hold through, breakdown, final viscosity, unit weight,formation time, and was negative correlated with peak time, protein content and stability time. According to cluster analysis, carotenoid contents in 31 varieties were divided into three major groups: one group had common carotenoid content, which included 19 varieties such as Meng 0318; one group had relatively low carotenoid content, which included 10 varieties such as Luomai23; and the last group had relatively high carotenoid content, which included 2 varieties such as Huaimai22. The average carotenoid contents of the three groups were 1.93, 1.07 and 2.99 mg/kg respectively.展开更多

The main physiological profiles, such as ash free dry weight (AFDW), chlorophyll, carotenoid, protein and total fatty acids (TFA), in this alga exposed to old culture supernatant (OCS) at the decline phase or its crude ethyl acetate extracts (CEAE) were investigated by using tubular photobioreactors of different diameters. Results showed that both OCS and CEAE had strong inhibitory effect on the above physiological parameters. The longer the culture was exposed to OCS and the more CEAE were added into the algal culture, the more the above physiological properties were inhibited. Arachidonic acid (AA), the dominant component of fatty acids in this alga, was also seriously inhibited with respect to total TFA, AFDW of cell mass, or culture volume, due to a probable reduction of enzymes activities catalyzing chain elongation from C18:1ω9 to AA. These results incontestably evidenced that some CEAE dissolving substances existing in OCS, like auto inhibitors, inhibited P. incisa growth through feedback. Hence, any efficient removal of auto inhibitors from algal culture to decrease their bioactivity could be good for maximal production of desired products like AA.展开更多

The extrac...The differences in XRD patterns, elemental compositions, FT-IR spectra and TG-DSC curves of extract residues obtained by NaOH, and NaOH assisted with anthraguinone (AQ) extraction procedures were studied. The extract residues are mainly comprised of humin fractions associated with inorganic minerals. XRD analysis shows that there is no typical peak of organic carbon because those organic humin fractions appear as a highly disordered substance. The peak of quartz is dominant. The elementary analysis shows that assistant AQ in NaOH solution can break the link of organic humic substances with inorganic minerals. And aromatization degree of humin fractions obtained by NaOH is smaller than that obtained by NaOH assisted with AQ. FT-IR analysis displays that various groups exist in those two humin fractions obtained by different extraction procedures. There are some differences in FT-IR curves between two humin fractions. TG-DSC analysis shows that thermal decomposition occurs during the heating of testing samples. By contrast, the humin fractions associated with inorganic minerals obtained by NaOH possess a higher thermal decomposition range.展开更多
Lutein is a dietary carotenoid of particular nutritional interest as it is preferentially taken up by neural tissues. Often linked with beneficial effects on vision, a broader role for lutein in neuronal differentiati...Lutein is a dietary carotenoid of particular nutritional interest as it is preferentially taken up by neural tissues. Often linked with beneficial effects on vision, a broader role for lutein in neuronal differentiation has emerged recently, although the underlying mechanisms for these effects are not yet dear. The purpose of this study was to investigate the effect of lutein on neuronal differentiation and explore the associated underpinning mechanisms. We found that lutein treatment enhanced the differentiation of SH-SYSY cells, specifically increasing neuronal arborization and expression of the neuronal process filament protein microtubule-associated protein 2. This effect was mediated by the intracellular phosphoinositide-3-kinase (PI3K) signaling pathway. While PI3K activity is a known trigger of neuronal differentiation, more recently it has also been shown to modulate the metabolic state of cells. Our analysis of bioenergetics found that lutein treatment increased glucose consumption, rates of glycolysis and enhanced respiratory activity of mitochondrial complexes. Concomitantly, the generation of reactive oxygen species was increased (con- sistent with previous reports that reactive oxygen species promote neuronal differentiation), as well as the production of the key metabolic intermediate acetyl-CoA, an essential determinant of epigenetic status in the cell. We suggest that lutein-stimulated neuronal differentiation is mediated by PI3K-dependent modulation of mitochondrial respiration and signaling, and that the consequential metabolic shifts initiate epigenetically dependent transcriptomic reprogramming in support of this morphogenesis. These obser- vations support the potential importance of micronutrients supplementation to neurogenesis, both during normal development and in regenerative repair.展开更多

