The present study observed neurological functional changes in a rat model of traumatic brain injury following ADMSC transplantation via the tail vein. Cell transplants were observed in injured cerebral cortex, and expression of brain-derived nerve growth factor was significantly increased in the injured hippocampus following transplantation. Results demonstrated that transvenous ADMSC transplants migrated to the injured cerebral cortex and significantly improved cognitive function.展开更多
Objective To detect the change of nerve growth factor (NGF) level in human amniotic fluid during gestation, and to explore the relationship between this change and fetal ventriculo-megaly (VM). Methods The studied sub...Objective To detect the change of nerve growth factor (NGF) level in human amniotic fluid during gestation, and to explore the relationship between this change and fetal ventriculo-megaly (VM). Methods The studied subjects (collected from 2004 to 2007) were divided into four groups, including the second-trimester pregnancy group (n=113), third-trimester pregnancy group (n=110), fetal cerebral VM group (n=12), and healthy control group (n=12) which matched with the VM group in gestational weeks. The amniotic fluid specimens were obtained during amniocentesis or cesarean section. The NGF levels in amniotic fluid were detected with en-zyme-linked immunosorbent assay. Results A significantly negative correlation was found between gestational age and the NGF level in amniotic fluid (r= 0.6149, P<0.0001). The NGF level in patients with fetal VM was significantly lower than that in healthy controls (33.95±29.24 pg/mL vs. 64.73±16.21 pg/mL, P=0.024). Conclusion NGF levels in amniotic fluid may be a sensitive marker for fetal VM.展开更多

Methods: Primary and passage choroid plexus epithelial cells were obtained from newborn, one-day Spragne-Dawley rats. The expressions of BDNF and NGF were measured by qRT-PCR and Western blottingting. The secretions of BDNF and NGF were detected by ELISA. Cell supematants of primary cells, purified cells and passage 1 cells were harvested. Results: The expression of BDNF in the purified cells was significantly lower than that in the primary cells (P〈0.05), and it in the primary cells and the purified cells was significantly higher than that in the passage 1 cells (P〈0.05). The expression of NGF was significantly higher in the purified cells than in the primary cells and the passage 1 cells (P〈0.05). It in the passage 1 cells was significantly higher than that in the primary cells (P〈0.05). Conclusion: The time of CPECs transplantation for central nervous system diseases should be selected based on their secretory function and features,which could lead to better and more effective treatment.展开更多

In this study, RT-PCR and Western Blotting were performed to quantify the regulation of endogenic nerve growth factor expression in neuroglia ceils by HCMV infection. The results showed that basal, endogenous NGF expression in U251 was unchanged during early HCMV infection. NGF expression is strongly down-regulated during the latent phase of infection. Objective: To examine the effects of rat marrow stromal cells (rMSCs) on gene expression of local brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) after injection of rMSCs into Cistern Magnum of ...Objective: To examine the effects of rat marrow stromal cells (rMSCs) on gene expression of local brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) after injection of rMSCs into Cistern Magnum of adult rats subjected to traumatic brain injury(TBI).Methods: A modified Feeney’s TBI model was created in 48 adult rats. rMSCs were harvested from 3-month-old rats, and injected into Cistern Magnum in 24 rats subjected to TBI(Group cell transplantation). Saline was given through Cistern Magnum to another 24 rats subjected to TBI (Group saline control). Animals were sacrificed 1, 2 and 3 weeks after intervention, and special brain tissue blocks were dissected for total RNA extraction from each block. BDNF and NGF mRNA were reverse-transcribed into cDNA and further expanded by polymerase chain reaction (PCR). The expression of target genes was evaluated using semi-quantitative methods. Results: Group cell transplantation had higher BDNF and NGF gene expressions than Group saline control during a period of less than 3 weeks (P< 0.05).Conclusions: rMSCs transplantation via Cistern Magnum in rats subjected to traumatic brain injury can enhance expressions of local brain NGF and BDNF to a certain extent.展开更多

