目的 探讨膜微阵列技术检测细菌的可行性。方法 在细菌的 1 6SrRNA基因内设计了通用引物和特异性探针 ,将探针点在膜上制备微阵列 ,再与地高辛标记的DNA杂交 ,比较了随机引物标记法和PCR掺入标记法的灵敏度 ,探讨了硝酸纤维素膜和尼... 目的 探讨膜微阵列技术检测细菌的可行性。方法 在细菌的 1 6SrRNA基因内设计了通用引物和特异性探针 ,将探针点在膜上制备微阵列 ,再与地高辛标记的DNA杂交 ,比较了随机引物标记法和PCR掺入标记法的灵敏度 ,探讨了硝酸纤维素膜和尼龙膜作基片的可行性 ,并比较了不同的固定条件和杂交条件下的杂交结果 ,以标准菌株DNA为样本 ,建立起微阵列技术。结果 随机引物标记法和PCR掺入标记法的灵敏度均为 0 .1pg ;尼龙膜较之硝酸纤维素膜可更好的结合寡核苷酸 ,经紫外线交联 3min ,与杂交液 3(5×SSC ,5g/LSDS ,1 %封闭剂 )在 5 5℃杂交结果最好。标准菌株除结核杆菌、变形杆菌外 ,均能与其对应的特异探针杂交。结论 利用膜微阵列技术检测细菌有快速、敏感、特异。 展开更多

OBJECTIVE To study the gene expression profiles of human endometrial cancers at various differentia0ted grade levels and to identify the genes related to differentiation of the endometrial cancers. METHODS cDNA microarray technology was used to analyze the differentially-expressed genes among different differentiated grades of 32 cases of endometrial cancer. Hierarchical cluster analysis (HCA) for the gene expression profiles of the cases was employed. RESULTS The tissue samples were grouped based on the various differentiated tumor grades with 33 differentiation-related genes identified out (P〈0.001). Based on the results from the HCA, the conformity rate was 91% among the 33 differentially-expressed genes, and the analysis of pathological classification.CONCLUSION Genes related to the differentiation of endometrial cancer can be identified by using gene chips to analyze the expression profiles of endometrial cancers at various differentiated grades; HCA of the gene expression profiles can be helpful for distinguishing high-risk endometrial cancers before surgery. 展开更多

Metastasis and micrometastasis in ultra-low rectal cancer

Objective:The aim of our study was to investigate the lymph node metastasis of mesorectum and ischiorectal foss in ultra-low rectal cancer and its influence on the surgical procedure selection.Methods:Large slices and tissue microarray technology were used to detect metastasis lymph nodes from 23 dissected specimens of ultra-low rectal cancer.Results:415 lymph nodes were detected in the 23 specimens.169 and 59 lymph nodes were metastasis and micrometastasis, respectively.Twelve specimens were diagnosed with lymph node metastasis, while other 4 specimens were with lymph node micro-metastasis.There were 29.0% (49/169) and 17.2% (29/169) metastatic lymph nodes located in the outer and anterior region of mesorectum.There were 2 cases with ischiorectal fossa lymph node metastasis and 1 case with micrometastasis.The 3 metastatic cases were only 13% (13/23) of all the 23 cases.Conclusion:There is regional lymph node metastasis existing in ultra-low rectal cancer.The metastatic incidences in distal mesorectum and ischiorectal fossa are relatively low.Considering Miles operation as the standard surgical procedure for ultra-low rectal cancer should be evaluated renewedly. 展开更多