[Method] By employing SDS-PAGE and 2-demensional electrophoresis (2-DE) techniques,proteins in various heteromorphic leaves from the same adult tree of P. euphratica were isolated and separated to the electrophoresis technique suitable for the separation and analysis of proteins in leaves of P. euphratica tree. [Results] There were significant differences in the expressions of proteins in various heteromorphic leaves of P. euphratica tree. SDS-PAGE pattern showed that bands of proteins with molecular weight of 57.2,13.2,30.2,23.9 and 33.3 kDa were remarkably different. 2-D electrophoresis pattern presented that proteins in leaves of P. euphratica tree mainly belong to acidic proteins distributed at pH value of 5.0-6.5 and with molecular weight of 20-40 kDa; totally 73 different protein spots were observed,of which 51 were up expressed and other 22 were down expressed in the serrated ovate leaves. [Conclusion] Based on these results,we speculate that regulated gene expression in leaves of P. euphratica tree results in the generation of different shapes of leaves,in order to adapt to the surroundings better.展开更多

The crop is mostly consumed, processed or sold. Extending shelf-life of roots is crucial for Malawi and Northern Ghana as the crop has high value. Trials were conducted in the countries at the community level. In the dry season, temperature is cool in Malawi while warm in Ghana, but thru harmattan, the night is cool with low relative humidity. In Malawi, orange-fleshed sweetpotato Zondeni var., white and yellow types were assessed in three types of storage, Afghan ventilated pit store, storage in dry sand of pit-steps, and of a granary In Ghana, local moistened heap and sandbox were compared. In Malawi, weight losses were calculated relative to the quantity stored at start, it was not cumulative. At 1.5 months no significant difference was among treatments. By 3.5 months the pit-steps method emerged to be superior and continued to 6.5 months. Losses in granary were due to shriveling, in the pit-stepsdue to termites and rats, and in ventilated pit due to termites, rats and Java black rot. Sprouting was high in pit-steps, but it was simply removed and roots returned to storage. At 6.5 months, the beta-carotene of Zondeni roots was traceable. Farmers gained high price when selling them as roots were scarce. Women favored the pit-steps because it was manageable. In Ghana, the sandbox was superior to local moistened heap. Methods designed were suitable for home consumption, but will require modification for commercialization.展开更多

Studies were conducted on the preservative effects of four plant extracts--Xylopia aethiopica, Piper nigrum, Tetrapleura tetraptera and Carica papaya seeds on tomato and pepper. Tomato and pepper were dipped into four different concentrations of aqueous extracts of the four plants as well as water serving as control. The result of the study showed that aqueous extract of all plants extended the shelf life of tomato and pepper with increasing concentration of the extracts, except for Carica papaya seeds which produced significant elongation of 12.0 ±0.6 d and 14.0 ± 0.7 d on tomato and pepper, respectively, at the least concentration of 0.25 mg/mL. The results showed that these ethnomedicinal plants can serve as good and potent sources of natural preservatives which can be used to substitute existing synthetics used in the food industry.展开更多

This study aimed to characterize physicochemical quality and minerals in the mesocarp (pulp) of grugru palm, Grugru palm fruits from NE Brazil production area were collected and analyzed. The determinations included: respiratory rate, sugars, soluble solids, pH, titratable acidity, humidity, water activity (aw), ashes, protein, lipids, total phenolics, total carotenoids, vitamin C, color and minerals. The fruit respiration rate was 74.45 mL CO2 kg-1 h1. High levels were found for sugars, soluble solids, pH, aw, lipids and carotenoids; however, the pulp showed low acidity, moisture and ashes. The values found for lipids, minerals and vitamin C are equivalent to those observed in the literature. The grugru palm fruit has a high rate of respiration, which turns necessary the use of conservation techniques and methods. The grugru palm fruit pulp shows physical quality parameters attractive to the consumer, physicochemical quality and nutritive composition which can be considered for food ingredients and alternative source of ingredient for supplementation of diet food for needy population.展开更多