ERK5 is ex...Extracellular signal-regulated protein kinase 5 (ERK5), also known as big mitogen-activated protein kinase 1 (MAPK1), is an important member of ERK family, which is a subfamily of the large MAPK family. ERK5 is expressed in many tissues, including the dorsal root ganglion (DRG) neurons and the spinal cord. In this review, we focus on elaborating ERK5-associated pathway in pathological pain, in which the ERK5/CREB (cyclic adenosine monophos- phate (cAMP)-response element-binding protein) pathway plays a crucial role in the transduction of pain signal and contributes to pain hypersensitivity. ERK5 activation in the spinal dorsal horn occurs mainly in microglia. The activation of ERK5 can be mediated by N-methyI-D-aspartate (NMDA) receptors. We also elaborate the relationship between ERK5 activation and nerve growth factor-tyrosine kinase A (NGF-TrkA), and the connection between ERK5 activation and brain-derived neurotrophic factor (BDNF) in pathological pain in detail.展开更多
Objective To observe the effect of electroacupuncture(EA)of“concurrent treatment of the brain and heart”on angiogenesis and cortical vascular endothelial growth factor(VEGF)and brain-derived neurotrophic factor(BDNF...Objective To observe the effect of electroacupuncture(EA)of“concurrent treatment of the brain and heart”on angiogenesis and cortical vascular endothelial growth factor(VEGF)and brain-derived neurotrophic factor(BDNF)in rats with focal cerebral ischemia,and to explore the mechanism of EA in cerebral ischemia treatment.Methods A total of 108 Sprague-Dawley rats,27 rats were randomly selected as the sham-operation group,and the rest rats received the right middle cerebral artery occlusion operation for model preparation firstly,and then were divided into a model group,a traditional acupoint group,and a concurrent treatment of the brain and heart group,with 27 rats in each group.In the sham-operation group,only the carotid artery was isolated.EA at Shuigou(CV26),Quchi(LI11),Hegu(LI4),and Zusanli(ST36)in the traditional acupoint group,and EA at Fengfu(GV16),Baihui(GV20),Xinshu(BL15),and Neiguan(PC6)in the concurrent treatment of the brain and heart group were performed 4 h after the operation,once a day,for 14 consecutive days.Rats in the sham-operation group and the model group were identically fixed without any treatment.Before and after treatment,the modified neurological severity score(mNSS),regional cerebral blood flow(rCBF),and CD34 positive expression by immunohistochemistry were measured.The positive protein expression levels of VEGF and BDNF were detected by immunofluorescence,and the mRNA expression levels of VEGF and BDNF were detected by quantitative reverse transcription-polymerase chain reaction(qRT-PCR).Results Compared with the sham-operation group,the mNSS,rCBF,and ischemic side cortical micro-vessel density(MVD)decreased,and the protein and mRNA expression levels of VEGF and BDNF increased in the model group(P<0.01).Compared with the model group,the mNSS of the two EA groups decreased,and the mNSS of the concurrent treatment of the brain and heart group was lower than that of the traditional acupoint group on the 14th day(P<0.05).Compared with the model group,the rCBF in the two EA groups increased,and the rCBF reached the highest on the 14th day(P<0.05 or P<0.01),and the rCBF in the concurrent treatment of the brain and heart group was higher than that in the traditional acupoint group(P<0.05);the MVD of the two EA groups was higher than that of the model group,and the MVD of the concurrent treatment of the brain and heart group was higher than that of the traditional acupoint group on the 7th and 14th days(P<0.05 or P<0.01).Compared with the model group,the protein and mRNA expression levels of VEGF and BDNF in the two EA groups increased(P<0.01).The VEGF expression level was the highest on the 7th day in the concurrent treatment of the brain and heart group(P<0.05),and the BDNF expression level was higher on the 7th and 14th days than on the 3rd day(P<0.05).The mRNA expression levels of VEGF and BDNF in both EA groups reached the highest on the 7th day(P<0.05 or P<0.01).Conclusion EA therapy can up-regulate the VEGF and BDNF expression levels and increase the rCBF in the cortex of rats with focal cerebral ischemia,which may be one mechanism of EA in the cerebral ischemia treatment.The therapeutic effect is accumulated with the effective time,and the concurrent treatment of the brain and heart group is superior to the traditional acupoint group in promoting angiogenesis.展开更多
