Tzvel.) callus was purified by sucrose gradient centrifugation to investigate the influence of sound stimulation on the lipid physical states and metabolism of plasma membrane. The results showed that sound stimulation decreased the content of phosphodiesters and the fluorescent intensity of DPH, but increased the light scattering value of the membrane, the fluorescent intensity of MC540 and the content of phosphomonoesters, indicating that the vesicles got looser, the charge density and hydrophobicity of membrane surface decreased under sound stimulation of some strength and frequency. However, the membrane fluidity increased under the condition. Meanwhile, the anabolism of membrane lipid increased and the catabolism decreased. It can be seen that the physical state and metabolism of membrane lipid is sensitive to sound stimulation.展开更多
Objective: To investigate the proliferation capabilities, immunophenotype changes, level of secreted cytokines and activities against lymphoma cells under the condition that cytokine-induced killer (CIK) cells co-c...Objective: To investigate the proliferation capabilities, immunophenotype changes, level of secreted cytokines and activities against lymphoma cells under the condition that cytokine-induced killer (CIK) cells co-cultured with dendritic cells (DC) in vitro. Methods: DC and CIK cells were induced from peripheral blood mononuclear cells of healthy volunteers. They were co-cultured meanwhile CIK cells were cultured alone as controls. Increased number of cells were counted by tapan-blue staining, killing activities were detected by MTT assay, immunophenotype changes were analyzed by flow cytometry, the IL-12 and INF-y levels of the cultured supernatants were detected by ELISA kits. Results: The proliferation capabilities of DC-CIK cells were significantly higher than that of CIK cells (P 〈 0.05). Under the same condition, the ratio of double positive cells such as CD3^+ CD8^+, CD3^+ CD56^+ in CIK cells was significantly enhanced by co-cultured with DC cells (P 〈 0.05). The level of IL-12 and INF-y secreted in supernatants was increased noticeably by co-cultured DC-CIK cells on day 3 compared to CIK cells which were cultured alone (P 〈 0.01 and P 〈 0.05). Within the effector-target ratio range between 5:1 to 40:1, the activities against lymphoma cells of DC-CIK cells were much higher than that of CIK cells (P 〈 0.05), and this effect was showed a positive correlation with the effector-target ratio. Conclusion: The proliferation capabilities, the level of secreted cytokines and the activities against lymphoma cells of DC-CIK cells were significantly higher than those of CIK cells. The research might provides theoretical and experimental basis for clinical immunotherapy of DC-CIK cells.展开更多

METHODS: Plasmids suppressing UCP-2 expression were constructed, and transfected into fatty liver cells cultured in vitro and the ob/ob mouse I/R injury model. Serum tumor necrosis factor (TNF)-α levels, UCP-2 mRNA expression, alanine aminotransferase (ALT) levels in ob/ob mice were tested, and the pathological changes in fatty liver were observed in experimental and control groups. RESULTS: In ob/ob mouse I/R models, serum TNF-α levels were significantly higher than in normal controls. After the plasmids were transfected into the cultured cells and animal models, expression of UCP-2 rnRNA was significantly reduced as compared with that in the control group (2^1.56± 0.15 vs 2^-0.45± 0.15, p 〈 0.05). In ob/ob mouse models, in which expression of UCP-2 was suppressed, serum ALT levels were significantly lower than those of other groups, and pathological analysis revealed that injury of liver tissues was significantly alleviated. Three kinds of hyphae are considered in the model: active...A morphologically structured model is proposed to describe the batch fermentation of lovastatin according to the growth kinetics of filamentous microorganisms. Three kinds of hyphae are considered in the model: actively growing hyphae, non-growing hyphae and deactivated hyphae. Furthermore, actively growing hyphae consist of three morphological compartments: apical compartment which gives rise to hyphal tip extension; subapical compartment which is related to hyphal branching; and hyphal compartment which is only responsible for secondary metabolite formation. The kinetics of mycelial growth mechanism is summarized and applied in modeling lovastatin fermentation. A Michaelis-Menten kinetic model with substrate inhibition is proposed for product formation. As expected, the model simulations fit well with experimental data obtained either from a laboratory scale 10L fer-menter or from a pilot-plant scale fermenter.展开更多

The sample collection was based on file descripti...The aim of this study was to analyze the chemical composition of landrace seeds of rice, maize and cassava grown in traditional communities in file state of Maranchao. The sample collection was based on file description of the farmers for identification of varieties of plants, based on protein content, lipids, carbohydrates and mineral through Association of Official Analytical Chemists (AOAC) methods. Landrace seeds of nine maize cultivars, 13 rice and 40 cassava, with differences in centesimal composition in all varieties were analyzed. For maize landrace seeds file lipid values, carbohydrates and ash are in the average for the species. The protein contents and ash to file surveyed rice varieties were high, while the levels of lipids and carbohydrates are within average in relation to conventional varieties. The results obtained for cassava showed high percentages of protein, with a significant level for the feeding of the people of the community who consume their byproducts. Thus, differences in file chemical composition of file varieties studied showed file interference of genotype, management and environmental factors in their chemical and physiological quality, reinforcing file importance of file study of creole varieties for the development of new germplasm and for file guarantee of food sovereignty of traditional communities.展开更多

During the late 17th and early 18th centuries, migration of the Minangs from West Sumatra to the state of Negeri Sembilan Darul Khusus in Peninsular Malaysia took place and their descendants now form the main sub ethnic group in this state. The genetic polymorphisms of Y chromosome at DYS 287 locus were analyzed in Minangkahau Malays. Methods: A total of 41 buccal cells from healthy unrelated individual's males from Minangkabau Malays were typed for the DYS 287. The PCR products were separated on 2% (w/v) agarose gel followed by visualization under UV light. Results: Three out of 41 samples (7.32%) showed insertion (YAP+) polymorphism, while the rest of the samples (92.68%) showed deletion (YAP-) polymorphism. This is the first report concerning the YAP in Malay population at Peninsular Malysia. Conclusion: The vAluable data obtained in this study will contribute to fill in the gap in the knowledge of YAP distribution in Malaysian population and will allow continuous interpretation of the evolution of YAP.展开更多