Data on physical characteristics such as weight, ...A research was conducted to assess the effects of three different drying techniques, namely open sun drying (OSD), solar dryers (SD) and solar tent dryers (STD). Data on physical characteristics such as weight, total length and standard length were recorded and proximate composition of the fish sample Clarias gariepinus (Burchell, 1822) was carried out before, during and after drying to assess changes on physical characteristics and nutrient composition at different level. After drying, the samples were subjected to storage for eight weeks. The result obtained shows that there was no significant (P 〉 0.01) difference in the drying weight of the fish samples between the direct SD and the STD. However, for the total and standard length there was significant (P 〈 0.01) difference in the drying techniques, but no significant difference (P 〉 0.05) in the drying period. There was no significant (P 〉 0.05) difference on the physical parameters, storage period, proximate composition of both direct SD and STD and the drying period, respectively. Therefore, direct SD can be substituted with the STD without altering the quality of the product.展开更多

Among treatments, ...Effect of fertilizer source (mineral fertilizer or organic manure) on photosynthetic pigments leaves content of four brassica vegetables (Pak choi, Kohlrabi, Cauliflower and Cabbage) was tested. Among treatments, cauliflower fertilized with chicken manure had the highest content of chlorophyll a and b (10.08 and 9.37 μm·g^-1), while, pak choi had the lowest values (2.00 and 2.44 μm·g^-1) in non-fertilized plants. These differences in chlorophyll a and b content may have a great impact on photosynthetic rate and activity. Total carotenoids and total xanthophylls (lutein, zeaxanthin, antheraxanthin and violaxanthin) were the highest in cauliflower while the lowest was in pakchoi, β-carotene, on the other hand, was the highest percentage of total carotenoids (65.7%) in pak choi while Kohlrabi had the lowest percent (57.4%). This high percentage offl-carotene of total carotenoids may compensate for lower content of chlorophylls and assist for efficient light harvesting process. Lutein showed to be the major components of xanthophyll pigments brassica vegetables and ranged from 0.462μm·g^-1 in cabbage to 0.626μm·g^-1 in kohlrabi but not differed significantly which may refer to genetically controlled. However, lutein, antheraxanthin and zeaxanthin content were increased significantly by the application of chicken manure regardless ofbrassica vegetable type. Violaxanthin was only detected in pak choi and not affected by fertilizer source. It can be concluding from the increasing contents of total xanthophylls pigments that may increase photosynthetic efficiency through non-photochemical quenching process.展开更多

Royle twigs were exposed to CdCI2 (50, 100, 200 and 500 BM) under continuous light for 48 hrs and the physiological parameters like photosynthetic pigment (chlorophylls a, b and carotenoids) contents, activities of antioxidative enzymes like superoxide dismutase (SOD), catalase (CAT) and peroxidase (POX) along with lipid peroxidation level were determined. With respect to increase in Cd concentration in the medium and exposure duration, decrease in pigment contents, and decrease in the activities of SOD, CAT and POX were found. The increased levels of lipid peroxidation indicated the prevalence of oxidative stress situation in the tissues which might be one of the reasons behind Cd induced toxicity in Hydrilla verticillata. Since there was significant decrease in the activities of key antioxidative ezymes, the study suggests that Hydrilla verticillata may not be effective for phytoremediation of cadmium in these concentration ranges.展开更多

We suggest that lutein-stimulated neuronal differentiation is mediated by PI3K-dependent modulation of mitochondrial respiration and signaling, and that the consequential metabolic shifts initiate epigenetically dependent transcriptomic reprogramming in support of this morphogenesis. These obser- vations support the potential importance of micronutrients supplementation to neurogenesis, both during normal development and in regenerative repair.
文摘[Objective] This study was to elucidate the cellular and molecular mechanism of the development of heteromorphic leaves of Populus euphratica Oliv. [Method] By employing SDS-PAGE and 2-demensional electrophoresis (2-DE) techniques,proteins in various heteromorphic leaves from the same adult tree of P. euphratica were isolated and separated to the electrophoresis technique suitable for the separation and analysis of proteins in leaves of P. euphratica tree. [Results] There were significant differences in the expressions of proteins in various heteromorphic leaves of P. euphratica tree. SDS-PAGE pattern showed that bands of proteins with molecular weight of 57.2,13.2,30.2,23.9 and 33.3 kDa were remarkably different. 2-D electrophoresis pattern presented that proteins in leaves of P. euphratica tree mainly belong to acidic proteins distributed at pH value of 5.0-6.5 and with molecular weight of 20-40 kDa; totally 73 different protein spots were observed,of which 51 were up expressed and other 22 were down expressed in the serrated ovate leaves. [Conclusion] Based on these results,we speculate that regulated gene expression in leaves of P. euphratica tree results in the generation of different shapes of leaves,in order to adapt to the surroundings better.
文摘Uni-modal rainfall pattern has long dry spell wherein sweetpotato is scarce, expensive but cheap at harvest. The crop is mostly consumed, processed or sold. Extending shelf-life of roots is crucial for Malawi and Northern Ghana as the crop has high value. Trials were conducted in the countries at the community level. In the dry season, temperature is cool in Malawi while warm in Ghana, but thru harmattan, the night is cool with low relative humidity. In Malawi, orange-fleshed sweetpotato Zondeni var., white and yellow types were assessed in three types of storage, Afghan ventilated pit store, storage in dry sand of pit-steps, and of a granary In Ghana, local moistened heap and sandbox were compared. In Malawi, weight losses were calculated relative to the quantity stored at start, it was not cumulative. At 1.5 months no significant difference was among treatments. By 3.5 months the pit-steps method emerged to be superior and continued to 6.5 months. Losses in granary were due to shriveling, in the pit-stepsdue to termites and rats, and in ventilated pit due to termites, rats and Java black rot. Sprouting was high in pit-steps, but it was simply removed and roots returned to storage. At 6.5 months, the beta-carotene of Zondeni roots was traceable. Farmers gained high price when selling them as roots were scarce. Women favored the pit-steps because it was manageable. In Ghana, the sandbox was superior to local moistened heap. Methods designed were suitable for home consumption, but will require modification for commercialization.
文摘This research aimed at the use of natural products to extend the shelf life of perishable vegetables. Studies were conducted on the preservative effects of four plant extracts--Xylopia aethiopica, Piper nigrum, Tetrapleura tetraptera and Carica papaya seeds on tomato and pepper. Tomato and pepper were dipped into four different concentrations of aqueous extracts of the four plants as well as water serving as control. The result of the study showed that aqueous extract of all plants extended the shelf life of tomato and pepper with increasing concentration of the extracts, except for Carica papaya seeds which produced significant elongation of 12.0 ±0.6 d and 14.0 ± 0.7 d on tomato and pepper, respectively, at the least concentration of 0.25 mg/mL. The results showed that these ethnomedicinal plants can serve as good and potent sources of natural preservatives which can be used to substitute existing synthetics used in the food industry.
文摘Studies are needed to know the grugru palm (Acrocomia aculeata) potential for food industry. This study aimed to characterize physicochemical quality and minerals in the mesocarp (pulp) of grugru palm, Grugru palm fruits from NE Brazil production area were collected and analyzed. The determinations included: respiratory rate, sugars, soluble solids, pH, titratable acidity, humidity, water activity (aw), ashes, protein, lipids, total phenolics, total carotenoids, vitamin C, color and minerals. The fruit respiration rate was 74.45 mL CO2 kg-1 h1. High levels were found for sugars, soluble solids, pH, aw, lipids and carotenoids; however, the pulp showed low acidity, moisture and ashes. The values found for lipids, minerals and vitamin C are equivalent to those observed in the literature. The grugru palm fruit has a high rate of respiration, which turns necessary the use of conservation techniques and methods. The grugru palm fruit pulp shows physical quality parameters attractive to the consumer, physicochemical quality and nutritive composition which can be considered for food ingredients and alternative source of ingredient for supplementation of diet food for needy population.
文摘A research was conducted to assess the effects of three different drying techniques, namely open sun drying (OSD), solar dryers (SD) and solar tent dryers (STD). Data on physical characteristics such as weight, total length and standard length were recorded and proximate composition of the fish sample Clarias gariepinus (Burchell, 1822) was carried out before, during and after drying to assess changes on physical characteristics and nutrient composition at different level. After drying, the samples were subjected to storage for eight weeks. The result obtained shows that there was no significant (P 〉 0.01) difference in the drying weight of the fish samples between the direct SD and the STD. However, for the total and standard length there was significant (P 〈 0.01) difference in the drying techniques, but no significant difference (P 〉 0.05) in the drying period. There was no significant (P 〉 0.05) difference on the physical parameters, storage period, proximate composition of both direct SD and STD and the drying period, respectively. Therefore, direct SD can be substituted with the STD without altering the quality of the product.
文摘Effect of fertilizer source (mineral fertilizer or organic manure) on photosynthetic pigments leaves content of four brassica vegetables (Pak choi, Kohlrabi, Cauliflower and Cabbage) was tested. Among treatments, cauliflower fertilized with chicken manure had the highest content of chlorophyll a and b (10.08 and 9.37 μm·g^-1), while, pak choi had the lowest values (2.00 and 2.44 μm·g^-1) in non-fertilized plants. These differences in chlorophyll a and b content may have a great impact on photosynthetic rate and activity. Total carotenoids and total xanthophylls (lutein, zeaxanthin, antheraxanthin and violaxanthin) were the highest in cauliflower while the lowest was in pakchoi, β-carotene, on the other hand, was the highest percentage of total carotenoids (65.7%) in pak choi while Kohlrabi had the lowest percent (57.4%). This high percentage offl-carotene of total carotenoids may compensate for lower content of chlorophylls and assist for efficient light harvesting process. Lutein showed to be the major components of xanthophyll pigments brassica vegetables and ranged from 0.462μm·g^-1 in cabbage to 0.626μm·g^-1 in kohlrabi but not differed significantly which may refer to genetically controlled. However, lutein, antheraxanthin and zeaxanthin content were increased significantly by the application of chicken manure regardless ofbrassica vegetable type. Violaxanthin was only detected in pak choi and not affected by fertilizer source. It can be concluding from the increasing contents of total xanthophylls pigments that may increase photosynthetic efficiency through non-photochemical quenching process.
文摘Hydrilla verticillata (I.f.) Royle twigs were exposed to CdCI2 (50, 100, 200 and 500 BM) under continuous light for 48 hrs and the physiological parameters like photosynthetic pigment (chlorophylls a, b and carotenoids) contents, activities of antioxidative enzymes like superoxide dismutase (SOD), catalase (CAT) and peroxidase (POX) along with lipid peroxidation level were determined. With respect to increase in Cd concentration in the medium and exposure duration, decrease in pigment contents, and decrease in the activities of SOD, CAT and POX were found. The increased levels of lipid peroxidation indicated the prevalence of oxidative stress situation in the tissues which might be one of the reasons behind Cd induced toxicity in Hydrilla verticillata. Since there was significant decrease in the activities of key antioxidative ezymes, the study suggests that Hydrilla verticillata may not be effective for phytoremediation of cadmium in these concentration ranges.
